    Notes: An aminopeptidase was isolated from a soil fungus, which specifically cleaves the unnatural N-terminal methionine in recombinant human growth hormone. Reaction mixtures with different ratios of aminopeptidase to recombinant methionyl human growth hormone showed that the removal of N-terminal methionine was complete at 1:200 (w/w), and more than 90% complete at ratios up to 1:2000 (w/w) when incubated for 24 h at 37°C. The data indicate that the aminopeptidase we have purified can be used for the efficient conversion of unnatural recombinant proteins to their natural form.

    Notes: Summary UV-irradiation of the vegetal hemisphere of amphibian eggs leads to developmental abnormalities in neural morphogenesis. The possibility that the egg's transient sensitivity to irradiation could be due to pigmentation changes was examined in albino eggs. The tissue specificity of the effects of irradiation was analyzed by exchanging the ectoderm between irradiated and control embryos.

    Notes: Abstract In this paper, local and global error estimates for the element-free Galerkin (EFG) method are proposed. The essence of proposed error estimates is to use the difference between the values of the projected stress and these given directly by the EFG solution. The stress projection can be obtained simply by taking product of shape function based on a different domain of influence with the stresses at nodes. In this study, it was found that the effectivity index is optimized if the domain of influence in stress projection procedure is the smallest that retains regularity of the matrices in EFG. Numerical tests are shown for various 1D and 2D examples illustrating the good effectiveness of the proposed error estimator in the global energy norm and in the local error estimates.

    Notes: Abstract The development is described of a flexible manufacturing cell (FMC) programming and simulation system called FMCPS, which generates robot programs for a 6-DOF industrial robot. The development of FMCPS is based on an FMC that is being developed at UMIST. The cell consists of a Fanuc 600 ARCmate industrial robot, a Takisawa MAC V3 CNC machining centre and an MHP MT-50 CNC turning centre. The software has been developed on a SUN 4/110 workstation running under the UNIX operating system, using three-dimensional SUNCORE interactive graphics and SUNVIEW window systems. An event-oriented robot-programming method is used in the FMCPS. This method divides the robot tasks into different events. The user needs to define the points of the robot gripper route and the corresponding data for each event. The Cartesian coordinates of the points in the robot gripper paths are then transformed to the six robot joint angles by a variant inverse kinematics iteration method. These robot joint angles are used in the simulation model which generates robot programs suitable for the Fanuc robot. A three-dimensional, wire-frame-based graphical model is used for simulation. The user can observe the simultaneous animation of the robot movement, and display machine tool paths in separate graphics windows on the screen simultaneously. The various activities in the cell are simulated in pseudo-parallel mode by taking advantage of the multi-tasking and piping features of UNIX. At the end of a simulation exercise, the user is provided with the facility to obtain a performance report for the FMC which gives information on total operation time and on robot and machine utilisation, in addition to the robot program.

    Notes: Abstract A signal flow graph representation of the density matrix equations appropriate to a serial three-wave interaction is introduced, from which perturbation theory and AC Stark shift information may be extracted. The conditions for a large interaction cross section for the three-photon transition are deduced and applied to selected experimental situations with the main conclusion being that the effect may be present in a large number of CW and pulsed experiments.

    Notes: Abstract Fine powders of LiCoO2 were successfully synthesized through an emulsion drying method. The oxide powders were characterized by thermogravimetric-differential thermal analyses, X-ray diffractometry, scanning electron microscopy, elemental analysis and electrochemical method including charge–discharge cycling. By post-annealing the powders obtained by the emulsion drying method in the temperature range 600 ∼ 900 ∘C, we obtained LiCoO2 powder which has a layer structure (R3¯m) and consists of fine particles with submicrometre order in diameter. The charge–discharge characteristics as a cathode for lithium ion battery depended on the post-annealing temperature. LiCoO2 powder made by the emulsion drying method displays a good electrochemical performance, suggesting that this soft chemistry approach has the potential to synthesize a high quality electrode material.

    Notes: Abstract Effective transposon tagging with theAc/Ds system in heterologous plant species relies on the accomplishment of a potentially high transposon-induced mutation frequency. The primary parameters that determine the mutation frequency include the transposition frequency and the transposition distance. In addition, the development of a generally applicable transposon tagging strategy requires predictable transposition behaviour. We systematically analysedDs transposition frequencies andDs transposition distances in tobacco. An artificialDs element was engineered with reporter genes that allowed transposon excision and integration to be monitored visually. To analyse the variability ofDs transposition between different tobacco lines, eight single copy T-DNA transformants were selected. Fortrans-activation of theDs elements, differentAc lines were used carrying an unmodifiedAc + element, an immobilizedsAc element and a stableAc element under the control of a heterologous chalcone synthas (chsA) promoter. With allAc elements, eachDs line showed characteristic and heritable variegation patterns at the seedling level. SimilarDs line-specificity was observed for the frequency by whichDs transpositions were germinally transmitted, as well as for the distances of theDs transpositions. ThesAc element induced transposition ofDs late in plant development, resulting in low germinal transposition frequencies (0.37%) and high incidences of independent transposition (83%). The majority of theseDs elements (58%) transposed to genetically closed linked sites (≤10 cM).
