    Notes: Summary Influence of food on the serum concentration and kinetics ambenonium chloride (AMBC) has been examined in thirteen patients with myasthenia gravis (MG). Mean serum concentrations and Cmax during fasting were higher than those in the non-fasting state. The AUC (0–3 h) was also about four-times larger. The drug effects versus the serum concentration were observed to be anti-clockwise or clockwise. The effective range of the Cmax varied between patients. The unexpected increase in Cmax led to adverse muscarinic actions of AMBC, when the condition was changed from the non-fasting to the fasting state. It is recommended that the dose be changed during non-fasting treatment when adjusting the optimum regimen for patients myasthenia gravis. Patients must be advised to keep to the dosing and dietary schedule in order to avoid unexpected adverse actions to AMBC.

    Notes: Abstract In order minimize the possible contamination during storing and pre-treatment of such pure samples as ice and snow collected in Antarctica, trace elements in experimental tools such as bottles, beakers, tubings and filters were determined by neutron activation analysis. By using well certified tools, ice and snow samples from Antarctica and high mountains in China and in Japan were analyzed. Relative concentrations of volatile elements such as Zn, Cd, As, Sb or Ag to Al or Fe which are major components in the earth crust were found to be 10 to 1000 times higher than in the ordinary soil for the samples from Antarctica and Mt. Naimonanyi in China.

    Notes: Abstract Proteasomes function mainly in the ATP-dependent degradation of proteins that have been conjugated with ubiquitin. We isolated a cDNA clone for a rice protein that exhibited high homology to subunit C2 of proteasomes. Southern blot analysis revealed that the corresponding gene was present as a single copy in the rice genome. After fractionation of a crude extract from cultured cells, a 35-kDa protein that cross-reacted with antibodies against the C2 subunit was recovered in the peak fraction of both 20 S and 26 S complexes. The same antibodies cross-reacted with two proteins in seedlings, one of which was the same as that detected in cultured cells. The level of the protein was reduced in roots under conditions of high salinity. The 35-kDa protein was not detected in the nuclei of rice or carrot cells. However, during somatic embryogenesis of carrot cells, the C2 subunit was found in the nucleus at the globular stage, and it gradually disappeared in the period from the heart to the torpedo stage. Cells at the globular stage are proliferating rapidly, thus, it is possible that proteasomes are associated with the proliferation of plant cells.

    Notes: Abstract Tachyplesin I, a broad-spectrum antimicrobial peptide fromTachypleus tridentatus has a basic (+7), amphiphilic, and cyclic β-sheet structure. We reported (Matsuzaki K. et al. (1991) Biochim. Biophys. Acta 1070:259–264) that 1) the action mechanism of tachyplesin I may be the permeabilization of bacterial membranes, 2) the peptide specifically permeabilizes acidic phospholipid bilayers, and 3) its Trp2 residue is located in the hydrophobic region near the surface of the bilayers. In this paper, we found that tachyplesin I dose-dependently induces not only the permeabilization but also aggregation/fusion and micellization of the phosphatidylglycerol large unilamellar vesicles (100 nm in diameter) either in the gel (L-α-dipalmitoylphosphatidyl-DL-glycerol (DPPG)) or liquid-crystalline (egg yolk L-α-phosphatidyl-DL-glycerol (egg PG)) phase, as revealed by light scattering and electron micrograph techniques. The solid DPPG vesicles were more susceptible to the peptide. At peptide to lipid molar ratios (P/L) of 1/500 to 1/200, interpeptide interactions formed a pore through which calcein, a fluorescent dye, can leak out of the vesicles. The pore lifetime was longer in the DPPG vesicles. Further addition of the peptide caused aggregation and/or fusion of the vesicles. At a charge-neutralizingP/L ratio of 1/7, the enlarged vesicles disintegrated into small spherical particles (10–20 nm in diameter). The mechanism for these morphological changes will be discussed.

    Notes: Abstract In the present paper, the lower semicontinuity of certain classes of functionals is studied when the domain of integration, which defines the functionals, is not fixed. For this purpose, a certain class of domains introduced by Chenais is employed. For this class of domains, a basic lemma is proved that plays an essential role in the derivations of the lower-semicontinuity theorems. These theorems are applied to the study of the existence of the optimal domain in domain optimization problems; a boundary-value problem of Neumann type or Dirichlet type is the main constraint in these optimization problems.

    Notes: Abstract The second-order sensitivity analysis for a domain optimization problem is studied for a linear elastic structure. In the primary elastic structure considered, the surface traction, a part of the boundary conditions, depends not only on the position but also on the shape of the structure. The first variation and the second variation of the objective functional are calculated in terms of the solution, the first variation of the solution for the primal elastic system, and of the adjoint variable introduced. Moreover, the first-order and the second-order necessary optimality conditions are derived for the structure under a hydrostatic pressure. As an illustrative problem, a mean compliance design is treated.

    Notes: Abstract In this paper, domain optimization problems for both linear and nonlinear elastic structures are studied. The first variation and the second variation of the objective function are calculated in terms of the solution, of the first variation of the solution for the primal elastic system, and of the adjoint variables introduced. The adjoint variables obey a (fictitious) linear elastic system in contrast with the nonlinear adjoint systems introduced by Dems and Mróz, and by Dems and Haftka. From these results, the first-order and the second-order necessary conditions that an optimal domain should satisfy are immediately derived.
