    Notes: Abstract  Fusion of mononucleated myoblasts into parallel arrays of mutinucleated myotubes is an essential step in skeletal myogenesis. The formation of such a highly ordered structure requires myoblasts to come together, orient and align in the correct location prior to fusion. We report here that fetal and neonatal myoblasts can use topographical features as strong guidance cues in vitro. Myoblasts were cultured on multiple grooved substrata of varying dimensions, and the axial orientations of individual cells were recorded. Both fetal and neonatal myoblasts aligned parallel with the direction of deep grooves (2.3–6.0 µm), which is correlated well with the location of myoblasts in similar sized grooves during secondary myogenesis. Fetal myoblasts also responded to shallower grooves (0.04–0.14 µm) by aligning parallel or perpendicular to the direction of the grooves, indicating the ability of these cells to respond to fine elements normally encountered within the developing muscle architecture. In contrast, neonatal myoblasts failed to respond to shallow grooves, adding to the suggestion that fetal and neonatal myoblasts may represent separate populations of myoblasts. Overall, the results demonstrate that myoblasts respond to large and small features of the physical topography in vitro and indicate that structural elements in the microenvironment of the muscle may play a critical role in myoblast spatial organization during myogenesis.

    Notes: Abstract Multiphoton absorption (MPA) of the CO2 laser 9 μmR(30) line (pulse width 60 ns, FWHM) by NH3 and NH3/N2 mixtures was studied as a function of gas composition, pressure and laser fluence (Φ). MPA occurs for Φ smaller than those that have been used for optically pumped NH3/N2 lasers; consequently a simple two-level absorption model will not adequately describe laser action in these systems. The photon energy deposited in NH3/N2 mixtures can be calculated from the MPA cross-section and the fluence dependence of the illumination geometry. An examination of the efficiency of conversion of this absorbed energy to the reported radiant energy of optically pumped NH3/N2 lasers shows an optimum value which depends on pressure, and gas composition.

    Notes: Abstract Both the multiphoton absorption and the luminescence brightness of osmium tetroxide induced by absorption of temporally smooth CO2 10P(20) laser pulses of 10 and 40 ns FWHM have been measured for pressures in the range 6.7 to 200 Pa. A dependence of these phenomena on laser pulse length is observed. Comparison of the luminescence brightness to the average excitation shows that the distribution of the multiphoton excitation depends on the laser pulse intensity for the two pulse lengths studied.

    Notes: Summary The pharmacokinetics of a single-dose acetanilide (AA) test, and the urinary D-glucaric acid output (UDGAO) were studied in healthy drug free volunteer subjects before and at the end of a fourteen-day period of medication with 5 mg diphenylhydantoin sodium (DPH) per kg metabolically active mass, thrice daily. The steady state plasma concentration (SSPC) of DPH was also determined. The SSPC of DPH was found to vary from 3.4 to 19.6 µg per ml. The mean UDGAO values increased significantly during DPH medication from 9.5 µmol per g creatinine to 29.5 (p〈0.002). The half-life of plasma AA concentration decreased significantly during DPH medication from a mean of 4.2 h to a mean of 2.8 (p〈0.001). However, the plasma clearance of AA did not change significantly during DPH medication. The increase of UDGAO whilst on treatment with DPH correlated positively with the SSPC of DPH. The evidence from this study suggests that measurement of UDGAO, which is a measure of the activity of the glucuronic pathway does not necessarily indicate the state of induction of oxidizing enzymes in the hepatic endoplasmic reticulum.

    Notes: Summary Acetanilide and diphenylhydantoin have a similar first stage biotransformation in that both are oxidized in the para position of the benzene ring incorporated in each of the two molecules. The elimination of acetanilide from the plasma was studied in thirty healthy volunteer subjects following a single oral dose of 50 mg per kg metabolically active mass (MAM = weight to the power of 0.7). Plasma clearance values varied from 12.4 to 25.11 per hour. A dose of 5 mg diphenylhydantoin sodium (DPH) per kg MAM was then given thrice daily for 13 days to the same volunteers. The steady state plasma concentrations of DPH varied from 3.4 to 19.6 µg per ml. Statistically significant correlation was demonstrated between plasma acetanilide clearance and DPH clearance (r=+0.4984). This finding suggests either a common enzyme acceptor or a common rate-limiting step in the metabolism of the two drugs. It is possible that the pharmacokinetics of other widely used drugs known to be oxidized, especially phenylbutazone, may also be correlated with the kinetics of acetanilide and of DPH. If this were so, then certain individuals might be at a relatively high risk (due to drug accumulation) of developing adverse effects from drugs metabolized mainly by oxidation, and certain other individuals who metabolize these compounds at a fast rate are likely not to derive therapeutic benefit. A single dose study with simple measurement of acetanilide pharmacokinetics could be used to identify these groups.

    Notes: Summary We have studied the disposition of the centrally-acting analgesic meptazinol in a group of age-matched non-pregnant and pregnant (36–38 weeks gestation) women. Ten non-pregnant and nine multiparous pregnant volunteers each received a single i.v. dose of meptazinol hydrochloride (equivalent to 25 mg base). A further group of 9 non-pregnant (including four of the original participants) and 10 multiparous pregnant subjects were given repeated i.v. doses of meptazinol hydrochloride (each equivalent to 10 mg base) at 30-min intervals for 2.5 h. Meptazinol plasma concentrations were determined by HPLC using fluorescence detection and the pharmacokinetic variables investigated. After single dosing there were no statistical differences in half-life, clearance, or apparent volume of distribution between the two groups, suggesting that the disposition of meptazinol was not altered by pregnancy. This was confirmed in the repeated dose study, in which no significant differences occurred in either the plasma concentrations achieved or in areas under the curves between the non-pregnant and pregnant subjects. Furthermore, the steady-state concentrations were comparable with those predicted from the single dose results. This indicates that there should be no requirement for dosage alteration of meptazinol during pregnancy.

    Notes: Summary We have studied the disposition of perhexiline and its two major metabolites, M1 and M3, in healthy volunteers and in patients with biliary T-tube drains after cholecystectomy. In healthy volunteers the genetic control for impaired hepatic oxidation is identical for debrisoquine, sparteine, and perhexiline. Poor metabolizers demonstrate markedly reduced production and excretion of the major metabolite, M1. Their production of M3 is also reduced, but to a lesser degree than for M1, confirming substrate stereoselectivity by hepatic oxidases. Biphasic urinary elimination of M1 and M3 is seen in intact extensive oxidizers, whereas only the first phase is apparent in patients with biliary T-tube drainage. This suggests the possibility of enterohepatic recycling of these compounds, which may account for their prolonged elimination More than 90% of an ingested dose of perhexiline maleate remains unaccounted for at 24h after ingestion, even in extensive metabolizers. A careful, radiolabelled tissue-distribution study is warranted to elucidate the complicated metabolic fate of perhexiline.
