    Notes: Summary We have studied the genetic linkage of two markers, the apolipoprotein C1 (APOC1) gene and a cytochrome P450 (CYP2A) gene, in relation to the gene for myotonic dystrophy (DM). A peak lod score of 9.29 at 2 cM was observed for APOC1-DM, with a lod score of 8.55 at 4cM for CYP2A-DM. These two markers also show close linkage to each other (θ max = 0.05, Z max = 9.09). From examination of the genotypes of the recombinant individuals, CYP2A appears to map proximal to DM because in one recombinant individual CYP2A, APOC2 and CKMM had all recombined with DM. Evidence from another CYP2A-DM recombinant individual places CYP2A proximal to APOC2 and CKMM. Localisation of CYP2A on a panel of somatic cell hybrids also suggests that it is proximal to DM and APOC2/C1/E gene cluster.

    Notes: Summary The order of fourteen polymorphic markers localised to the long arm of human chromosome 19 has been established by multipoint mapping in a set of 40 CEPH (Centre d'Étude de Polymorphisme Humain, Paris) reference families. We report here the linkage relationship of the myotonic dystrophy (DM) locus to twelve of these markers as studied in 45 families with DM. The resulting genetic map is supported by the localisation of the DNA markers in a panel of somatic cell hybrids. Ten of the twelve markers have been shown to be proximal to the DM gene and two, PRKCG and D19S22, distal but at distances of approximately 25 cM and 15 cM, respectively. The closest proximal markers are APOC2 (apolipoprotein C-II) and CKM (creatine kinase, muscle) approximately 3 cM and 2 cM from the DM gene respectively, in the order APOC2-CKM-DM. The distance between APOC2, CKM and DM (of the order of 2 million base pairs) and their known orientation should permit directional chromosome walking and jumping. The data presented here should enable us to determine whether or not new markers are distal to APOC2/CKM and thus potentially flank the DM gene.

    Notes: Summary Two pyridine bases were isolated from the marine hoplonemertineAmphiporus angulatus (Fabricius) and identified by mass and PMR-spectroscopy as 2,3′-bipyridyl and 3,2′; 3′,2″; 4″,3′''-tetrapyridyl (Nemertelline). Nemertelline, the first tetrapyridyl natural product to be reported, shows a structural resemblance to the tobacco constituent nicotelline. The crustacean toxicity of 2,3′-bipyridyl is very similar to that of nicotine, but its mammalian toxicity is negligible.

    Notes: Abstract Repeated sampling of deep-sea bottom-living fishes was conducted at two stations in the Rockall Trough at depths of 2 200 and 2 900 m. The 2 200 m station (M) was sampled 16 times by an Agassiz trawl between April 1978 and April 1985 and yielded 473 fish belonging to 17 species. The 2 900 m permanent station (PS) yielded 781 fish belonging to 11 species from 9 Agassiz trawls, 2 small box otter trawls, 4 semi-balloon otter trawls and 7 epibenthic sledges between March 1975 and April 1985. Macrourid fishes were numerically dominant at both stations with Coryphaenoides guentheri and Coryphaenoides (Nematonurus) armatus being the most abundant species at M and PS, respectively. In terms of biomass the morid, Antimora rostrata, was dominant at M but C. (N.) armatus was dominant at PS. The only species that showed clear evidence of a seasonal reproductive cycle was C. guentheri. Many species showed no indication of reproducing in the Rockall Trough. The diets of almost all the species are described and considered in relation to the food resources exploited. Investigations on the pelagic and benthic invertebrate faunas at these stations should reveal the extent to which the bottom-living fishes exploit the available resources.

    Notes: Summary The apolipoprotein gene cluster on human chromosome 19 (APOC1, APOC2, APOE) has been localised by pulsed-field gel electrophoresis to within 200 kb of a chronic lymphocytic leukemia-associated translocation breakpoint. A restriction map covering 1300 kb around these loci has been constructed and contains two polymorphic MluI sites, which appear to show Mendelian inheritance. The orientation of the map on the chromosome has been established as 19cen CLL breakpoint-APOC2-19qter. Pedigree analysis using APOC2, a probe derived from the CLL breakpoint, and other localised markers on 19q suggests that the myotonic dystrophy locus is distal to APOC2 on 19q.

    Notes: Abstract. Yeast cells are capable of accumulation of various heavy metals, preferentially accumulating those of potential toxicity and also those of value. They retain their ability to accumulate heavy metals under a wide range of ambient conditions. In the present study it was shown that yeast cells in suspension accumulate heavy metal cations such as Cu2+, Co2+, and Cd2+. The level of copper accumulation was dependent on the ambient metal concentration and was markedly inhibited by extremes of ambient pH. Temperature (5–40°C) and the presence of the alkali metal sodium had much smaller effects on the level of copper accumulation. This suggests that in waste-waters of pH 5.0–9.0, yeast biomass could provide an effective bioaccumlator for removal and/or recovery of the metal. During bioaccumulation and subsequent processes it is necessary to retain the biomass. It was shown in the present study that this could be achieved by cell immobilization. Immobilization allowed for complete removal of Cu2+, Co2+, and Cd2+ from synthetic metal solutions. The immobilized material could be freed of metals by use of the chelating agent ethylenediamine tetraacetic acid (EDTA) and recycled for further bioaccumulation events with little loss of accumulation capacity.

    Notes: Abstract Propp and Wilson (Random Structures and Algorithms (1996) 9: 223–252, Journal of Algorithms (1998) 27: 170–217) described a protocol called coupling from the past (CFTP) for exact sampling from the steady-state distribution of a Markov chain Monte Carlo (MCMC) process. In it a past time is identified from which the paths of coupled Markov chains starting at every possible state would have coalesced into a single value by the present time; this value is then a sample from the steady-state distribution. Unfortunately, producing an exact sample typically requires a large computational effort. We consider the question of how to make efficient use of the sample values that are generated. In particular, we make use of regeneration events (cf. Mykland et al. Journal of the American Statistical Association (1995) 90: 233–241) to aid in the analysis of MCMC runs. In a regeneration event, the chain is in a fixed reference distribution– this allows the chain to be broken up into a series of tours which are independent, or nearly so (though they do not represent draws from the true stationary distribution). In this paper we consider using the CFTP and related algorithms to create tours. In some cases their elements are exactly in the stationary distribution; their length may be fixed or random. This allows us to combine the precision of exact sampling with the efficiency of using entire tours. Several algorithms and estimators are proposed and analysed.
