Notes: Summary Cells of the line Kc, derived fromDrosophila melanogaster embryos, extend long processes when exposed to ecdysteroid hormones. We have devised a quantitative assay for this morphological response, using the subline Kc-H. The assay was used to characterize the conditions required for the response. A halfmaximal response is elicited by approximately 10−8M 20-hydroxyecdysone; the response is saturated by 10−7M 20-hydroxyecdysone, which causes detectable elongation within a few hours, and a maximal response after 2–3 days. The response occurs substantially normally in the absence of serum, during growth in suspension, and in over-crowded cultures. It is not elicited by cyclic nucleotides, vertebrate growth factors, or a variety of other non-ecdysteroid reagents. Of 60 ecdysteroid compounds tested, only those which were active in other insect test systems elicited the response, and the concentrations required were approximately proportional to the concentrations active in other in vitro systems. We conclude that the response of Kc cells to 20-hydroxyecdysone retains basic features of the ecdysteroid response of intact tissues and therefore that Kc cells are a useful model system for studying ecdysteroid action.

Notes: Abstract Segregation of the complement component, C4, was analyzed in six families that each included an individual who inherited an HLA haplotype where a crossover event had occurred in the region between HLA-B and HLA-DR. Two cDNA clones corresponding to the C4 gene were utilized as probes in Southern blot analysis of DNA from members of each family. Restriction fragment length polymorphisms (RFLP) were observed and were assigned to haplotypes. In one family RFLP, hybridizing with the C4 probes, segregrated with HLA-B, and in four families RFLP segregated with HLA-DR; one family was not informative in this respect. These analyses have made it possible to localize the genes for C4 between HLA-B and HLA-DR by molecular genotyping and to characterize three different genomic configurations of C4 genes by limited restriction mapping.

Notes: Summary DiGeorge syndrome (DGS) is a human developmental defect of the structures derived from the third and fourth pharyngeal pouches. It apparently arises due to deletion of 22q11. We describe a strategy for the isolation of DNA probes for this region. A deleted chromosome 22, which includes 22q11, was flow-sorted from a lymphoblastoid cell line of a patient with cat eye syndrome and used as the source of DNA. A DNA library was constructed from this chromosome by cloning into the EcoR1 site of the vector Lambda gt10. Inserts were amplified by PCR and mapped using a somatic cell hybrid panel of this region. Out of 32 probes, 14 were mapped to 22q11. These probes were further sublocalised within the region by dosage analysis of DGS patients, and by the use of two new hybrid cell lines which we have produced from DGS patients. One of these lines (7939B662) contains the altered human chromosome segregated from its normal homologue. This chromosome 22 contains an interstitial deletion in 22q11, and will be useful for localising further probes to the DGS region.

Notes: Summary In this paper the uniqueness results found in simultaneous Chebychev approximation are extended to simultaneousL 1 approximation. In particular a sufficient condition to guarantee uniqueness of a best approximate to aL 1 compact set is given.

Notes: Summary Sulfo-N-succinimidyl derivatives of the long-chain fatty acids, oleic and myristic, were synthesized and covalently reacted with isolated rat adipocytes. The plasma membrane proteins labeled by these compounds and the effect of labeling on the transport of long-chain fatty acids were investigated. Sulfo-N-succinimidyl oleate (SSO) and myristate (SSM) inhibited the transport of fatty acids (by about 70%). Inhibition of fatty acid transport was not a result of alterations in cell integrity, as intracellular water volume was not changed. It did not reflect effects on fatty acid metabolism, since it was observed under conditions where greater than 90% of the fatty acid taken up was recovered in the free form. The inhibitory effect was specific to the fatty acid transport system, as the transport of glucose and the permeation of retinoic acid, a substance with structural similarities to long-chain fatty acids, were unaffected. Sulfosuccinimidyl oleate reacted exclusively with a plasma membrane protein with an apparent size of 85 kDa while sulfosuccinimidyl myristate also labeled a 75-kDa while sulfosuccinimidyl myristate also labeled a 75-kDa protein. These proteins were among the ones labeled by diisothiocyanodisulfonic acid (DIDS) which also inhibits fatty acid transport irreversibly. The data suggest that the 85-kDa protein, which is the only one labeled by all three inhibitors is involved in facilitating membrane permeation of long-chain fatty acids.

Notes: Abstract We study ππ correlations in the exclusive reaction pp → 2π+2π− at rest measured by the CPLEAR experiment. Avoiding the introduction of an arbitrary reference sample, we analyse differential distributions for equal charge pion pairs removing the phase-space factor event by event. A peak at small relative momenta is most pronounced for large total momentum of the pair. The physical implications of bosonic symmetrization for the properties of the pion source, in particular its radius, are briefly discussed. The two extremes considered are the chaotic Hanbury-Brown-Twiss mechanism and the coherent Skyrmion model.

Notes: Abstract A time-sinusoidal circularly polarized plane standing wave solution is obtained for isotropic lossless dielectric media with arbitrary nonlinearity. The spatial variation of the standing wave depends on the nonlinearity and is found by solving a problem of centralforce motion in which the electric complementary energy density furnishes the potential and the spatial coordinate has the role of time. Two special nonlinear dielectric response laws-cubic and quintic-are treated, and explicit solutions for the spatial variation of the wave amplitude and phase are obtained in terms of elliptic functions and elliptic integrals, respectively. The standing-wave solution is applied to give steady state solutions of two reflection/transmission problems (a) reflection from, or resonance modes between, ideally conducting planes and (b) reflection and transmission at a plane interface between two nonlinear dielectric media.

Notes: Abstract A finite oscillatory shearing motion is shown to be possible, in the absence of body force, in every homogeneous isotropic compressible or incompressible elastic solid. The spatial geometry is the same for all materials and the nature of the time-dependence, for a particular material, is determined by the generalized shear modulus. A motion of this type and a spatially uniform, time-dependent temperature can be supported in thermoelastic solids, without application of body force or volume supply of heat.

Notes: Abstract Motions of a certain type are shown to be possible in every compressible or incompressible, homogeneous, isotropic simple material, in the absence of body force. These motions have a specific spatial dependence and the nature of the time dependence, for a particular material, is determined by the material response function or functional for the shear stress in simple shear. It is shown further that motions of this type, and spatially homogeneous, time dependent temperatures, can be supported without application of body force or radiative supply of heat.

Notes: Abstract Within the scope of the nonlinear theory of an elastic Cosserat surface, this paper is mainly concerned with the influence of the reference geometry and the related aspects of material symmetry restriction on the response of thermoelastic shells. The significance of the effect of the reference geometry is discussed in the case of isotropic shells, which may be of variable thickness in a reference state.
