    Description / Table of Contents: Résumé L'étude de différents allotypes du système Gm a été pratiquée sur les sérums provenant de onze familles dont au moins un des membres présentait un déficit mineur ou grave portant sur une ou plusieurs classes d'immunoglobulines. Dans six de ces familles, deux ordres de faits sont observés, simultanément ou non, chez des sujets ne présentant, par les méthodes classiques, aucun désordre immunologique: une diminution de l'expression phénotypique d'un ou plusieurs facteurs qui permet de conclure à un déficit sélectif de certaines sous-classes de γG; ces déficits sont, parfois, transmis directement à la génération suivante; des anomalies d'ordre qualitatif se caractérisant par la présence de phénotypes, en règle générale, rarement rencontrés parmi des Caucasiens. L'hypothèse faisant appel à l'existence de gènes de structure rares ou anormaux pour expliquer ces faits ne peut être retenue car elle aboutit à la constatation de trop nombreuses exclusions de paternité ou de maternité. Dans une seconde hypothèse, nous supposons qu'existent des gènes de régulation responsables de la synthèse des chaines γ1, γ2 et γ3 et que les anomalies phénotypiques observées sont l'expression de la modification de ces gènes. La transmission de ces gènes de régulation se ferait indépendamment de celle des gènes de structure.
    Notes: Summary Different allotypes of the Gm system were studied on sera from eleven families where at least one of the members had a minor or important deficiency in one or several classes of immunoglobulins. In six of these families two facts were observed, simultaneously or not, in subjects showing no immunologic disorder, as revealed by conventional methods: a reduction in the phenotypic expression of one or several factors which allowed to conclude that there is a selective deficiency in certain γG subclasses; these deficiencies are sometimes directly transmitted to the following generation; qualitative anomalies characterized by the presence of phenotypes, as a general rule, rarely encountered among Caucasians. The hypothesis based on the existence of rare or abnormal structural genes could not be applied to explain these facts because it would have led to the statement that there are too many exclusions of paternity or maternity. In a second hypothesis, we supposed that there exist regulator genes, responsible for the synthesis of the γ1, γ2, and γ3 chains, and that the observed phenotypic anomalies are an expression of the mutation of these genes. The transmission of these regulator genes could be independent of that of the structural genes.

    Notes: Summary Midazolam is a water soluble benzodiazepine, with a short elimination half-life in adults and children. An IV bolus (0.2 mg·kg−1) immediately followed by continuous infusion of 0.06 mg·kg−1·h−1 was administered to 15 critically ill neonates at a gestational age of 32.8 weeks, who required sedation for mechanical ventilation. Heart rate and blood pressure were closely monitored. Hypotension occurred in 4 patients after the bolus dose or during the continuous infusion. Three of them had also been given fentanyl. Individual pharmacokinetic parameters were calculated: plasma clearance was 3.9 ml·min−1, elimination half-life was 12.0 h. Because of its short half-life compared to diazepam, midazolam may be used during the neonatal period to achieve rapid, brief sedation. However, it should be administered cautiously to neonates, particularly in premature infants, or if fentanyl is also given.

    Notes: Abstract  Radial distribution of soluble phenolics was investigated at different heights in stems of Juglans nigra, J. regia and hybrids J. nigra 23 × J. regia. Four major phenolic compounds were studied: hydrojuglone glucoside (HJG), quercitrin (QUER) and two unknown compounds characterized as two ellagic acid derivatives E1 and E2. HJG and E1 content increased gradually in the sapwood, peaked in the sapwood-heartwood transition zone, and decreased drastically in the heartwood. QUER was accumulated preferentially around the transition zone, and its content was relatively low in the outer part of the sapwood and in the inner part of the heartwood. E2 content was low in the sapwood and increased in the heartwood. The heartwood formation was marked by the accumulation of new soluble compounds. The relationship between wood extractives and wood colour were evaluated and discussed. HJG was considered to be a major precursor of heartwood colour providing chromophores through hydrolysis (deglucosylation), oxidation and polymerization processes.

    Notes: Abstract  The influence of texture and local climate, especially precipitation, on C decomposition was evaluated in topsoils under forest and pastures at different sites located in the western, central, and eastern Brazilian Amazon Basin. Independent of the geographical location and vegetation cover, C decomposition was lower in topsoils of fine texture with a sand/clay ratio of less than 1, compared to topsoils of coarse a texture with a sand/clay ratio ranging from 2 to 8. In the former, the low sand/clay ratio appears to reduce C decomposition. In the latter, besides the effect of the high sand/clay ratio, the local climate, especially the annual precipitation, seems to play an important role in controlling C decomposition.

    Notes: Abstract  Transgenic rice (Oryza sativa L.) cell lines and plants expressing an oat arginine decarboxylase (Adc) cDNA under the control of the CaMV 35 S promoter were recovered using particle bombardment. Molecular analyses confirmed stable integration of the transgene and active transcription (mRNA). A four- to sevenfold increase in arginine decarboxylase (ADC) activity was observed in transformed plants compared to wild-type controls. Biochemical analysis of cellular polyamines (PAs) indicated up to fourfold increase in putrescine (Put) levels in transgenic callus and regenerated plants. This is the first report which demonstrates an increase in PA levels in plants engineered with the Adc gene. Implications of this increase are discussed in terms of development, physiology and nutrition. We observed a correlation between high levels of Adc gene expression and inability of callus tissue to develop normally into differentiated plants. This correlates well with reports in other species, in which perturbation of the PA pathway using genes involved in PA biosynthesis results in aberrant phenotypes. We have shown for the first time that PA biosynthesis can be manipulated in cereal species using genetic engineering.

    Notes: Abstract An in vitro human reconstructed epidermis model (SkinEthic) used for screening acute and chronic skin irritation potential was validated against in vivo data from skin tolerability studies. The irritation potential of sodium lauryl sulfate (SLS), calcipotriol and trans-retinoic acid was investigated. The in vitro epidermis-like model consists of cultures of keratinocytes from human foreskin on a polycarbonate filter. The modulation of cell viability, the release and gene expression of proinflammatory cytokines, interleukins 1α and 8, and morphological changes were evaluated during 3 days as endpoints representative for an inflammatory reaction. The cumulative irritation potential of the topical products was evaluated in a human clinical study by visual scoring and biophysical measurement of inflammatory skin reaction after repeated 24 h applications over 3 weeks under Finn chamber patches. All topical products that were nonirritating in the human study were noncytotoxic and did not induce cytokine expression in the in vitro acute model (day 1 exposure). All irritating controls exhibited specific cell viability and cytokine patterns, which were predictive of the in vivo human data. The ranking of mild to moderate skin irritation potential was based on the lack of cytotoxicity and the presence of cytokine patterns including gene expression specific for each irritant, using the chronic in vitro model (up to 3 days exposure). The human reconstructed epidermis model SkinEthic was shown to be a reliable preclinical tool predicting the irritation potential of topical products. Moreover, it is a useful model in a two-step tiered strategy for screening acute and chronic irritation potential for the selection of vehicles for new topical drugs.

Oxygen consumption $$\dot M_{O_2 } $$ , ventilation, $$\dot Vw$$ , and circulatory flow-rate, $$\dot Vb$$ , were studied in settled crayfishAstacus leptodactylus at different O2 partial pressures in the inspired water, $$PI_{O_2 } $$ , ranging from 250 to 8 Torr. From $$PI_{O_2 } $$ =250 to 25 Torr, $$\dot M_{O_2 } $$ was maintained constant while $$\dot Vw$$ increased 16 times the hyperoxic value. Since $$P_{O_2 } $$ of both arterialized hemolymph and expired water remained in a narrow range independent of $$PI_{O_2 } $$ , we propose that $$\dot Vw$$ is adjusted in order to maintain the level of oxygenation at the gas exchanger. Down to $$PI_{O_2 } $$ =40 Torr, the observed arterio-venous concentration difference and $$\dot Vb$$ both remained constant. At $$PI_{O_2 } $$ =25 Torr, however, a further increase in $$\dot Vw$$ could not maintain the (a-v)O 2 concentration difference. As $$\dot M_{O_2 } $$ did not fall, it is concluded that $$\dot Vb$$ increased. 2. The role of the peripheral O2 chemoreceptors in these ventilatory and circulatory adjustments was analysed by a study of responses to short periods of exposure to $$PI_{O_2 } $$ ≃600 Torr (one-minute ‘O2 test’). Giving O2 tests to animals at $$PI_{O_2 } $$ plateaux situated between 150 and 15 Torr induced a rapid change in the respiratory frequency which started before the hyperoxic bolus could have reached the central nervous system (CNS). Response latencies were shorter the higher the reference $$\dot Vw$$ ; they were independent of the time required for the hemolymph to reach the heart and hence the CNS. It is concluded that the responses were initiated by O2 chemoreceptors located in the branchial cavities. The branchial location of O2 reception is consistent with the above proposition of a $$P_{O_2 } $$ regulation at the gas exchanger. The importance of a central O2 chemosensitivity is questioned.

Chronically hypoxic crayfishAstacus leptodactylus at 20°C were transiently exposed (1 or 2 minute tests) to either hyperoxic, hypercapnic, normoxic, or normoxic and hypercapnic waters. Using O2 probes to monitor changes in O2 levels of the inspired water and of the hemolymph in the pericardial sinus following brief O2 exposures, the ventilatory response delay and circulation time required for hemolymph to reach the heart and hence the central nervous system (CNS) were followed. In some CO2 tests, fast pH probes recorded changes of hemolymph pH in the pericardial cavity. 2. All crayfish exhibited a CO2 induced hyperventilation. At $$PI_{O_2 } $$ = 40–50 Torr, the ventilatory response to CO2 tests started before the O2 labelled hemolymph could have reached the CNS. The CO2 response delay was shortened when the rate of ventilation was increased by lowering $$PI_{O_2 } $$ from 70 to 40 Torr. As circulation time is not shortened in these conditions (Massabuau and Burtin 1984), it is concluded that CO2 chemoreceptors are present in the branchial cavities. The importance of a central CO2 chemosensitivity is discussed.
