    Description / Table of Contents: Resumen A partir de una cepa de Conidiobolus coronatus aislada de Aenolamia postica, se inocularon 152 ratones, 42 cobayos y 47 hamsters por vías intraperitoneal (IP), subcutánea (SC), intratraqueal y por escarificación en la mucosa oral; cada animal se inoculó 3 veces a intervalos de una semana. Los animales se sacrificaron a los 1, 8, 15, 45 y 225 días de la primera inoculación, haciéndose estudios de alteraciones macroscópicas (nódulos, abscesos, adherencias), exámenes directos, cultivos e histopatología. Las alteraciones macroscópicas de los tejidos fueron las más frecuentes y todas evolucionaron a la curación espontánea. Los animales presentaron un mayor múmero de estudios positivos cuando se inocularon por vía IP y SC. Los estudios histopatológicos hechos al 1° y 8° día, demostraron un infiltrado inflamatorio agudo y filamentos fúngicos bien conservados; a los 15 días se observaron granulomas con células gigantes y filamentos fragmentados en desintegración; a los 45 y 225 días se observó una reacción de fibrosis sin filamentos fúngicos. Se consideró que en ningún caso se logró reproducir la rinoentomoftoromicosis y de acuerdo a éstos resultados se estima que esta cepa no tiene patogenicidad o bien ésta es muy baja.
    Notes: Abstract The pathogenicity of a strain of Conidiobolus coronatus isolated from Aenolamia postica was investigated in 152 mice, 42 guinea pigs and 47 hamsters. Four routes of inoculation were used: intraperitoneal, subcutaneous, intratracheal and scarification in the oral mucosa. Each animal was inoculated 3 times at 3 week intervals. Animals were sacrificed at 1, 8, 15, 45 of 225 days after the first inoculation. Observations of macroscopic lesions (nodules, abscesses, adhesions, etc.) direct examinations, cultures and histopathological studies of different tissues were performed. The aforementioned lesions of tissue were the most frequently found and all of them were resolved by spontaneous cure. The animals inoculated intraperitoneally and subcutaneouslly presented the highest number of positive results. The histopathological features at the 1st and 8th days after inoculation showed an acute inflamatory infiltrate with well preserved fungus filaments; at the 15th day granulomas with giant cells were observed containing fragmented and disintegrated fungal hyphae; at the 45th and 225th days a fibrosis reaction was observed and no fungal filaments, could be found. In no animal was rhinoentomophthoromycosis never reproduced, and according to these results it is considered that either this strain is not a pathogen or its pathogenicity is very low.

    Notes: Summary MDCK cells form monolayers which have many of the properties usually found in transporting epithelia. The present article is devoted to the study of the structural and functional polarization of MDCK cells, which is one of the central features of transporting epithelia. The results show: (i) that MDCK monolayers transport 2.6 μmol hr−1 cm−2 of sodium in the apical to basolateral direction; (ii) the passive flux of this ion is relatively large (20.3 mole hr−1 cm−2), which is a characteristic of leaky epithelia; (iii) a large fraction of the penetration of sodium into the cells proceeds through an amiloride-sensitive channel, and the exit is operated mainly by a ouabain-sensitive pump; (iv) the net transport of sodium from the apical to the basolateral side agrees with the asymmetric labeling of the pumps with3H-ouabain; (v) this asymmetric labeling agrees, in turn, with a higher concentration of intramembrane particles (IMPs) in freeze-fracture replicas of the basolateral side of the plasma membrane; (vi) the structural polarization of confluent MDCK cells is also revealed by the location of microvilli, occluding junctions, and pinocytotic vesicles; and (vii) the presence of a continuous ring formed by actin microfilaments visualized by immunofluorescence under the lateral aspect of the plasma membrane that may be related to the distribution of the occluding junctions, which act as barriers separating apical from basolateral membrane components.

    Notes: Summary MDCK cells (epithelioid of renal origin) form monolayers which are structurally and functionally similar to transporting epithelia. One of these similarities is the ability to form occluding junctions and act as permeability barriers. This article studies the junctions of MDCK monolayers formed on a permeable and transparent support (a disk of nylon cloth coated with collagen) by combining two different approaches: (i)Scanning of the electric field: the disk is mounted as a flat sheet between two Lucite chambers and pulses of 20–50 μA cm−2 are passed across. The apical surface of the monolayer is then scanned with a microelectrode to detect those points where the current is flowing. This shows that the occluding junctions of this preparation are not homogeneous, but contain long segments of high resistance, intercalated with sites of high conductance. (ii)Freeze fracture electron microscopy: the junctions are composed of regions of eight to ten strands intercalated with others where the strands are reduced to one or two ridges. The sites of high conductance may correspond to those segments where the number of junctional strands is reduced to 1 or 2. It is concluded that the occluding junctions of MDCK monolayers are functionally and morphologically heterogeneous, with “tight” regions intermixed with “leaky” ones.

    Notes: Abstract The specific growth rate (μ) of a respiration-deficient mutant of Saccharomyces cerevisiae growing under defined experimental conditions in batch culture (mineral medium plus glucose and vitamins at 25°C) varied from experiment to experiment over a wide range (0.10–0.24 h-1) and showed a normal distribution. Neither the age of the culture, the history of the inoculum, nor experimental error accounted wholy for the variability of μ. The variation was positively correlated with the specific rate of glucose transfer and negatively with the specific rate of production of non-fermentative CO2. The yield decreased with μ implying higher maintenance requirements in batch culture (4.7 mmoles g-1 h-1) than in continuous culture (0.8 mmoles g-1 h-1). It was concluded that the strain is capable of establishing any one of several steady states of growth under the same experimental conditions, each steady state displaying some buildin inertia with respect to change. The variations of the specific rates of glucose transfer and non-fermentative CO2 production, and of the yield appeared to be consequences rather than causes of the variation of μ. The ultimate causes of the variation of μ remained unidentified.

    Notes: Summary Clostridium thermocellum ATCC 27405 (and its improved cellulase-producing mutant, AS-39) is an anaerobic thermophile that produces endo-β-glucanase and exo-β-glucanase when grown on cellobiose or cellulose as major carbon source (Shinmyo et al. 1979). The site of cellulase accumulation was at least 95% extracellular. Optimum conditions for endo-β-glucanase production in flasks included 1% (w/v) cellobiose, 0.2% (w/v) urea as a nitrogen source, 0.1 M morpholinopropane-sulfonic acid buffer, an initial pH of 7.4, and a yeast extract concentration of 0.6% (w/v). An improved medium (GS medium) was devised for future studies. Xylan was degraded by an extracellular enzyme (s) produced during cultivation on cellobiose, although C. thermocellum does not grow on xylan.
