    Notes: Abstract. We previously cloned a MaxiK channel α-subunit isoform, rbslo1, from rabbit kidney with an amino acid sequence highly homologous to mslo but with a 59 amino acid insertion between S8 and S9 (Morita et al., 1997. Am. J. Physiol. 273:F615–F624). rbslo1 activation properties differed substantially from mslo with much greater Ca2+ sensitivity, half-activation potential of −49 mV in 1 μm Ca2+. We now report single-channel analysis of rbslo1 and delA, a construct produced by removal of the 59 amino acid insertion at site A. delA is identical to mslo from upstream of S1 to downstream of S10 with the exception of 8 amino acids. Slope of the steady-state Boltzmann voltage activation curve was 8.1 mV per e-fold change in probability of opening for both rbslo1 and delA. The apparent [Ca2+] i properties in delA were more like mslo but the voltage-activation properties remained distinctly rbslo1. Ca2+ affinity decreased and transmembrane voltage effects on apparent Ca2+ affinity increased in delA. The differences between rbslo1 and other cloned channels appear to be localized at insertion site A with both the insertion sequence and amino acid substitutions near site A being important. The steeper activation slope makes the channel more responsive to small changes in transmembrane voltage while the insertion sequence makes the channel functional at physiological low levels of [Ca2+] i .

    Notes: Abstract The inheritance of 15 enzymes, comprising at least 22 genetic loci, was investigated in crosses between sexual diploid individuals of Taraxacum sections Vulgaria and Mongolica. Patterns were consistent with simple Mendelian segregation. From the inheritance information isozyme phenotypes in agamospermous plants from natural populations were inferred. In some crosses part or all of the progeny originated from self-fertilization, sofar a very rare phenomenon in the sections Vulgaria and Mongolica. It is possible that the probability of self-fertilization increases after pollination by triploid pollen, affecting the cohabitation dynamics of the various ploidy components in mixed natural stands.

    Notes: Abstract Polymerization reactions of colloidal silica spheres via the hydrolysis and dehydration processes of tetraethyl orthosilicate with ammonia and a tiny amount of water in ethyl alcohol have been studied in microgravity by the parabolic flights of a MU-300 rear-jet aircraft. Induction periods and polymerization rates are determined by fast-scanning transmitted-light-intensity measurements and the fast-scanning dynamic light-scattering method. Direct observation of the reaction mixtures is also made with a charge-coupled device video camera. Reproducible and reliable data are obtained in microgravity compared with those in gravity. Increases in the induction times and decreases in the polymerization rates are observed in microgravity compared with those in gravity. One of the main reasons for these observations is the fact that the translational Brownian movement of the reactants and/or product spheres is free from downward translational movement in microgravity. Very weak convection of the reaction suspensions in microgravity is another important factor.

The expression of mouseRad51 mRNA was examined in synchronized mouse m5S cells. TheRad51 transcript was observed from late G1 phase through to M phase. During the period of late G1-S-G2, the RAD51 proteins were observed exclusively in nuclei. Activation by mitogens of T cell and B cell proliferation in spleen induced the expression ofRad51 mRNA. By immunohistochemical analyses, the mouse RAD51 protein was detected in proliferating cells: spermatogonia in testis, immature T cells in thymus, germinal center cells of the secondary lymphatic nodules of spleen and intestine, follicle cells in ovary and epithelial cells in uterus and intestine. It was also expressed in spermatocytes during early and mid-prophase of meiosis and in resting oocytes before maturation. Thus, mouseRad51 expression is closely related to the state of cell proliferation and is presumably involved in DNA repair coupled with DNA replication, as well as in meiotic DNA recombination in spermatocytes.

    Notes: Abstract We report a change in the proliferative activity of mouse colonic epithelium due to development and aging. In order to measure the proliferative activity, colonic epithelium was immunostained for cyclin proliferating cell nuclear antigen (PCNA/cyclin), which appears from the Gl to the S phase of the cell cycle, and compared with labeling obtained by [3H]-thymidine radioautography. Litter mice of six age groups from the fetal period (embryonic day 19), newborn period (postnatal day 1), suckling period (postnatal day 5), weaning period (postnatal dy 21), adult period (2 month old) to the senescent period (11 month old) were examined by immunohistochemistry. The descending colons were fixed in methacarn (method-Carnoy) and embedded in paraffin. Sections were stained for PCNA/cyclin activity using 19A2 monoclonal antibody and the avidin-biotin peroxidase complex (ABC) technique. For radioautography, litter mice of nine age groups using in vivo intraperitoneal administration of [3H]-thymidine. The labeling indices of colonic epithelial cells in the proliferative zone were then analyzed and compared between the two investigative methods. Our results show that the prliferative activity of mice colon was high in the fetal and newborn periods and almost constant from the suckling period to senescence, as demonstrated by both PCNA/cyclin immunohistochemistry and [3H]-thymidine radioautography. The labeling index seen by PCNA/cyclin immunohistochemistry was, however, higher than that seen by [3H]-thymidine radioautography.

    Notes: Abstract Diurnal variation of hue discriminatory capability under artificial constant illumination of 4000 lux was studied with 10 young female adults using the 100-hue test. There were conspicuous diurnal variations in the yellow-blue and red-green systems, with marked reductions of error score in the evening. However, observations of the blue, yellow, green and red systems separately disclosed that there existed clear diurnal rhythms in the blue and green systems, but not in the yellow and red systems. This suggests the existence of diurnal variation in function of the S and M cones responsible for the blue and green hue.

    Notes: Abstract The dependence of the dissolution temperature (T sol) of isotactic polyacrylonitrile (PAN) on tacticity was studied for three dinitrile solvents. A linear relationship was obtained in the inverse plots of the tacticity dependence of the T sol of PAN. A phenomenological analogy between the tacticity dependence of the T sol of isotactic PAN and the molecular-weight dependence of the glass-transition temperature of amorphous polystyrene is discussed from a thermodynamic point of view. The linear relationships in both phenomena are explained in terms of a common mechanism: a breakdown of thermodynamic competition, enthalpy, and entropy through the segment mobility. The significance of segment concept and molecular mobility at elevated temperatures are discussed.

    Notes: Abstract A redundancy is noted in the set of Lagrange multipliers—effective fields and effective interactions—used in the expressions of the reduced density matrices given by Morita(4) in some problems with less symmetry(6). The expressions of the reduced density matrices without this redundancy are given. An argument showing that a certain expression of the free energy in terms of the Lagrange multipliers is stationary with respect to the variations of these is presented.
