    Notes: Abstract Glucose-limited continuous cultures were used to analyze σB activity at decreasing growth rates. Expression of the σB-dependent genes gsiB and ctc started to increase at a growth rate of 0.2 h–1, and both genes were induced approximately fivefold at a growth rate of 0.1 h–1 as compared to expression at the maximal growth rate. However, maximal σB activity was only reached when the growth stopped as a result of the exhaustion of the carbon and energy source glucose. During glucose-limited growth, increased expression of the general stress regulon at growth rates below 0.2 h–1 did not provide wild-type cells with a growth advantage over sigB mutants. Instead, expression of the stress regulon seems to constitute a significant burden during glucose-limited growth, resulting in a selective growth advantage of the sigB mutant as compared to the wild-type at a growth rate of 0.08 h–1.

    Notes: Abstract In Bacillus subtilis, heat shock proteins can be classified into two main groups: specific heat shock proteins (about 5) and general stress proteins (at least 14). Salt stress was very effective in the induction of general stress proteins (5 to 50-fold stimulation), but the synthesis of heat-specific stress proteins was not stimulated. Furthermore there were some proteins whose synthesis was accelerated only by salt stress.

    Notes: Summary To evaluate phenprocoumon elimination its possible biliary excretion was evaluated in addition to the known pathway of renal elimination. Bile samples were obtained during diagnostic endoscopy in patients receiving chronic phenprocoumon therapy and were analyzed for phenprocoumon and its metabolites by HPLC and GC-MS. The following substances were detected, mainly in conjugated form: unchanged phenprocoumon and the metabolites 7-hydroxy-, 4'-hydroxy-, and 6-hydroxy-phenprocoumon. The data provide direct evidence of the biliary elimination of unchanged phenprocoumon and its metabolites in humans.

    Notes: Summary The haemodynamic effects of a combined intravenous treatment of nifedipine and propranolol in ten patients with coronary artery disease compared to a single treatment with nifedipine or placebo were investigated. Nifedipine infusion resulted in a reduction of left ventricular (LV) afterload and LV volumes with an increase in heart rate and EF and no change of the double product, coronary sinus flow, LV diastolic parameters and dp/dtmax. Addition of propranolol lowers myocardial oxygen demand by reducing heart rate and dp/dtmax together with a sustained afterload reduction with no change in LV volumes and EF. The vasodilatatory action of nifedipine pretreatment balanced the negative effects of acute beta-receptor blockade on LV function and allows the reduction of myocardial oxygen demand without a deterioration of LV function.

    Notes: Abstract In end-stage heart failure due to idiopathic dilated cardiomyopathy β1-adrenoceptors are downregulated and G1α-proteins are upregulated. The aim of the present study was to investigate the influence of the angiotensin-converting enzyme inhibitor captopril on β-adrenoceptor density and Giα-proteins in sequential endomyocardial biopsies. Nineteen patients with mild to moderate congestive heart failure due to idiopathic dilated cardiomyopathy (NYHA Class II–III) were studied before and after 8–11 weeks of therapy. Patients were randomised into a captopril and a control group; 9 patients received captopril 12.5–50 mg per day, (divided in 2–3 doses) p.o. in addition to “conventional” therapy with digoxin and diuretics, and 10 controls received “conventional” therapy only. Echocardiography, spiroergometry, right heart catheterisation and endomyocardial biopsies were performed before (baseline) and after treatment. Compared to baseline, captopril increased total β-adrenoceptor density by selectively increasing β1-adrenoceptors (31.6 vs 41.2 fmol·mg−1; p〈0.05) but had no significant effect on Giα-proteins. The results indicate that treatment with angiotensin-converting enzyme inhibitors partly restores myocardial β1-adrenoceptor density, and this action effect may contribute to the clinical improvement of patients with idiopathic dilated cardiomyopathy treated in this way.

    Notes: Abstract  The De Gerlache Seamounts actually consist of two medium-sized guyots, with summits at depths of 350–600 m. Acoustic profiler data show no significant sediment on these guyots. Alkaline basalts dredged from the summit of the eastern guyot yield K/Ar ages between 20.1±1.0 and 23.2±1.2 Ma. Basement ridges and sediment-filled troughs between the guyots are associated with the prominent gravity anomaly extending north from the Antarctic margin. This structure possibly played a role in the guyot formation, however, the question of how the De Gerlache Seamounts are related to this gravity anomaly remains uncertain.

    Notes: Summary In electron micrographs it could be shown that hazelnut (Corylus avellana) pollen grains are covered on their surface by a diffusible 10 nm thick lamellar layer. On pollen surface as well as in pollen extract this layer could be precipitated and stained by the polycationic dye Cuprolinic blue. By subsequent application of both immunogold labeling with serum from a hay-fever patient allergic to tree pollen grains and histochemical detection with Cuprolinic blue this pollen surface layer proved to be an effective antigen.

    Description / Table of Contents: Zusammenfassung Die Cytokinese in jungen Brutkörpern vonMarchantia polymorpha wurde licht- und elektronenmikroskopisch untersucht. Die Teilungswand entsteht aus einer einseitigen oder einer irisblendenartigen Invagination des Plasmalemma der Mutterzelle. Mikrotubuli markieren die Zuwachsfront der jungen Zellwand. Ein Einbau von Golgi-Vesikeln in die wachsende Wand konnte nicht sicher nachgewiesen werden. Die beobachtete Primordialwandbildung entspricht nicht der typischen Cytokinese höherer Pflanzen, sondern ähnelt eher furchungsähnlichen Teilungen niederer Eukarionten. Die phylogenetische Bedeutung dieses Zellteilungstypus, der zwischen der zentripetalen Septenbildung der Prokarionten und der zentrifugalen Zellplattenbildung der höheren Eukarionten steht, wird diskutiert.
    Notes: Summary Cytokinesis in young gemmae ofMarchantia polymorpha was studied both with light and electron microscope. The crosswall is formed by invagination of the plasmalemma either from one side or by annular ingrowth. Microtubules are found oriented rectangular to the growing septum. There is no clear evidence, whether or not the new wall is formed by coalescence of golgi vesicles. This kind of crosswall formation rather resembles the cleavage of lower algae than the cellplate formation of higher plants. ThusMarchantia may represent an intermediate stage of cell division between those plants and the annular ingrowth of the septum in procaryonta.

    Notes: Abstract The activator-encoding gene phlR was identified upstream of the plasmid-encoded operon for phenol degradation in Pseudomonas putida strain H by cassette mutagenesis and DNA sequence analysis. The deduced amino acid sequence of PHLR shows high homology to DmpR of P. putida sp. CF600 and to the chromosomally encoded PhhR of P. putida P35X reported previously. Trans-activation of phenol degradation was observed when phlR was overexpressed in a phlR insertion mutant. Transconjugants of Escherichia coli carrying pPGH11, which contains the complete set of phl genes, are unable to grow on phenol as carbon source. However, two types of mutants were selected for further characterization that were able to metabolize phenol as sole source of carbon and energy. In both types of mutants enhanced expression of phlR is responsible for the Phl+ phenotype. In type I (pPGH13) a deletion of 1 bp made the −35 region and the spacing between the −35 and −10 regions of the phlR promoter more similar to the consensus structure. In type II (pPGH14) a duplication of the phlR 5′ region was identified that includes part of the −35 motif and reduces the spacing between the −35 and −10 regions. In addition, due to the duplication of part of phlR, the distance from the phlR promoter to the catabolic phl operon is increased. Different transcriptional start sites have been identified by primer extension analysis in clones harboring pPGH14 or the wild type phlR. Quantitative primer extension analysis revealed that the greatest amount of phlR transcript is expressed from the partial, phlR duplication. Growth on phenol and phenol hydroxylase activity reflect the high level of phlR transcript in E. coli transconjugants. Overexpression of PhlR was also observed when pPGH14 was transferred into P. putida, and results in earlier induction of the phenol degradation operon relative to the wild-type strain.
