    Notes: Abstract R388rep(Ts)::Tn5 a thermosensitive, Tn5 vector (pCHR81) developed by Sasakawa and Yoshikawa [12], was found to be compatible with two strains ofErwinia carotovora and a strain ofRhizobium meliloti. pCHR81 was introduced into these organisms at lower temperatures and rendered suicidal at higher temperatures, giving rise to Tn5 transposed. To the transconjugants ofE. carotovora, which were cured of the R388 moiety and carrying Tn5 transposed, another Tn5 vector R388rep(Ts)::Tn5-Tcl (pCHR82) was re-introduced; this is a derivative of R388rep(Ts)::Tn5 with a tetracycline resistance marked instead of the original antibiotic resistances of Tn5. Gua+ gene ofE. carotovora was transferred by the cultures carrying only R388rep(Ts)::Tn5 or by those carrying R388rep(Ts)::Tn5-Tc and transposed Tn5. Though one strain of each ofAgrobacterium tumefaciens andA. radiobacter showed restriction to R388rep(Ts)::Tn5 plasmid maintenance, derivatives devoid of R388 and carrying Tn5 transposed were obtained. Streptomycin resistance gene on Tn5 was expressed in the cultures of all four species.

    Notes: Abstract Most lenses composed of spherical or aspherical surfaces have symmetrical focusing properties about an optical axis. On the other hand, unconventional optical systems such as cylindrical lenses, astigmatic or progressive power glasses have axially asymmetrical focusing properties. In this study, the method of measuring focusing properties by the moire technique is extended to asymmetrical lenses. Some unconventional lenses were experimentally tested, and the focusing power in the interesting region and azimuthal direction of the aperture was evaluated.

    Notes: Abstract. A newly established human lymphoma cell line (OZ) has the t(14;18)(q32;q21) translocation and expresses large amounts of Bcl-2 compared to CCRF-CEM cells. VP-16 (40 μg/mL), a promising agent against lymphoma, caused DNA fragmentation (26.9% of total DNA) typical for apoptosis at 6 h in CCRF-CEM cells, but no significant changes in OZ cells until 24 h after the addition of VP-16. However, coincubation with calphostin C (0.2 μg/mL), a protein kinase C (PKC) inhibitor, induced DNA fragmentation in VP-16-treated OZ cells (13.5% of total DNA) at 6 h after the treatment. Simultaneous immunoblot analysis revealed that this induction of apoptosis coincided with the downregulation of serine-phosphorylated Bcl-2 (13% of control cells). By contrast, apoptosis induced by VP-16 in CCRF-CEM cells was attenuated by the addition of 0.5 μM phorbol 12-myristate 13-acetate, a potent PKC stimulator. These observations suggest that Bcl-2 function is partly regulated by phosphorylation/dephosphorylation mechanisms of the PKC system, and that phosphorylated Bcl-2 in lymphoma cells may play a role in the prevention of apoptosis.

    Notes: Summary The composition of urinary acidic glycosaminoglycans (AGAG) in 4 patients with Werner's syndrome was determined by an enzymatic assay system using chondroitinases and hyaluronidase. In Werner's syndrome, the amount of hyaluronic acid and heparan sulfates in the total AGAG increases. A compositional change in the chondroitin sulfate isomers occurs. The change of urinary AGAG in Werner's syndrome appears to reflect age-related changes.

The phylogenetic relationship among primates, ferungulates (artiodactyls + cetaceans + perissodactyls + carnivores), and rodents was examined using proteins encoded by the H strand of mtDNA, with marsupials and monotremes as the outgroup. Trees estimated from individual proteins were compared in detail with the tree estimated from all 12 proteins (either concatenated or summing up log-likelihood scores for each gene). Although the overall evidence strongly suggests ((primates, ferungulates), rodents), the ND1 data clearly support another tree, ((primates, rodents), ferungulates). To clarify whether this contradiction is due to (1) a stochastic (sampling) error; (2) minor model-based errors (e.g., ignoring site rate variability), or (3) convergent and parallel evolution (specifically between either primates and rodents or ferungulates and the outgroup), the ND1 genes from many additional species of primates, rodents, other eutherian orders, and the outgroup (marsupials + monotremes) were sequenced. The phylogenetic analyses were extensive and aimed to eliminate the following artifacts as possible causes of the aberrant result: base composition biases, unequal site substitution rates, or the cumulative effects of both. Neither more sophisticated evolutionary analyses nor the addition of species changed the previous conclusion. That is, the statistical support for grouping rodents and primates to the exclusion of all other taxa fluctuates upward or downward in quite a tight range centered near 95% confidence. These results and a site-by-site examination of the sequences clearly suggest that convergent or parallel evolution has occurred in ND1 between primates and rodents and/or between ferungulates and the outgroup. While the primate/rodent grouping is strange, ND1 also throws some interesting light on the relationships of some eutherian orders, marsupials, and montremes. In these parts of the tree, ND1 shows no apparent tendency for unexplained convergences.

    Notes: Abstract Ninety-nine members of the salmonid HpaI and AvaIII families of short interspersed repetitive elements (SINEs) were aligned and a general consensus sequence was deduced. The presence of 26 correlated changes in nucleotides (diagnostic nucleotides) from those in the consensus sequence allowed us to divide the members of the HpaI family into 12 subfamilies and those of the AvaIII family into two subfamilies. On the basis of the average sequence divergences and the phylogenetic distributions of the subfamilies, the relative antiquity of the subfamilies and the process of sequential changes in the respective source sequences were inferred. Despite the higher mutation rates of CG dinucleotides in individual dispersed members, no hypermutability of CG positions was observed in changes in the source sequences. This result suggests that sequences of SINEs located in a nonmethylated or hypomethylated genomic region could have been selected as source sequences for retroposition and/or that some CG sites are the parts of recognition sequences of retropositional machineries.

    Notes: Abstract To clarify the phylogenetic position of Vestimentifera (tube worms), 346-bp fragments of the elongation factor-lα (EF-lα) gene (939–1286 according to the numbering of the human gene) of a vestimentiferan, Lamellibrachia sp., a sternaspid polychaete, Sternaspis scutata, an earthworm, Pheretima sp., and a gastropod, Alviniconcha hessleri, were sequenced. From the amino acid sequences of these EF-lα, and those of two other vertebrates and two arthropods, phylogenetic relationships were deduced by the maximum likelihood (ML) method, by which the phylogenetic tree can be inferred without assuming constancy of the molecular evolutionary rate. For the ML tree and all of seven alternative trees, whose log-likelihoods could not be discriminated from that of the ML tree by the criterion of the standard error, the vestimentiferan, the polychaete, and the oligochaete formed a clade, excluding the arthropods and the gastropod as outgroups. This result is convincing evidence that Vestimentifera are protostomes that are closely related to Annelida. The ML tree suggests that Vestimentifera are more closely related to Polychaeta than to Oligochaeta, though the data were not sufficient to discriminate these three groups at a significant level. From recent evidence such as morphological characteristics and molecular information, it may safely be said that vestimentiferans should be included in the Annelida provided this phylum contains polychaetes and oligochaetes.
