    Notizen: Summary This report describes a method using direct photon absorptiometry (DPA) for determination in vivo of the bone mineral content to bone width ratio (BMC/W) in the rat femur. A significant correlation was noted between measurements by DPA and whole femur dry weight and ash weight. Measurements were reproducible within repeated determinations of both the same and successive days. Soft tissue, rotation of the femur, or slight differences in the longitudinal positioning of the femur did not significantly affect usefulness of the measurements. A significant positive correlation was seen between body weight and the femur BMC/W, indicating that the method accurately reflected expected physiological changes in bone mineral. The method is suitable for the rapid, simple, accurate, and reproducible measurement of bone mineral in the anesthetized rat and is applicable to serial measurements.

    Notizen: Abstract Fibroblast cells derived from juvenile bluegill sunfish were tested for their response to the effects of known mutagens/carcinogens. The cells grew well and cloned in Eagle's minimal essential medium which contained fetal bovine serum. Growth rate was temperature dependent and increased steadily as temperature was raised from 15 to 33°C. Direct mutagens and promutagens were toxic to the cells, and fluorescence spectroscopy revealed the capability of metabolizing at least one promutagen, benzo(a)pyrene, to water-soluble intermediates. Ouabain resistant mutants were produced by exposing the cells toN-methyl-N-nitro-nitrosoguanidine (MNNG) or to benzo(a)pyrene (B(a)P). The mutant clones were resistant to 100-fold increases in concentration of the selective agent ouabain, over that which was lethal to wild type cells. Spontaneous mutation frequency to ouabain in mass cultures averaged 1.2×10−6 for all experiments. A fluctuation test confirmed an expected random occurrence of spontaneous mutations. Visible crystals formed within the cells when high concentrations of B(a)P (〉5 μg/ml) were added to the tissue culture medium. Crystal formation resulted in no apparent cell damage but did reduce the growth rate of the cultures. The crystals were rhombic, resembling those described for pure native B(a)P, and gradually disappeared when the cells were exposed to mutagen-free medium which contained serum.

    Notizen: Summary Two doses of buspirone (5 and 10 mg tds), 1 dose of diazepam (5 mg tds) and placebo were administered to 8 normal subjects for a period of 8 days. Each subject received each drug in a balanced order with a minimum interval of 1 week between courses. Psychotropic effects were assessed with a battery of physiological, psychomotor and subjective tests on the first, third and last days of treatment both before the first daily dose and at 1 h and 3 h after it. Diazepam showed a characteristic profile of action producing EEG changes and psychological impairment after a single dose which were still present after a week's treatment. Such effects were minimal after buspirone. Both drugs increased subjective ratings of drowsiness but these feelings tended to decrease after a week's treatment on the clinical doses. Buspirone (10 mg tds) produced some unpleasant side-effects initially but tolerance to these invariably developed after 3 days treatment.

    Notizen: Summary Twelve normal subjects were tested on a number of measures both before and 1,3 and 5 h after 10 mg diazepam, 100 and 200 mg tofisopam and a placebo. The measures included self-ratings of mood, bodily symptoms, hostility and sleep, the electroencephalogram (EEG), reaction time, tapping, digit symbol substitution, the symbol copying test, and plasma levels. Diazepam showed a clear profile of action, producing EEG changes, pronounced sedation and psychological impairment. The last two effects were maximal at 1 h and had worn off by 5 h. The EEG was recorded at 3 h only. Tofisopam in no way resembled diazepam. It produced no changes on the EEG or psychological tests and a very mild stimulant effect was apparent on the ratings. While diazepam was easily detectable in the blood, tofisopam did not bind to benzodiazepine receptors.

    Notizen: Summary Many aloe species in southern Africa have stems clothed with a layer of persistent dead leaves. The degree of stem coverage is species-specific. The suggestion that persistent dead leaves have an insulatory function and are adaptive in fire-prone habitat was tested on Aloe ferox. Field surveys demonstrated a significant negative relationship between mortality and degree of stem coverage and laboratory studies confirmed the insulating properties of dead leaves. The distribution of southern African tree aloes supprts the prediction that bare-stemmed species would be confined to fire-free habitat whilst fully clothed species would occur in both fire-prone and fire-free habitat. The study suggests that harvesting of Aloe ferox leaves for medicinal purposes could result in significant mortality in fires.

    Notizen: Abstract Among the many possible contributions to joint or splice loss in an optical fibre system are those attributable to the manufacturing tolerances of the fibres themselves. This paper predicts, from a probability approach, the likely effect of errors in the cladding surface diameter, the circularity of this surface and the concentricity of core and cladding. It is shown that, for one representative set of excitation conditions, the tolerances proposed in a draft CCITT recommendation lead to a mean joint loss of 0.043 dB with a standard deviation of 0.045 dB. The loss exceeds 0.33 dB with a probability of 0.001.
