    Notizen: Abstract: A time-dependent electric field gives rise to a stationary non-equilibrium current I (2) around a mesoscopic metal ring threaded by a magnetic flux. We show that this current, which is proportional to the intensity of the field, is closely related to the exchange part of the interaction contribution to the equilibrium persistent current, and that the corresponding non-linear conductivity directly measures the weak localization correction to the polarization. We explicitly calculate the disorder average of I (2) in the diffusive regime as function of the frequency of the electric field and the static flux piercing the ring, and suggest an experiment to test our theory.

    Notizen: Abstract  The De Gerlache Seamounts actually consist of two medium-sized guyots, with summits at depths of 350–600 m. Acoustic profiler data show no significant sediment on these guyots. Alkaline basalts dredged from the summit of the eastern guyot yield K/Ar ages between 20.1±1.0 and 23.2±1.2 Ma. Basement ridges and sediment-filled troughs between the guyots are associated with the prominent gravity anomaly extending north from the Antarctic margin. This structure possibly played a role in the guyot formation, however, the question of how the De Gerlache Seamounts are related to this gravity anomaly remains uncertain.

    Notizen: Abstract We report on the application of a pyrogallol red-vanadium complex (PR-V) for ultracytochemical staining of proteinaceous structures in animal tissues and cell cultures. This dye may be used as a general purpose stain in electron microscopy. In contrast to osmium tetroxide, the price of the material is low and no toxic vapors are produced. The PR-V complex was prepared by addition of vanadium (IV) oxide sulfate to pyrogallol red dissolved in acetate buffer (pH 5.6). The formation of the complex was indicated by a color change from purplered (λmax=520 nm) to violet (λmax=539 nm) which occurred at equimolar concentrations of the dye and the metal salt. Under these conditions PR-V was stable for several days. The mechanism of PR-V binding was checked in dot blots using different proteins as well as heparin for control. While heparin remained unstained, proteins were stained in a dose-dependent manner. Deamination of proteins with nitric oxide strongly reduced PR-V staining in dot blots as well as in cell cultures. Optimal staining results of animal cells and tissues were obtained in specimens that had been mildly fixed for at least 1 h or longer with a mixture of 0.1% glutaraldehyde and 1.0% paraformaldehyde dissolved in phosphate-buffered saline, pH 7.2, washed with acetate buffer, pH 5.6, and subsequently treated with PR-V in the presence of 50% ethanol at room temperature. Control specimens without PR-V but treated en bloc with uranyl acetate or sodium molybdate showed similar contrast but less details in the ultrastructure of the tissue. All specimens were embedded in epoxy resin and ultrathin sections were stained conventionally with uranyl and lead salt solutions. In electron micrographs, membrane-associated particles, stress fibers and filaments of the cell cortex, collagen fibrils, tight junctions and desmosomes, and other proteinaceous components were clearly visualized only in the PR-V-treated speciments. In conclusion, the ability to bind selectively and specifically to proteinaceous structures makes PR-V a versatile stain to study the localization and distribution of these structures in cells and tissues at the ultrastructural level.

    Notizen: Abstract We have examined production of mediumchain fatty acids by Brassica napus L. plants transformed with a California bay (Umbellularia californica) medium-chain acyl-acyl carrier protein (ACP) thioesterase (UcFatB1) cDNA under the control of the constitutive cauliflower mosaic virus 35S promoter. These plants were found to accumulate medium-chain fatty acids in seeds but not in leaves or roots. Assay of thioesterase activity in extracts of leaves indicated that lauroyl-ACP thioesterase activity is comparable to oleoyl-ACP thioesterase (EC 3.1.2.14) activity in transformant leaves. Furthermore, leaf lauroyl-ACP thioesterase activity was in excess of that which produced a significant increase in the amount of laurate (12:0) in seed. Studies in which isolated chloroplasts were 14C-labelled were used to evaluate whether medium-chain fatty acids were produced in transformed leaves. Up to 34% of the fatty acids synthesized in vitro by isolated chloroplasts were 12:0. These results demonstrate that the normally seed-localized lauroyl-ACP thioesterase can be expressed in active form in leaves, imported into chloroplasts and can access acyl-ACP intermediates of leaf de-novo fatty acid synthesis. The most likely explanation for the lack of accumulation of 12:0 in transformed leaves is its rapid degradation by β-oxidation. In support of this hypothesis, isocitrate lyase (EC 4.1.3.1) activity was found to be significantly increased in plants transformed with 35S-UcFatB1.

    Beschreibung / Inhaltsverzeichnis: Zusammenfassung Die mikrophotometrische Auswertung von Spektren erlaubt über den Vergleich mit den Spektren eines UV-Normales nach Rössler, Krefft und Rüttenauer die spektrale Emission eines Quecksilber-Einspritz-Brenners zu bestimmen. Schon geringe Beimengungen zum in den Kern der Entladung injizierten Quecksilber verändern die spektrale Strahlungsverteilung. Brennt die Entladung mit kathodischem Brennfleckensatz in einer Gasatmosphäre, so liegt in der Wahl dieses Zusatzstoffes eine weitere Möglichkeit, die spektrale Emission des Quecksilber-Einspritz-Brenners zu variieren. Über diese Spektren und ihre Interpretation wird in der vorliegenden Arbeit berichtet.

    Notizen: Abstract Glucose-limited continuous cultures were used to analyze σB activity at decreasing growth rates. Expression of the σB-dependent genes gsiB and ctc started to increase at a growth rate of 0.2 h–1, and both genes were induced approximately fivefold at a growth rate of 0.1 h–1 as compared to expression at the maximal growth rate. However, maximal σB activity was only reached when the growth stopped as a result of the exhaustion of the carbon and energy source glucose. During glucose-limited growth, increased expression of the general stress regulon at growth rates below 0.2 h–1 did not provide wild-type cells with a growth advantage over sigB mutants. Instead, expression of the stress regulon seems to constitute a significant burden during glucose-limited growth, resulting in a selective growth advantage of the sigB mutant as compared to the wild-type at a growth rate of 0.08 h–1.

    Notizen: Abstract In end-stage heart failure due to idiopathic dilated cardiomyopathy β1-adrenoceptors are downregulated and G1α-proteins are upregulated. The aim of the present study was to investigate the influence of the angiotensin-converting enzyme inhibitor captopril on β-adrenoceptor density and Giα-proteins in sequential endomyocardial biopsies. Nineteen patients with mild to moderate congestive heart failure due to idiopathic dilated cardiomyopathy (NYHA Class II–III) were studied before and after 8–11 weeks of therapy. Patients were randomised into a captopril and a control group; 9 patients received captopril 12.5–50 mg per day, (divided in 2–3 doses) p.o. in addition to “conventional” therapy with digoxin and diuretics, and 10 controls received “conventional” therapy only. Echocardiography, spiroergometry, right heart catheterisation and endomyocardial biopsies were performed before (baseline) and after treatment. Compared to baseline, captopril increased total β-adrenoceptor density by selectively increasing β1-adrenoceptors (31.6 vs 41.2 fmol·mg−1; p〈0.05) but had no significant effect on Giα-proteins. The results indicate that treatment with angiotensin-converting enzyme inhibitors partly restores myocardial β1-adrenoceptor density, and this action effect may contribute to the clinical improvement of patients with idiopathic dilated cardiomyopathy treated in this way.

    Notizen: Abstract This article describes an approach to the development of highly reliable and reusable CORBA applications. Interface description language (IDL) specifications are extended with LOTOS specifications of the functional and dynamic behavior of the interface operations. These extended interface specifications are compiled into header files and stub code using standard CORBA mechanisms. The LOTOS part can be compiled into C-code yielding prototype implementations. An extensible set of LOTOS tools is used to test and prove critical properties of distributed applications. From the event-oriented LOTOS description, a state-oriented program in a CSP-like programming language is derived. This program can be analyzed using very efficient, BDD-based model checking verification tools. Furthermore, it serves as an ideal intermediate step between verified, high level, reusable LOTOS processes which solve typical distributed programming tasks and their implementation as CORBA objects in production programming languages.
