    Description: In a multidisciplinary ex situ experiment, benthic bacterial deep-sea communities from 2,500 m water depth at the Long-Term Ecological Research Observatory HAUSGARTEN (stationPS93/050-5 and 6), were retrieved using a TV-guided multiple corer. Surface sediments (0 - 2 cm) of 16 cores were mixed with sterile filtered deep-sea water to a final sediment dilution of 3.5 fold. The slurries were split and supplemented with five different types of habitat-related detritus: chitin, as the most abundant biopolymer in the oceans, and four different naturally occurring Arctic algae species, i.e. Thalassiosira weissflogii, Emiliania huxleyi, Bacillaria sp. and Melosira arctica. Incubations were performed in five replicates, at in situ temperature and at atmospheric pressure, as well as at in situ pressure of 250 atm. At the start of the incubation and after 23 days, changes in key community functions, i.e. extracellular enzymatic activity, oxygen respiration and secondary production of biomass (bacterial cell numbers and biomass), were assessed along with changes in the bacterial community composition based on 16S rRNA gene and 16S rRNA Illumina sequencing. In summary, differences in community structure and in the uptake and remineralization of carbon in the different treatments suggest an effect of organic matter quality on bacterial diversity as well as on carbon turnover at the seafloor.

    Description: We determined the concentrations of different types of bacteria via Catalysed reporter deposition-fluorescence in situ hybridization (CARD-FISH) at different depths of the water column as well as several centimeters of the surficial deep-sea sediment. Sampling sites were stations HGIV, EGI, and N4 of the long-term ecological research observatory HAUSGARTEN in the Fram Strait.

    Description: Samples in this dataset were collected at the long-term ecological research (LTER) site HAUSGARTEN in Fram Strait, and the central Arctic Ocean. On board, the samples were fixed with formalin in a final concentration of 2% for 10 – 12 hours, then filtered onto 0.2 µm polycarbonate Nucleopore Track-Etched filters, and stored at -20°C for further analysis. Cell abundances of the groups Alteromonas, Bacteroidia, Polaribacter, Gammaproteobacteria and the SAR11 clade were asses using CAtalyzed reporter deposition Fluorescence In Situ Hybridization (CARD-FISH) following the protocol established by (Pernthaler et al., 2002). The filters were evaluated microscopically under an automated microscope (Zeder et al., 2011). Cell enumeration was performed with the software Automated Cell Measuring and Enumeration Tool (ACMETool3, 2018-11-09; Zeder et al., 2011). Cells were counted as objects according to manually defined parameters separately for the DAPI and FISH channels.

    Description: The mineralogy and P, Fe and Mn contents of basal metalliferous and nonmetalliferous sediments of two Deep Sea Drilling Project cores (Sites 77 and 80) were investigated. When compared with superjacent nonmetalliferous material, basal metalliferous sediments are significantly enriched in P, Fe and Mn. Among the phases present in the metalliferous sediment samples are a poorly-crystalline Fe-rich smectite and X-ray amorphous Fe oxyhydroxides. P is associated primarily with the oxyhydroxides. Adsorption of phosphate on smectite does not seem to play any role in the uptake of P by metalliferous sediments. An estimate of the removal of P by on- and off-ridge metallogenic deposition suggests that this process strongly affects the overall geochemical balance of phosphorus in the World Ocean.
