    Notes: : Cores were obtained from several tree species located both on a river floodplain and a nearby terrace. A ratio of annual tree growth on floodplains to terrace growth was developed and shown to be related to the annual river discharge. Growth ratios from the time prior to written records can therefore be used to reconstruct river discharge and infer past unrecorded flood frequency. Oak and basswood ratios yielded the best models for discharge reconstruction, whereas those of elm and birch were less useful. This method permits reconstruction of river discharge from an assemblage of growth cores obtained within a relatively small area.

    Notes: : Drought is evaluated in terms of the magnitude and duration of the 1988 spring and summer precipitation shortfall, and according to various components of the hydrologic budget, both surface and sub-surface. The response time of some of these components is investigated, relative to the time of precipitation. Individual water users perceived a beginning and ending of the drought at different times relative to their activities. Some statistics better describe some components of a drought to some users, and better answer some questions, than do others.

    Notes: SYNOPSIS. We demonstrated previously microbodies in Euglena gracilis grown in the dark on 2-carbon substrates. We have now established in Euglena the particulate nature of enzymes known in other organisms to be localized in microbodies (glyoxysomes and leaf peroxisomes). On a linear sucrose gradient the glyoxylate cycle enzymes band together at a nigner equilibrium density (1.20 g/cm3) than mitochondrial marker enzymes (1.17 g/cm3), establishing the existence in Euglena of glyoxysomes similar to those of higher plants. Glyoxylate (hydroxypyruvate) reductase and, under certain conditions, also glycolate dehydrogenase co-band with the glyoxylate cycle enzymes, suggesting that Euglena glyoxysomes, like those of higher plants, may contain peroxisomal-type enzymes. Catalase, an enzyme characteristic of microbodies from a variety of sources, was not detected in Euglena.

    Notes: . The life cycle of Vairimorpha necatrix was studied by electron microscopy. Disporous development has two distinct stages: 1) diplokaryotic meronts which are actively mitotic, and 2) diplokaryotic sporonts which are distinguished by reduced ribosome density and a thickened plasmalemma. After final division of the sporont, sporoblasts form spores which are ovocylindrical and measure 4.4 ± 0.08 × 2.3 ± 0.05 μm (mean ± SE). Octosporous development results in eight haploid spores being formed in a sporophorous vesicle. The uninucleate octospores were smaller than the binucleate dispores and the exospore was thicker but less crenulate in outline. Early in octosporogony, tubules are produced from the sporont plasmalemma and electron-dense material accumulates in the episporontal space. The latter may be amorphous, vesiculated, or vacuolated in appearance and in later stages may take a stacked, lamellar form. At sporoblast formation, exospore material coats the plasmalemma and attached tubules; all inclusions in the episporontal space gradually disappear as spores are formed. These secretory products may have application to taxonomic distinction at the species level.

Electron microscope and time lapse, phase contrast cinematography studies on Babesia equi organisms within equine red blood cells revealed that this hemoprotozoon has two organelles possibly involved with ingestion of nutrients: a cytostome that takes in hemoglobin from the host cell and a tubule that extends from the main body of the parasite through the erythrocyte to the blood plasma and appears to ingest plasma during periods of rapid growth and development.

    Notes: SYNOPSIS. We demonstrated previously the presence of glyoxysomes containing the glyoxylate cycle enzymes in Euglena gracilis grown in the dark on ethanol. We have now established that the glyoxysomes of Euglena grown on hexanoate also contain the following enzymes of the pathway for β-oxidation of fatty acids: hexanoyl-CoA synthetase, 3-β-hydroxyacyl-CoA dehydrogenase and thiolase. Estimations of specific activities indicate that these enzymes are over 20 times as active in glyoxysomes as they are in mitochondria, suggesting that the β-oxidation of fatty acids occurs almost entirely in Euglena glyoxysomes under these conditions. Thus, the entire portion of the gluconeogenic pathway from fatty acid to succinate is localized in the glyoxysome of Euglena.

    Notes: : Illinois data from 168 months (1986–1999) were investigated to determine the responses of surface-water and ground-water resources to precipitation. Such responses were generally within the month of occurrence or one to two months later, with recovery being reached another one to three months into the future, depending on season of the year. Although the drought of 1988 immediately impacted surface-water and ground-water resources, the time of recovery was substantially longer compared to those of individual dry months, generally continuing for several months. The extremely wet summer of 1993 resulted in elevated responses in water resources almost immediately, but in this instance continued through the following fall and winter, into the spring of 1994.

    Description: The mechanistic basis of regulatory variation and the prevailing evolutionary forces shaping that variation are known to differ between sexes and between chromosomes. Regulatory variation of gene expression can be due to functional changes within a gene itself ( cis ) or in other genes elsewhere in the genome ( trans ). The evolutionary properties of cis mutations are expected to differ from mutations affecting gene expression in trans . We analyze allele-specific expression across a set of X substitution lines in intact adult Drosophila simulans to evaluate whether regulatory variation differs for cis and trans , for males and females, and for X-linked and autosomal genes. Regulatory variation is common (56% of genes), and patterns of variation within D. simulans are consistent with previous observations in Drosophila that there is more cis than trans variation within species (47% vs. 25%, respectively). The relationship between sex-bias and sex-limited variation is remarkably consistent across sexes. However, there are differences between cis and trans effects: cis variants show evidence of purifying selection in the sex toward which expression is biased, while trans variants do not. For female-biased genes, the X is depleted for trans variation in a manner consistent with a female-dominated selection regime on the X. Surprisingly, there is no evidence for depletion of trans variation for male-biased genes on X. This is evidence for regulatory feminization of the X, trans -acting factors controlling male-biased genes are more likely to be found on the autosomes than those controlling female-biased genes.

    Description: We explore the evolutionary origins of dosage compensation (DC) in sex chromosomes in the context of metabolic control theory. We consider first the cost of gene loss (hemizygosity) per se in reducing flux, and examine two relationships between flux and fitness (linear and Gaussian) to calculate a fitness cost of hemizygosity. Recognizing that new sex chromosomes are derived from autosomes, we also calculate the cost of unmasking deleterious mutations segregating on the nascent sex chromosomes as loci become hemizygous. The importance of deleterious mutations to the fitness cost of hemizygosity depends on their frequency, and on the relative costs of halving gene dose for wild-type alleles. We then consider the evolution of DC in response to gene loss, and include a cost of overexpression (i.e., DC such that expression exceeds the wild-type homozygote). Even with costs to excess flux, hypomorphic mutations can cause the optimal level of DC to be higher than 2-fold when the absolute cost of hemizygosity is small. Finally, we propose a three-step model of DC evolution: 1) once recombination ceases and the Y begins to deteriorate, genes from longer metabolic pathways should be lost first, as halving these genes does not drastically reduce flux or, thereby, fitness; 2) both the cost of hemizygosity and the presence of hypomorphic mutations will drive an increase in expression, that is, DC; 3) existing DC will now permit loss of genes in short pathways.
