    Description: The histone chaperone Rtt106 binds histone H3 acetylated at lysine 56 (H3K56ac) and facilitates nucleosome assembly during several molecular processes. Both the structural basis of this modification-specific recognition and how this recognition informs Rtt106 function are presently unclear. Guided by our crystal structure of Rtt106, we identified two regions on its double-pleckstrin homology domain architecture that mediated histone binding. When histone binding was compromised, Rtt106 localized properly to chromatin but failed to deliver H3K56ac, leading to replication and silencing defects. By mutating analogous regions in the structurally homologous chromatin-reorganizer Pob3, we revealed a conserved histone-binding function for a basic patch found on both proteins. In contrast, a loop connecting two β-strands was required for histone binding by Rtt106 but was dispensable for Pob3 function. Unlike Rtt106, Pob3 histone binding was modification-independent, implicating the loop of Rtt106 in H3K56ac-specific recognition in vivo. Our studies described the structural origins of Rtt106 function, identified a conserved histone-binding surface, and defined a critical role for Rtt106:H3K56ac-binding specificity in silencing and replication-coupled nucleosome turnover.

    Notes: Abstract New thermostable enzyme activities of seven Thermus strains were compared using the API ZYM system. All the strains exhibited high levels of α- and β-glycosidases, esterase (C4) and esterase-lipase (C8) activities intracellularly. Only T. thermophilus HB8 (ATCC 27634) showed α-glucosidase and esterase activities in the supernatant. According to the intensity of β-galactosidase activity, Thermus strains were divided in three groups. Group 0, which showed a weak β-galactosidase activity, included Thermus spp. ATCC 31674 (T351) and 27978 (X-1) as well as T. thermophilus ATCC 27634 (HB8). Group I which consisted of T. aquaticus ATCC 25104 (YT-1), ATCC 25105 (Y-VII-51B) and Thermus sp. ATCC 27737 (T2), had a specific activity of approximately 40.0 U mg−1 and galactose as inducer. T. aquaticus ATCC 31558 (group 2) was particularly effective for β-galactosidase production (2840 U) with a specific activity of 98 U mg−1. For each strain, galactose (0.5%) was a better inducer of β-galactosidase production than lactose (1%). The detection of β-galactosidase activity was dependent on the derivative chromogenic substrates used (naphthyl or nitrophenol coupled to sugar). Oligosaccharides were synthesized from cellobiose, lactulose, maltose or lactose as substrates at high temperature in some strains of Thermus.

    Notes: Abstract A method of computing driving tractions for phase transformations is presented. The driving traction calculation requires evaluating the jump in the displacement gradient and the average stress acting across the interface. A bimaterial Green's function is used in a boundary element formulation for the calculation. The Green's function satisfies all interface boundary conditions for the bimaterial providing an efficient formulation for evaluating the necessary jump and average interface terms. We derive a boundary integral equation for calculating the driving traction and apply the developed numerical tool to driving traction calculations in copper-aluminum-nickel.

    Notes: Abstract We present a boundary integral formulation for anisotropic interface crack problems based on an exact Green's function. The fundamental displacement and traction solutions needed for the boundary integral equations are obtained from the Green's function. The traction-free boundary conditions on the crack faces are satisfied exactly with the Green's function so no discretization of the crack surfaces is necessary. The analytic forms of the interface crack displacement and stress fields are contained in the exact Green's function thereby offering advantage over modeling strategies for the crack. The Green's function contains both the inverse square root and oscillatory singularities associated with the elastic, anisotropic interface crack problem. The integral equations for a boundary element analysis are presented and an example problem given for interface cracking in a copper-nickel bimaterial.
