    Description: The seed is the pharmaceutical and breeding organ of Cassia obtusifolia, a well-known medical herb containing aurantio-obtusin (a kind of anthraquinone), food, and landscape. In order to understand the molecular mechanism of the biosynthesis of aurantio-obtusin, seed formation and development, and stress response of C. obtusifolia, it is necessary to understand the genomics information. Although previous seed transcriptome of C. obtusifolia has been carried out by short-read next-generation sequencing (NGS) technology, the vast majority of the resulting unigenes did not represent full-length cDNA sequences and supply enough gene expression profile information of the various organs or tissues. In this study, fifteen cDNA libraries, which were constructed from the seed, root, stem, leaf, and flower (three repetitions with each organ) of C. obtusifolia, were sequenced using hybrid approach combining single-molecule real-time (SMRT) and NGS platform. More than 4,315,774 long reads with 9.66 Gb sequencing data and 361,427,021 short reads with 108.13 Gb sequencing data were generated by SMRT and NGS platform, respectively. 67,222 consensus isoforms were clustered from the reads and 81.73% (61,016) of which were longer than 1000 bp. Furthermore, the 67,222 consensus isoforms represented 58,106 nonredundant transcripts, 98.25% (57,092) of which were annotated and 25,573 of which were assigned to specific metabolic pathways by KEGG. CoDXS and CoDXR genes were directly used for functional characterization to validate the accuracy of sequences obtained from transcriptome. A total of 658 seed-specific transcripts indicated their special roles in physiological processes in seed. Analysis of transcripts which were involved in the early stage of anthraquinone biosynthesis suggested that the aurantio-obtusin in C. obtusifolia was mainly generated from isochorismate and Mevalonate/methylerythritol phosphate (MVA/MEP) pathway, and three reactions catalyzed by Menaquinone-specific isochorismate synthase (ICS), 1-deoxy-d-xylulose-5-phosphate synthase (DXS) and isopentenyl diphosphate (IPPS) might be the limited steps. Several seed-specific CYPs, SAM-dependent methyltransferase, and UDP-glycosyltransferase (UDPG) supplied promising candidate genes in the late stage of anthraquinone biosynthesis. In addition, four seed-specific transcriptional factors including three MYB Transcription Factor (MYB) and one MADS-box Transcription Factor (MADS) transcriptional factors) and alternative splicing might be involved with seed formation and development. Meanwhile, most members of Hsp20 genes showed high expression level in seed and flower; seven of which might have chaperon activities under various abiotic stresses. Finally, the expressional patterns of genes with particular interests showed similar trends in both transcriptome assay and qRT-PCR. In conclusion, this is the first full-length transcriptome sequencing reported in Caesalpiniaceae family, and thus providing a more complete insight into aurantio-obtusin biosynthesis, seed formation and development, and stress response as well in C. obtusifolia.

    Description: The effects of temperature on bulk hydrogen concentration and diffusion have been tested with the Devanathan–-Stachurski method. Thus, a model based on hydrogen potential, diffusivity, loading frequency, and hydrostatic stress distribution around crack tips was applied in order to quantify the temperature’s effect. The theoretical model was verified experimentally and confirmed a temperature threshold of 320 K to maximize the crack growth. The model suggests a nanoscale embrittlement mechanism, which is generated by hydrogen atom delivery to the crack tip under fatigue loading, and rationalized the ΔK dependence of traditional models. Hence, this work could be applied to optimize operations that will prolong the life of the pipeline.

    Description: With the development of the “Hallyu” (Korean Wave) and people’s deep understanding of South Korea, the phenomenon of “Hallyu” idols driving tourism has become very common. Tourists, especially fans are no longer satisfied with the traditional tourist attractions with special symbolic meanings when visiting South Korea, but instead focus on common places associated with idols. For example, restaurants, cafes, bookstores, parks and convenience stores that have appeared in an idol’s social network accounts. With the application of the theory of attachment and sense of place in the tourism field, this study will start from the celebrity attachment, to exploring the specific processes and mechanisms by which idols affect the behavioral intentions of the fans. The associations between visitors and idol-related places based on celebrity attachments has also become possible, and the fans/tourists’ opinions and attitudes toward idol-related places will also change due to this emotional attachment. Relevant data were collected in the form of online questionnaires, and 440 valid responses were finalized for data analysis. Through data analysis, all the hypotheses proposed in this paper are supported, and there are significant relationships and positive influences among the variables. Data analysis results show that idol attachment can positively influence the four cognitive dimensions of sense of place (place identity, place dependence, social bonding and atmosphere). The various dimensions of sense of place can promote the generation of place attachment.

    Description: As a kind of Amaryllidaceae alkaloid which is accumulated in the species of Lycoris plants, lycorine has a range of physiological effects. The biosynthesis pathway of lycorine has been partly revealed, but the transport and accumulation mechanisms of lycorine have rarely been studied. In this study, an ATP-binding cassette (ABC) transporter from Lycoris aurea (L’Hér) Herb., namely LaABCB11, was cloned and functionally characterized. Heterologous expression showed that LaABCB11 transported lycorine in an outward direction, increased the tolerance of yeast cells to lycorine, and caused a lower lycorine accumulation in transformants than control or mutant in yeast. LaABCB11 is associated with the plasma membrane, and in situ hybridization indicated that LaABCB11 was mainly expressed in the phloem of leaves and bulbs, as well as in the cortical cells of roots. These findings suggest that LaABCB11 functions as a lycorine transport and it might be related to the translocation and accumulation of lycorine from the leaves and bulbs to the roots.
