    Description: Adoptive cell therapy is an emerging anti-cancer modality, whereby the patient’s own immune cells are engineered to express T-cell receptor (TCR) or chimeric antigen receptor (CAR). CAR-T cell therapies have advanced the furthest, with recent approvals of two treatments by the Food and Drug Administration of Kymriah (trisagenlecleucel) and Yescarta (axicabtagene ciloleucel). Recent developments in proteomic analysis by mass spectrometry (MS) make this technology uniquely suited to enable the comprehensive identification and quantification of the relevant biochemical architecture of CAR-T cell therapies and fulfill current unmet needs for CAR-T product knowledge. These advances include improved sample preparation methods, enhanced separation technologies, and extension of MS-based proteomic to single cells. Innovative technologies such as proteomic analysis of raw material quality attributes (MQA) and final product quality attributes (PQA) may provide insights that could ultimately fuel development strategies and lead to broad implementation.

    Description: Systemic toxicity and severe side effects are commonly associated with anticancer chemotherapies. New strategies based on enhanced drug selectivity and targeted delivery to cancer cells while leaving healthy tissue undamaged can reduce the global patient burden. Herein, we report the design, synthesis and characterization of a bio-inspired hybrid multifunctional drug delivery system based on diatom microalgae. The microalgae’s surface was chemically functionalized with hybrid vitamin B12-photoactivatable molecules and the materials further loaded with highly active rhenium(I) tricarbonyl anticancer complexes. The constructs showed enhanced adherence to colorectal cancer (CRC) cells and slow release of the chemotherapeutic drugs. The overall toxicity of the hybrid multifunctional drug delivery system was further enhanced by photoactivation of the microalgae surface. Depending on the construct and anticancer drug, a 2-fold increase in the cytotoxic efficacy of the drug was observed upon light irradiation. The use of this targeted drug delivery strategy, together with selective spatial–temporal light activation, may lead to lower effective concentration of anticancer drugs, thereby reducing medication doses, possible side effects and overall burden for the patient.

    Description: States are urged frequently by the UN, policymakers, and activists to recognise the human right to water domestically. However, does such legal incorporation, often in national constitutions, affect water policy and the realisation of the right? While several qualitative studies report positive impacts, initial quantitative assessments have questioned the systematic positive impact of the national recognition of the human right to water. Yet, such quantitative analyses of the effects of constitutional rights to water often overlook important mediating policy factors. We test specifically whether strong democratic governance is a significant condition for ensuring that the constitutional recognition of the human right to water has concrete outcomes. Results of a multivariate regression analysis on a global sample of 123 states over a 15-year period provide two findings. First, the constitutionalisation of the right to water and other economic, social, and cultural rights (ESCRs), in national constitutions alone is not associated with material benefits related to the human right to water. Second, the constitutionalisation of those rights can have positive material benefits for water access when the rights are foregrounded in democratic governance.

    Description: Developments in the capabilities and affordability of unmanned aerial vehicles (UAVs) have led to an explosion in their use for a range of ecological and agricultural remote sensing applications. However, the ubiquity of visible light cameras aboard readily available UAVs may be limiting the application of these devices for fine-scale, high taxonomic resolution monitoring. Here we compare the use of RGB and multispectral cameras deployed aboard UAVs for assessing intertidal and shallow subtidal marine macroalgae to a high taxonomic resolution. Our results show that the diverse spectral profiles of marine macroalgae naturally lend themselves to remote sensing and habitat classification. Furthermore, we show that biodiversity assessments, particularly in shallow subtidal habitats, are enhanced using six-band discrete wavelength multispectral sensors (81% accuracy, Cohen’s Kappa) compared to three-band broad channel RGB sensors (79% accuracy, Cohen’s Kappa) for 10 habitat classes. Combining broad band RGB signals and narrow band multispectral sensing further improved the accuracy of classification with a combined accuracy of 90% (Cohen’s Kappa). Despite notable improvements in accuracy with multispectral imaging, RGB sensors were highly capable of broad habitat classification and rivaled multispectral sensors for classifying intertidal habitats. High spatial scale monitoring of turbid exposed rocky reefs presents a unique set of challenges, but the limitations of more traditional methods can be overcome by targeting ideal conditions with UAVs.

    Description: This research was motivated by the nurses’ decision-making process in the current emergency department (ED) triage process in the United States. It explores how continuous vital signs monitoring can be integrated into the ED. The article presents four shortcomings on current ED triage systems and proposes a new conceptual clinical decision support model that exploits the benefits of combining wireless wearable devices with Multi-Attribute Utility Theory to address those shortcomings. A literature review was conducted using various engineering and medical research databases, analyzing current practices and identifying potential improvement opportunities. The results from the literature review show that advancements in wireless wearable devices provide opportunities to enhance current ED processes by monitoring patients while they wait after triage and, therefore, reduce the risk of an adverse event. A dynamic mathematical decision support model to prioritize patients is presented, creating a feedback loop in the ED. The coupling of wearable devices (to collect data) with decision theory (to synthesize and organize the information) can assist in reducing sources of uncertainty inherent to ED systems. The authors also address the feasibility of the proposed conceptual model.

    Description: Coastal marine ecosystems are under stress, yet actionable information about the cumulative effects of human impacts has eluded ecologists. Habitat-forming seaweeds in temperate regions provide myriad irreplaceable ecosystem services, but they are increasingly at risk of local and regional extinction from extreme climatic events and the cumulative impacts of land-use change and extractive activities. Informing appropriate management strategies to reduce the impacts of stressors requires comprehensive knowledge of species diversity, abundance and distributions. Remote sensing undoubtedly provides answers, but collecting imagery at appropriate resolution and spatial extent, and then accurately and precisely validating these datasets is not straightforward. Comprehensive and long-running monitoring of rocky reefs exist globally but are often limited to a small subset of reef platforms readily accessible to in-situ studies. Key vulnerable habitat-forming seaweeds are often not well-assessed by traditional in-situ methods, nor are they well-captured by passive remote sensing by satellites. Here we describe the utility of drone-based methods for monitoring and detecting key rocky intertidal habitat types, the limitations and caveats of these methods, and suggest a standardised workflow for achieving consistent results that will fulfil the needs of managers for conservation efforts.
