    Description: : The frequency and intensity of heat waves (HWs) has increased in subtropical regions in recent years. The mechanism underlying the HW response of subtropical trees remains unclear. In this study, we conducted an experiment with broad-leaved Schima superba (S. superba) and coniferous Cunninghamia lanceolata (C. lanceolata) seedlings to examine HW (5-day long) effects on stem water transport, leaf water use efficiency (WUE), morphology and growth, and to elucidate differences in the responses of both species. Our results indicated that HWs can significantly reduce hydraulic conductivity in both species. C. lanceolata experienced significant xylem embolism, with the percentage loss of conductivity (PLC) increasing by 40%, while S. superba showed a non-significant increase in PLC (+25%). Furthermore, HW also caused a reduction in photosynthesis rates (An), but transpiration rates (Tr) increased on the 5th day of the HW, together leading to a significant decrease in leaf WUE. From diurnal dynamics, we observed that the HW caused significant decrease of S. superba An only in the morning, but nearly the all day for C. lanceolata. During the morning, with a high vapor pressure deficit (VPD) environment, the HW increased Tr, which contributed a lot to latently cooling the foliage. In comparing the two tree species, we found that HW effects on S. superba were mostly short-term, with leaf senescence but limited or no xylem embolism. The surviving S. superba recovered rapidly, forming new branches and leaves, aided by their extensive root systems. For C. lanceolata, continued seedling growth initially but with subsequent xylem embolism and withering of shoots, led to stunted recovery and regrowth. In conclusion, apart from the direct thermal impacts caused by HW, drought stress was the main cause of significant negative effects on plant water transport and the photosynthetic system. Furthermore, S. superba and C. lanceolata showed clearly different responses to HW, which implies that the response mechanisms of broad-leaved and coniferous tree species to climate change can differ.

    Description: A real scalar variable integral is known in the literature by different names in different disciplines. It is basically a Bessel integral called specifically Krätzel integral. An integral transform with this Krätzel function as kernel is known as Krätzel transform. This article examines some mathematical properties of Krätzel integral, its connection to Mellin convolutions and statistical distributions, its computable representations, and its extensions to multivariate and matrix-variate cases, in both the real and complex domains. An extension in the pathway family of functions is also explored.

    Description: Starches of low digestibility are associated with improved glucose metabolism. We hypothesise that a lowly digestible-starch diet (LDD) versus a highly digestible-starch diet (HDD) improves the capacity to oxidise starch, and that this is sex-dependent. Mice were fed a LDD or a HDD for 3 weeks directly after weaning. Body weight (BW), body composition (BC), and digestible energy intake (dEI) were determined weekly. At the end of the intervention period, whole-body energy expenditure (EE), respiratory exchange ratio (RER), hydrogen production, and the oxidation of an oral 13C-labelled starch bolus were measured by extended indirect calorimetry. Pancreatic amylase activity and total 13C hepatic enrichment were determined in females immediately before and 4 h after administration of the starch bolus. For both sexes, BW, BC, and basal EE and RER were not affected by the type of starch, but dEI and hydrogen production were increased by the LDD. Only in females, total carbohydrate oxidation and starch-derived glucose oxidation in response to the starch bolus were higher in LDD versus HDD mice; this was not accompanied by differences in amylase activity or hepatic partitioning of the 13C label. These results show that starch digestibility impacts glucose metabolism differently in females versus males.

    Description: Human macrophage inflammatory protein 3α (MIP-3α), also known as CCL20, is a 70 amino acid chemokine that selectively binds and activates chemokine receptor 6 (CCR6). This chemokine is responsible for inducing the migration of immature dendritic cells, effector, or memory T-cells, and B-cells. Moreover, the MIP-3α protein has been shown to display direct antimicrobial, antiviral and antiprotozoal activities. Because of the potential therapeutic uses of this protein, the efficient production of MIP-3α is of great interest. However, bacterial recombinant production of the MIP-3α protein has been limited by the toxicity of this extremely basic protein (pI 9.7) toward prokaryotic cells, and by solubility problems during expression and purification. In an attempt to overcome these issues, we have investigated the bacterial recombinant expression of MIP-3α by using several common expression and fusion tags, including 6× histidine (His), small ubiquitin modifier protein (SUMO), thioredoxin (TRX), ketosteroid isomerase (KSI), and maltose binding protein (MBP). We have also evaluated a recently introduced calmodulin (CaM)-tag that has been used for the effective expression of many basic antimicrobial peptides (AMPs). Here, we show that the CaM fusion tag system effectively expressed soluble MIP-3α in the cytoplasm of Escherichia coli with good yields. Rapid purification was facilitated by the His-tag that was integrated in the CaM-fusion protein system. Multidimensional nuclear magnetic resonance (NMR) studies demonstrated that the recombinant protein was properly folded, with the correct formation of disulfide bonds. In addition, the recombinant MIP-3α had antibacterial activity, and was shown to inhibit the formation of Pseudomonas aeruginosa biofilms.

    Description: Human donor milk (HDM) provides appropriate nutrition and offers protective functions in preterm infants. The aim of the study is to examine the impact of different storage conditions on the stability of the human breast milk peptidome. HDM was directly frozen at −80 °C or stored at −20 °C (120 h), 4 °C (6 h), or room temperature (RT for 6 or 24 h). The milk peptidome was profiled by mass spectrometry after peptide collection by ultrafiltration. Profiling of the peptidome covered 3587 peptides corresponding to 212 proteins. The variance of the peptidome increased with storage temperature and time and varied for different peptides. The highest impact was observed when samples were stored at RT. Smaller but significant effects were still observed in samples stored at 4 °C, while samples showed highest similarity to those immediately frozen at −80 °C when stored at −20 °C. Peptide structures after storage at RT for 24 h point to the increased activity of thrombin and other proteases cleaving proteins at lysine/arginine. The results point to an ongoing protein degradation/peptide production by milk-derived proteases. They underline the need for immediate freezing of HDM at −20 °C or −80 °C to prevent degradation of peptides and enable reproducible investigation of prospectively collected samples.

    Description: Renal cell carcinoma (RCC) is a heterogeneous malignancy which often develops and progresses asymptomatically. Benign oncocytomas are morphologically similar to malignant chromophobe RCC and distinguishing between these two forms on cross-sectional imaging remains a challenge. Therefore, RCC-specific biomarkers are urgently required for accurate and non-invasive, pre-surgical diagnosis of benign lesions. We have previously shown that dysregulation in glycolytic and tricarboxylic acid cycle intermediates can distinguish benign lesions from RCC in a stage-specific manner. In this study, preoperative fasting urine samples from patients with renal masses were assessed by ¹H nuclear magnetic resonance (NMR). Significant alterations in levels of tricarboxylic acid cycle intermediates, carnitines and its derivatives were detected in RCC relative to benign masses and in oncocytomas vs. chromophobe RCC. Orthogonal Partial Least Square Discriminant Analysis plots confirmed stage discrimination between benign vs. pT1 (R2 = 0.42, Q2 = 0.27) and benign vs. pT3 (R2 = 0.48, Q2 = 0.32) and showed separation for oncocytomas vs. chromophobe RCC (R2 = 0.81, Q2 = 0.57) and oncocytomas vs. clear cell RCC (R2 = 0.32, Q2 = 0.20). This study validates our previously described metabolic profile distinguishing benign tumors from RCC and presents a novel metabolic signature for oncocytomas which may be exploited for diagnosis before cross-sectional imaging.

    Description: Plant metabolites that have shown activity against bacteria and/or environmental fungi represent valuable leads for the identification and development of novel drugs against clinically important human pathogenic fungi. Plants from the genus Eremophila were highly valued in traditional Australian Aboriginal medicinal practices, and E. alternifolia was the most prized among them. As antibacterial activity of extracts from E. alternifolia has been documented, this study addresses the question whether there is also activity against infectious fungal human pathogens. Compounds from leaf-extracts were purified and identified by 1- and 2-D NMR. These were then tested by disk diffusion and broth microdilution assays against ten clinically and environmentally relevant yeast and mould species. The most potent activity was observed with the diterpene compound, 8,19-dihydroxyserrulat-14-ene against Cryptococcus gattii and Cryptococcus neoformans, with minimum inhibition concentrations (MIC) comparable to those of Amphotericin B. This compound also exhibited activity against six Candida species. Combined with previous studies showing an antibacterial effect, this finding could explain a broad antimicrobial effect from Eremophila extracts in their traditional medicinal usage. The discovery of potent antifungal compounds from Eremophila extracts is a promising development in the search for desperately needed antifungal compounds particularly for Cryptococcus infections.

    Description: Surface degradation of Ni-enriched layered cathode material Li[Ni0.6Mn0.2Co0.2]O2 (NMC622) is the main reason that leads to large capacity decay during long-term cycling. In the frame of this research, an amorphous SiO2 coating was applied onto the surface of the commercially available NMC622 powder by a wet coating process, through the condensation reaction of tetraethyl orthosilicate. The chemical composition of the coating layer was analyzed by inductively-coupled plasma. The morphology was studied by scanning electron microscopy and transmission electron microscopy. Electrochemical properties, including cyclic voltammetry, galvanostatic cycling, and rate capability measurements in a half-cell configuration, were tested to compare the electrochemical behavior of the non-coated and coated NMC622 materials. It is shown that the rate performance of the NMC622 materials is not affected by the coating layer. After 700 cycles in the range of 3.0–4.3 V at 2 C discharge, the cells with SiO2-coated NMC622 materials retained 80% of their initial capacity, which is higher than the uncoated ones (74%). Physicochemical characterizations, e.g., XRD and SEM, were performed post-mortem to reveal the stabilizing mechanism of the SiO2-coated NMC622 electrodes after long-term cycling. Based on these results, this is due to the shielding effect of the coating between the NMC622 particle surface and the liquid electrolyte, along with its scavenging effect on HF. SiO2 coating is therefore a facile surface modification method that results in potentially significant enhancement of the cyclic stability of Ni-rich NMC materials.

    Description: This paper deals with a general class of integrals, the particular cases of which are connected to outstanding problems in physics and astronomy. Nuclear reaction rate probability integrals in nuclear physics, Krätzel integrals in applied mathematical analysis, inverse Gaussian distributions, generalized type-1, type-2, and gamma families of distributions in statistical distribution theory, Tsallis statistics and Beck–Cohen superstatistics in statistical mechanics, and Mathai’s pathway model are all shown to be connected to the integral under consideration. Representations of the integral in terms of Fox’s H-function are pointed out.

    Description: Pseudorabies virus (PRV) is a member of the alphaherpesvirus subfamily of the herpesviruses and is the causative agent of Aujeszky’s disease in pigs, causing respiratory, neurological, and reproductive symptoms. Given the heavy economic losses associated with Aujeszky’s disease epidemics, great efforts were made to develop efficacious vaccines. One of the best modified live vaccines to this day is the attenuated Bartha K61 strain. The use of this vaccine in extensive vaccination programs worldwide has assisted considerably in the eradication of PRV from the domesticated pig population in numerous countries. The Bartha K61 strain was described in 1961 by Adorján Bartha in Budapest and was obtained by serial passaging in different cell cultures. Ever since, it has been intensively studied by several research groups, for example, to explore its efficacy as a vaccine strain, to molecularly and mechanistically explain its attenuation, and to use it as a retrograde neuronal tracer and as a vector vaccine. Given that the Bartha K61 vaccine strain celebrates its 60th birthday in 2021 with no sign of retirement, this review provides a short summary of the knowledge on its origin, characteristics, and use as a molecular tool and as a vaccine.
