    Notes: The formation of protein single crystals grown with the shape controlled by the geometry of the capillary used as a growth cell is presented. The shaped crystals show strong birefringence under crossed nicols and diffract as single crystals up to 1.74 Å.

    Notes: Mosaic spread values for crystals grown in microgravity were measured using synchrotron radiation. Full width at half maximum (FWHM) values for diffraction line profiles in the range 10–20′′ (arc seconds, 1′′ = 1°/3600) were observed. These values are similar to those measured for crystals grown on earth using the gel-acupuncture method. The crystals analysed are composed of from two to five domains producing peaks having widths from 5 to 15′′. The distribution of these domains is neither homogeneous (with domains of lower quality concentrated in the centre of the crystal) nor isotropic (producing peaks whose width changes depending on the observation direction). Methodological aspects are also discussed, with special consideration of the effects of mosaic spread on the data-collection procedures for high-resolution (low-intensity) reflections.

    Notes: In this work we explore the possibilities of the gel-acupuncture technique, proposed previously for the growth of protein single crystals [García-Ruiz, Moreno, Viedma & Coll (1993). Mater. Res. Bull. 28, 541–546]. The main advantage of the technique is that the crystals are obtained inside an X-ray capillary and, unlike classical microdiffusion techniques, it involves a very simple and accurate technical arrangement that permits the continuous monitoring of the crystals in their growth environment. In particular, we describe the growth of single crystals of lysozyme, concanavalin A and ribonuclease A. Different starting conditions have been used to grow single crystals of these proteins into different types of capillaries at several protein and precipitating-agent concentrations. It is demonstrated that the technique works for a wide range of precipitating agents commonly used in protein crystal growth, such as large polymers (PEG 4000 and PEG 6000), organic solvents (from methanol to butanol) and salts [NaCl, (NH4)2SO4]. The range of inner diameter of the capillaries for which the technique works correctly has been also studied. The growth process and possible crystal movement was followed by video microscopy. Lysozyme crystals up to 3.1 mm were obtained but the average maximum linear crystal sizes were 2.0 mm for lysozyme, 0.4 mm for concanavalin A and 1.2 mm for ribonuclease, respectively. The waiting times to reach such a size, measured from the set-up of the experiments, were 72 h, 10 d and 5 d, respectively. Gels of tetramethoxysilane, tetraethoxysilane, sodium silicate, agar, high-strength agar and gel-gro have been tested in relation to their mechanical properties and their chemical interaction with the reactants. Finally, we discuss briefly the advantages of the gel-acupuncture technique and plausible applications other than crystal growth.

    Notes: Intercalation effect on BSCCO (2212) system, although it increases the interlayer distance and the c-axis remarkably, produces only a small change in the transition temperature. Thus, amongst other things, intercalation provides an effective method to investigate the influence of the inter-block coupling. Electrons are transferred from the host Cu-O2 layers to the guest lmolecules I2, HgBr2, HgI2 leading to evolution of the Tc. For this we have made high resolution XANES study on the O K and Cu L3 edges to estimate the density of the doping holes. We attempt on basis of our and earlier results the evolution of Tc in these as also the much larger decrease produced in Tc for I2-intercalation for which the increase in basal spacing is the smallest of the three halides.

    Notes: This paper presents a modification of the gel acupuncture method to grow isolated crystals inside X-ray capillaries. Protein crystals are grown from 2–12 µl of gelled agarose–protein solution, cryoprotected and immobilized by the gel matrix. The same X-ray capillary that acts as a crystallization reactor is used to transport the crystals to the X-ray source and to collect data at both room temperature and 100 K, without any post-crystallization manipulation. To enhance the flash-cooling stage, two additional elements are proposed for inclusion in the cryosystems currently in use: a laser pointer to illuminate the crystal to be flash-cooled and a trap to divert the N2 flow and switch from room temperature to 100 K without misalignment of the crystal. With the proposed implementation, data can be collected at different temperatures from the same crystal in exactly the same orientation. This permits the study, at lattice level, of changes in unit-cell parameters, mosaic spread and crystal quality induced by cryogenic temperatures and annealing techniques.

    Notes: Pr doping has been extensively studied in cuprate superconductors to understand the mechanism of quenching of superconductivity in cuprate perovskites. Experience has revealed that it acts differently in different cuprate perovskites. We have made high resolution polarization XANES measurements on a Pr-doped single crystal within the ab-plane on the Cu K and Pr LIII absorption edges to ascertain the valence state of Pr cation and thereby try learning how it is quenching superconductivity in the present case. Our results show that besides the Pr quenching holes the possibility of its localizing some holes through hybridization with O 2p and Cu 3d cannot be ruled out.

    Description / Table of Contents: Zuverlässigkeit elektrochemischer Methoden bei der Untersuchung von Prothesen aus nichtrostendem Stahl mit AluminiumoxidüberzugIm vorliegenden Bericht wird die Korrosionsbeständigkeit von Prothesen aus nichtrostendem Stahl mit Aluminiumoxidüberzug erörtert; als Versuchsmaterial dienten Stahl AISI 316 L mit plasmagespritztem Aluminiumoxidüberzug (mit und ohne Nickel als Zwischenschicht).Die Nickelschicht soll die Haftung zwischen dem keramischen Material und dem Stahl gewährleisten. Als Prüfmedium diente die Hank'sche Lösung, da diese Ähnlichkeit mit organischen Flüssigkeiten aufweist. Zur Verbesserung der Aussagefähigkeit der Ergebnisse wird empfohlen, mehrere der nachstehenden elektrochemischen Methoden nebeneinander zu verwenden: Bestimmung der Zeitabhängigkeit des Korrosionspotentials; Polarisationswiderstandsmessung; coulostatische Methoden und Wechselstrommethoden. Bei der elektrochemischen Untersuchung von keramikbeschichtetem nichtrostendem Stahl im passiven Zustand ergeben sich einige spezifische Schwierigkeiten durch die Störung der Arbeitselektrode infolge der einwirkenden elektrischen Signale, wodurch die Genauigkeit der Korrosionsgeschwindigkeitsmessung beeinträchtigt wird. Diese Schwierigkeiten treten nicht auf, wenn eine Kombination mit einer aktiveren Zwischenschicht benutzt wird.
    Notes: The corrosion resistance of alumina-covered stainless steel prosthesis is discussed in this report. The test material was an AISI 316L steel with a plasma sprayed on alumina coating, applied directly or through an intermediate nickel base layer, whose mission is to guarantee a perfect adherence between the ceramic material and the steel. Use was made in the laboratory of Hank's solution as test medium since this solution is similar to organic fluids. In order to increase the reliability of the results the simultaneous use of various of the following electrochemical methods are recommended: Determination of corrosion potential with the time, polarization resistance method, coulostatic method and a.c. methods. The use of electrochemical methods to study ceramic-covered stainless steel in the passive state poses some specific difficulties accruing from the disturbance of the working electrode by the applied electrical signals which limit the accuracy of the corrosion rate measurements. These difficulties do not appear in the case of a material having a intermediate layer which is more active.
