Notes: Abstract. Isochorismate hydroxymutase (i.e. isochorismate synthase, EC 5.4.99.6) was purified from an anthraquinone-producing cell-suspension culture of Galium mollugo L. Although attempts to stabilize the labile enzyme met with little success, a substantial increase in enzyme activity was observed in the presence of glycine betaine (500 mM). Column chromatography on solid supports other than diethylaminoethyl (DEAE)-Sephacel, Phenylsepharose Cl-4B or Cibacron Blue 3G-A did not give active enzyme preparations. In spite of these drawbacks the enzyme was purified 573-fold. Enzyme activity depended strictly on the presence of Mg2+. Kinetic data for chorismate in the forward reaction (K m = 807 μM, V max = 6.2 pkat · mg−1) and for isochorismate in the reverse reaction (K m = 675 μM, V max = 5.9 pkat · mg−1) were determined.

Notes: Summary Inherent relative growth rate has been suggested as a major determinant of plant species' capacity to regrow and compensate for tissues lost to herbivores. We investigated: 1) the relationship between compensatory growth capacity and relative growth rate (RGR) in six semiarid-land shrubs following removal in winter or spring of 90% of the previous year's growth, 2) the influence of loss of buds on production of new growth and 3) the relationship between meristematic potential and the capacity to produce new growth in four of the six semiarid-land shrub species. Four-year-old plants growing under field conditions were used in the study. The species with the highest inherent growth rate, sagebrush [Artemisia tridentata ssp. vaseyana (Rydb.) Beetle], died following the severe clipping treatments. The other five species exactly compensated for lost tissues. Inherent growth rates and compensatory growth capacity of the shrubs were not correlated. Loss of 90% of the buds on the previous year's growth did not limit production of new growth. Instead, shrubs that lost buds produced more new growth biomass than the controls. The shrub species had significantly different meristematic potential. Curlleaf mountain mahogany (Cercocarpus ledifolius Nutt.) and serviceberry (Amelanchier alnifolia Nutt.) had the greatest and least number of buds and long shoots per plant, respectively. The number of long shoots produced following bud removal was positively correlated with new growth biomass, while the percentage of long shoots produced at the basal position on twigs was negatively correlated with new growth biomass. Our results suggest that inherent growth rate is not likely to influence production of new growth following browsing when resources for growth are not limiting. In contrast, the ability of a shrub to initiate long shoot growth is likely to influence production of new growth even when resources for growth are abundant.

Notes: Abstract. Microtomography (micro-computed-tomography, μ-CT) is a method to image and quantify trabecular bone. It has the capability to address the role of trabecular architecture on the mechanical properties of bone and to study trabecular bone remodeling. The system described in this work is based on a compact fan-beam type tomograph that can work in spiral scanning or multislice mode. An X-ray tube with a microfocus is used as a source, a CCD-array as a detector. Samples with diameters from a few millimeters to a maximum of 14 mm can be measured, typically, bone biopsies with a diameter of 8 mm and a length of approximately 10 mm are measured. Spatial resolution is 28 μm. Usually the volume of interest contains 4 × 4 × 4 mm3 and is represented in 14 × 14 × 14 μm3 voxels. 3D stereological indices are extracted according to the standard definitions used in histomorphometry. Triangular surface representation is effected with an extended marching cube algorithm and forms a convenient basis for finite element analysis. Microtomographic measurements may be employed to ``calibrate'' lower-dose, lower-resolution images in vivo as well as to nondestructively assess unprocessed surgical bone biopsy specimens. These specimens remain intact for mechanical or histological testing.

Notes: Summary For the reaction Bi209 (n, γ) Bi210, theQ-value as determined from a suitablereaaction chain exceeds that indicated by the γ-line of maximum energy as measured with a magnetic pair spectrometer. Coincidence investigations have therefore been undertaken to search for γ-γ-cascades: transitions for 117, 165, 275, 396 and 507 keV have been detected in coincidence with the lines at 4 MeV. The intensities of the former lines have been elucidated and are discussed. The most probable value for the neutron binding energy is thereby indicated to be 4·56 MeV.

Keywords: Chemical ozone loss (Arctic vortex 1995–96; halogen chemistry)
Source: Springer Online Journal Archives 1860-2000
    Topics: Geosciences , Physics
Notes: Abstract Severe chemical ozone loss has been detected in the Arctic in the winter and spring of 1995–96 by a variety of methods. Extreme reductions in column ozone due to halogen catalysed chemistry were derived from measurements of the Halogen Occultation Experiment (HALOE) on board the Upper Atmosphere Research Satellite in the Arctic vortex. Here, we discuss further aspects of the HALOE observations in the Arctic over this period. Potential problems, both in the data themselves and in the methodology of the data analysis are considered and the reason for the differences between the Arctic ozone losses deduced from HALOE data version 17 and 18 is analysed. Moreover, it is shown that HALOE measurements in the Arctic in winter and spring 1995–96 compare well with observations by other ground-based and satellite instruments.

Notes: Abstract  The growth yield coefficients of phenol as well as of chloro- and methyl-substituted derivatives and the respective phenoxyalkanoic acids have been considered in a theoretical study. The yield coefficient of phenol assimilated via the ortho pathway is at a given P/O quotient 9–23% higher than that of the meta route. Chlorine, removed by a reductive mechanism, decreases the yield figure by 10% per chlorine atom; hydrolytic mechanisms are more favourable in this respect. Methylation has a positive effect on the energetics of phenol utilization. Phenoxyacetic acid derivatives have a reduced yield per carbon atom in comparison to the respective phenol derivative. This is compensated for by higher chain lengths of the alkanoic acid moiety. The yield coefficients calculated for phenol correspond to a P/O quotient of 2–3 as compared to the experimental carbon conversion. With chlorinated derivatives, e.g. 2,4-dichlorophenoxyacetic acid or pentachlorophenol, the apparent gain in energy reflects a P/O〈1, indicating uncoupling effects during growth on such compounds.

Notes: Abstract When transformed with a recombinant vector carrying the ubiC gene (encoding chorismate pyruvate-lyase, EC 4.1.3.27) the triple mutant (Phe−, Trp−, Tyr−) Klebsiella pneumoniae 62-1 excretes 4-hydroxybenzoic acid instead of chorismic acid. The recombinant strain can be used to produce in high yield specifically ring-labelled 4-hydroxybenzoic acid from isotopically labelled glucose.
