    Notes: We previously identified HpuB, an 85 kDa Fe-repressible protein required for utilization of Fe from, and binding to, haemoglobin and the haemoglobin–haptoglobin complex. The gene for hpuB was cloned from Neisseria meningitidis strain DNM2 and the predicted amino acid sequence indicates that HpuB is an outer membrane receptor belonging to the TonB family of high-affinity transport proteins. A second open reading frame, predicted to encode a 34.8 kDa lipoprotein, was discovered 5′ to hpuB, and was designated hpuA. HpuA was identified in a total-membrane-protein preparation by construction of a mutant lacking HpuA. Acylation of HpuA was confirmed by [3H]-palmitic acid labelling of meningococci. Consensus promoter sequences were not apparent 5′ to hpuB. The hpuA insertion mutation exerted a polar effect, abolishing expression of hpuB, suggesting that hpuA and hpuB are co-transcribed. The 3.5 kb polycistronic hpuAB mRNA was identified and shown to be transcriptionally repressed by iron. The transcriptional start site was identified 33 nucleotides 5′ to the hpuA translational start site, appropriately positioned around consensus promoter and ferric uptake regulator (Fur)-box sequences. The structure of this operon suggests that HpuA–HpuB is a two-component receptor analogous to the bipartite transferrin receptor TbpB–TbpA.

    Notes: Models of mass and energy exchanges between the biosphere and the atmosphere generally contain a nonlinear dependence between fluxes and model parameters, and thus estimation of these parameters from measurements in a heterogeneous landscape depends on the scale of the observations. The scale-dependence of a typical surface-exchange model (the CSIRO Biospheric Model, CBM) is examined using the diurnal variation of hourly fluxes of CO2, latent heat, sensible heat and soil heat. The fluxes were measured using micrometeorological techniques over six sites in a grazing/pasture system in SE Australia during a period of three weeks in 1995. Nonlinear parameter inversion was used to determine model parameters.Analysis of the covariance of the estimates of the parameters and the unexplained residuals of the model showed that a maximum of three or four parameters could be determined independently from the observations for all six sites. Estimates of a key model parameter, jmax, the mean of maximum potential electron transport rate of all leaves within the canopy, was best determined by the measurements of net CO2 flux at all sites examined. Measurements of ground heat flux provide little information about any of the model parameters in CBM.Because of nonlinearities in the surface exchange model, calculated fluxes will be in error if parameters for the component vegetation types are simply averaged in proportion to their areal fraction. The magnitude of these errors was examined for CBM using a hypothetical land surface consisting of two surface types, each with different parameter values. Predictions of net CO2, latent heat and ground heat fluxes using a linear combination of model parameters for the two surface types were quite similar with those found using optimal estimates of the parameters for the landscape, but were significantly poorer for sensible heat fluxes.

    Notes: Rhizobium melioti DctD activates transcription from the dctA promoter by catalysing the isomerization of closed complexes between σ54-RNA polymerase holoenzyme and the promoter to open complexes. DctD must make productive contact with σ54-holoenzyme and hydrolyse ATP to catalyse this isomerization. To define further the activation process, we sought to isolate mutants of DctD that had reduced affinities for σ54-holoenzyme. Mutagenesis was confined to the well-conserved C3 region of the protein, which is required for coupling ATP hydrolysis to open complex formation in σ54-dependent activators. Mutant forms of DctD that failed to activate transcription and had substitutions in the C-terminal half of the C3 region were efficiently cross-linked to σ54 and the β-subunit of RNA polymerase, suggesting that they bound normally to σ54-holoenzyme. In contrast, some mutant forms of DctD with amino acid substitutions in the N-terminal half of the C3 region had reduced affinities for σ54 and the β-subunit in the cross-linking assay. These data suggest that the N-terminal half of the C3 region of DctD contains a site that may contact σ54-holoenzyme during open complex formation.

    Description: In order to reduce the foundation settlement, conserve resources, and be environmental-friendly while increasing the use of soil resources, an artificial crust layer formed by in situ stabilization is proposed to combine with prestressed pipe piles over soft ground in road construction. In this study, a centrifuge test and two-dimensional coupled-consolidation finite-element analyses are conducted to simulate the construction of an embankment. And a two-dimensional parametric study is conducted to study the performance indicated by maximum long-term settlement, excess pore water, and tensile stress under various conditions. The results of the centrifuge test clearly show that the measured settlement, excess pore water, and tensile stress are in good agreement with the calculated results. In addition, the key factors of pile spacing and thickness and strength of the crust have an influence on the maximum settlement, stress of the foundation, and tensile stress of the crust using the two-dimensional coupled-consolidation finite-element analyses. And the stress transfer regular of the foundation is analyzed under various conditions. Moreover, the failure of the crust contained tensile cracking and shearing failure and the thickness of the pile that pierced the crust are also affected by the key factors.

    Description: With the development of market competition, company faces more and more pressures. Meanwhile, procurement has a vital effect on achieving competitive advantages in a supply chain. Selecting the appropriate suppliers is one of the most important sections in purchase management. However, in real situation, supplier selection is a multiple objective problem about different items with vagueness and randomness of the data. It is very complex. Hence, research about supplier selection is relatively scarce under considering multiple items, discount price, and fuzzy and stochastic information. In our paper, we develop a fuzzy multiobjective supplier selection model for overcoming uncertainty and multiple items. Stochastic demand, fuzzy objectives, and weights are simultaneously applied to help the managers to select the suitable suppliers about different items. For illustration purpose, a numerical example is presented to verify the effectiveness of the proposed model.

    Description: Histamine H2 receptor (HRH2) was previously suggested to affect the proliferation of breast cancer cells and disease-free survival of breast cancer patients. Furthermore, a common polymorphism, rs2067474, was identified in an enhancer element of the HRH2 gene promoter and was reported to be associated with various diseases including cancer. However, the relationship between this polymorphism and breast cancer risk and malignant degree remains unclear. The aim of this study was to clarify the clinical association of rs2067474 polymorphism with breast cancer. A total of 201 unrelated Chinese Han breast cancer patients and 238 ethnicity-matched health controls were recruited and rs2067474 polymorphism was genotyped. Logistic regression analyses were performed to calculate the odds ratios (ORs) as a measure of association of genotype with breast cancer according to 3 genetic models (dominant, recessive, and additive). Although the percentage of hormone receptor negative cases tended to be higher in AA genotypes, we did not find any significant associations of rs2067474 polymorphism with breast cancer risk or with related clinicopathological parameters in the present study, which indicates that rs2067474 polymorphism of HRH2 gene might not be a risk factor in the development of breast cancer in Chinese Han population.

    Description: Objective. To investigate the impact of seven-valent pneumococcal conjugate vaccine on tube insertions in a partial immunized pediatric population. Study Design. Retrospective ecological study. Methods. This study used Taiwan National Health Insurance Research Database for the period 2000–2009. Every child under 17 years old who received tubes during this 10-year period was identified and analyzed. The tube insertion rates in different age groups and the risk to receive tubes in different birth cohorts before and after the release of the vaccine in 2005 were compared. Results. The tube insertion rates for children under 17 years of age ranged from 21.6 to 31.9 for 100,000 persons/year. The tube insertion rate of children under 2 years old decreased significantly after 2005 in period effect analysis (β = −0.074, P 〈 0.05, and the negative β value means a downward trend) and increased in children 2 to 9 years old throughout the study period (positive β values which mean upward trends, P 〈 0.05). The rate of tube insertion was lower in 2004-2005 and 2006-2007 birth cohorts than that of 2002-2003 birth cohort (RR = 0.90 and 0.21, 95% CI 0.83–0.97 and 0.19–0.23, resp.). Conclusion. The seven-valent pneumococcal conjugate vaccine may reduce the risk of tube insertion for children of later birth cohorts. The vaccine may have the protective effect on tube insertions in a partial immunized pediatric population.

    Description: In order to predict productivity of multiple-fractured horizontal well in fractured reservoir, flow models of reservoir and hydraulic fractures based on the volumetric source idealization are developed. The models are solved by utilizing Laplace transformation and orthogonal transformation, and flow rate of the well is calculated by coupling the two models. Compared to traditional point source functions, volumetric source function has many advantages in properties of function and programming calculation. The productivity predicting model is verified via an analytical ternary-porosity model. Moreover, a practical example of fractured horizontal well is studied to analyze the productivity and its influent factors. The result shows that flow rate of each fracture is different and inner fracture contributes least to productivity. Meanwhile, there are optimizing ranges for number, length, and conductivity of hydraulic fractures. In low-permeability reservoir, increasing surface area in contact with reservoir by increasing number and length of hydraulic fractures is the most effective method to improve the productivity.

    Description: Lane-changing and overtaking are conventional maneuvers on roads, and the reference trajectory is one of the prerequisites to execute these maneuvers. This study proposes a universal trajectory planning method for automated lane-changing and overtaking maneuvers, in which the trajectory is regarded as the combination of a path and its traffic state profiles. The two-dimensional path is represented by a suitable curve to connect the initial position with final position of the ego vehicle. Based on the planned path, its traffic state profiles are generated by solving a nonlinear mathematical optimization model. Moreover, the study discretizes the time horizon into several time intervals and determines the parameters to obtain the continuous and smooth profiles, which guarantees the safety and comfort of the ego vehicle. Finally, a series of simulation experiments are performed in the MATLAB platform and the results show the feasibility and effectiveness of the proposed universal trajectory planning method.

    Description: Growth in environmental sustainability has prompted the logistics industry to seek sustainable development, and carbon tax policies are considered an effective approach to reducing carbon emissions. This study investigates the optimization of sustainable transportation and inventory under a carbon tax policy and explores effective methods for coordinating the interests of governments and enterprises. The results can provide insights into sustainable logistics for decision-making by enterprises and policy-making by governments. We first examine a Stackelberg game model and design an iterative solution to optimize sustainable transportation and inventory under the carbon tax policy. We then establish a three-stage dynamic game model to optimize the wholesale price, carbon tax rate, and proportion of sustainable investment shared by the government. Furthermore, we perform a simulation to identify the optimal solution of the three-stage game, and we compare the simulation results with a numerical example. The results indicate that a carbon tax policy can improve social welfare and the sustainability of transportation and inventory but could hinder corporate profits. An appropriate sustainable investment-sharing strategy could compensate for the shortcomings of the carbon tax policy and result in positive outcomes for governments and enterprises.
