Mechanisms of manganese(II) oxidation by filamentous ascomycete fungi vary with species and time as a function of secretome composition. Frontiers in Microbiology, 12, (2021): 610497, https://doi.org/10.3389/fmicb.2021.610497.
Description: Manganese (Mn) oxides are among the strongest oxidants and sorbents in the environment, and Mn(II) oxidation to Mn(III/IV) (hydr)oxides includes both abiotic and microbially-mediated processes. While white-rot Basidiomycete fungi oxidize Mn(II) using laccases and manganese peroxidases in association with lignocellulose degradation, the mechanisms by which filamentous Ascomycete fungi oxidize Mn(II) and a physiological role for Mn(II) oxidation in these organisms remain poorly understood. Here we use a combination of chemical and in-gel assays and bulk mass spectrometry to demonstrate secretome-based Mn(II) oxidation in three phylogenetically diverse Ascomycetes that is mechanistically distinct from hyphal-associated Mn(II) oxidation on solid substrates. We show that Mn(II) oxidative capacity of these fungi is dictated by species-specific secreted enzymes and varies with secretome age, and we reveal the presence of both Cu-based and FAD-based Mn(II) oxidation mechanisms in all 3 species, demonstrating mechanistic redundancy. Specifically, we identify candidate Mn(II)-oxidizing enzymes as tyrosinase and glyoxal oxidase in Stagonospora sp. SRC1lsM3a, bilirubin oxidase in Stagonospora sp. and Paraconiothyrium sporulosum AP3s5-JAC2a, and GMC oxidoreductase in all 3 species, including Pyrenochaeta sp. DS3sAY3a. The diversity of the candidate Mn(II)-oxidizing enzymes identified in this study suggests that the ability of fungal secretomes to oxidize Mn(II) may be more widespread than previously thought.

Description / Table of Contents: Summary The population of the White Stork in Baden-Württemberg (South-west Germany) was approximately constant in the post-war years until 1959. Beginning with 1960 it started to decline with an average rate of 13% per year. After 1960 the average annual mortality rate increased both in adults and in first year birds. In addition the average age of first breeding and the average percentage of pairs without breeding success increased in the second period. The latter means a decrease of breeding success. Some possible causes for the changes of these parameters are mentioned.

Notes: Abstract Coumarin dyes are encapsulated in silicate and aluminosilicate polymeric glasses synthesized by the sol-gel technique. The coumarin dyes retain their luminescent properties in all of the aged gels and in many of the dried gels (xerogels). The luminescence spectra of the new optical materials are reported. The luminescence of coumarin 4 provides a probe at the molecular level of the changes which occur during the gelation and drying. The spectral changes are studied and discussed in detail. Gels and xerogels doped with coumarin 460, 480, and 540A exhibit optical gain and laser action. The laser properties of these new solid-state dye laser materials are reported and discussed.

Notes: Abstract Quantitative structure-activity relationships for piritrexim and analogues acting as inhibitors of tumour cell growth have been derived. First the Free-Wilson-method was used on a homologous training set of eight derivatives. The selection of variables important for the biological activity of the compounds was carried out with different multivariate methods as multiple linear regression, the partial least squares method und a genetic algorithm. The derivation of three-dimensional structure activity relationships started with a systematic conformational analysis of all compounds. For the conformations having minimal energy and being in agreement with the crystal structure of piritrexim charges were calculated with the AM1 hamiltonian. For the superimposition of the derivatives two methods were used: maximal similarity of the common substructure or of the molecular fields. A Comparative Molecular Field Analysis with steric and electrostastic fields identified regions important for the activity of the studied compounds independent of the chosen alignment and also correctly predicted the activity of two nonhomologous compounds.
