    Notes: 〈list style="custom"〉1The population density, diversity and productivity of the microbial plankton in an oligotrophic maritime Antarctic lake were studied for a 15-month period between December 1994 and February 1996.2In the lake, concentrations of nutrients and dissolved organic carbon were uniformly low, temperature varied over a small annual range of 0.1–3 °C, and the surface was ice-covered except during a period of approximately 6 weeks in summer.3The total of 57 morphotypes of protozoa observed during the study is a higher taxonomic diversity than previously reported from continental Antarctic lakes, but lower than that found in more eutrophic maritime Antarctic lakes. Likewise, planktonic abundance and productivity were lower than has been reported in other lakes on Signy Island, but generally higher than those of lakes on the Antarctic continent.4There were marked seasonal and interannual variations in planktonic population density.5Chlorophyll a concentrations ranged from undetectable to 4.2 µg L-1 and the greatest rate of primary productivity measured was 4.5 mg C m-3 h-1. The phytoplankton was dominated by small chlorophytes and chrysophytes, with phototrophic nanoflagellate abundance ranging from 1.1 × 103 to 1.2 × 107 L-1.6Bacterial densities of 3.6 × 108 to 1.9 × 1010 L-1 were recorded and bacterial productivity reached a peak of 0.36 µg C L-1 h-1. Numbers of heterotrophic nanoflagellates between 5.0 × 104 and 1.8 × 107 L-1, and of ciliates from undetectable to 1.1 × 104 L-1 were observed. Naked amoebae were usually rare, but occasionally reached peaks of up to 1.5 × 103 L-1.

    Notes: Streptococcus agalactiae is a major bacterial pathogen that is the cause of serious economic losses in many species of freshwater, marine and estuarine fish worldwide. A highly efficacious S. agalactiae vaccine was developed using extracellular products (ECP) and formalin-killed whole cells of S. agalactiae. The vaccine efficacy following storage of S. agalactiae ECP and formalin-killed S. agalactiae cells at 4 °C for 1 year was determined. The stored ECP containing S. agalactiae formalin-killed cells failed to prevent morbidity and mortality among the vaccinated fish, and the relative percentage survival was 29. Serum antibody responses of the stored ECP and freshly prepared ECP against soluble whole cell extract of S. agalactiae indicated that significantly less antibody was produced in fish immunized with stored ECP and S. agalactiae cells than in those fish immunized with freshly prepared ECP and S. agalactiae cells at day 31 post-vaccination. Silver staining of sodium dodecyl sulphate-polyacrylamide gels and immunostaining of Western blots with tilapia antiserum to S. agalactiae revealed that predominant 54 and 55 kDa bands were present in the freshly prepared ECP fraction. The 55 kDa band was absent from the stored ECP and new bands below 54 kDa appeared on the Western blot. The results of this study on S. agalactiae ECP provide evidence for a correlation between protection and antibody production to ECP and for the importance of the 55 kDa ECP antigen for vaccine efficacy.

    Notes: Streptococcus agalactiae was isolated from cultured gilthead seabream, Sparus auratus L., and diseased wild Klunzinger's mullet, Liza klunzingeri (Day), in Kuwait Bay, Arabian Gulf. Isolates were catalase negative, β-haemolytic, Gram-positive cocci and serogroup B. Experimental infectivity trials with mullet and seabream brain isolates in Nile tilapia, Oreochromis niloticus L., caused 100 and 90% mortality, respectively, within 7 days post-inoculation indicating virulent S. agalactiae as the bacterial pathogen responsible for the epizootic in Kuwait Bay.

    Notes: Flavobacterium columnare is an important pathogen of freshwater fish, implicated in skin and gill disease, often causing high mortality. An outbreak of skin disease in fingerling and adult Nile tilapia, Oreochromis niloticus (L.), cultivated in a recirculation system, was investigated. Four strains were isolated and characterized by biochemical reactions, enzyme production, fatty acid profile and analysis of the 16S-23S rDNA intergenic spacer region. All strains were identified as F. columnare. Experimental infection assays with one of these strains (BZ-5-02) were conducted and pathogenicity (by intramuscular route) was demonstrated in Nile tilapia and channel catfish, Ictalurus punctatus (Rafinesque). This is the first report of characterization of Brazilian strains of F. columnare.

    Notes: The biological properties of Edwardsiella ictaluri RE-33 rifampicin-mutant and its parent strain EILO were analysed. RE-33 is an avirulent isolate used as a modified live vaccine against enteric septicaemia of catfish. Electrophoretic analysis of lipopolysaccharide (LPS) patterns showed high homology between both isolates. Further characterization of LPS by immunoblotting revealed the main differences in LPS composition. The RE-33 isolate lacks the high molecular weight bands of LPS (HMW-LPS). Outer membrane protein analysis also showed some immunological differences between RE-33 and the EILO parent strain. Only two fingerprinting techniques, fatty acid composition analysis and Biolog phenotypic profiles, were able to discriminate between both isolates.

    Notes: The profile of photosynthetic rate with depth through a leaf depends on the profiles of light absorption and photosynthetic capacity. Using a combination of several techniques, a comprehensive description of spinach leaves has been obtained. Profiles of CO2 fixation were obtained by exposing leaves to 14CO2 for 10 s under blue or green light before freeze clamping and paradermal sectioning. Profiles of light absorption were measured on adjacent parts of the leaf by quantifying chlorophyll fluorescence images of the transversely cut face obtained when blue or green light was applied to the adaxial or abaxial surface. The profile of CO2 fixation was modelled using the measured profiles of light absorption and photosynthetic capacity. There was remarkably good agreement between the observed and modelled CO2 fixation profiles for the eight combinations of colour, orientation and irradiance tested. Gas exchange of an intact leaf was also measured concurrently with conventional chlorophyll fluorescence under blue or green light. These data were consistent with the multi-layer leaf model with the exception of blue light applied to the abaxial surface, where chlorophyll fluorescence appeared to come from layers deeper than expected. Photosynthetic capacity matched the profile of green but not white light absorption through the leaf.

    Notes: Chlorophyll fluorescence was used to estimate profiles of absorbed light within chlorophyll solutions and leaves. For chlorophyll solutions, the intensity of the emitted fluorescence declined in a log–linear manner with the distance from  the  irradiated  surface  as  predicted  by  Beer's  law. The amount of fluorescence was proportional to chlorophyll  concentration  for  chlorophyll  solutions  given  epi-illumination on a microscope slide. These relationships appeared to hold for more optically complex spinach leaves. The profile of chlorophyll fluorescence emitted by leaf cross sections given epi-illumination corresponded to chlorophyll content measured in extracts of leaf paradermal sections. Thus epifluorescence was used to estimate relative chlorophyll content through leaf tissues. Fluorescence profiles across leaves depended on wavelength and orientation, reaching a peak at 50–70 µm depth. By infiltrating leaves with water, the pathlengthening due to scattering at the airspace : cell wall interfaces was calculated. Surprisingly, the palisade and spongy mesophyll had similar values for pathlengthening with the value being greatest for green light (550 〉 650 〉 450 nm). By combining fluorescence profiles with chlorophyll distribution across the leaf, the profile of the apparent extinction coefficient was calculated. The light profiles within spinach leaves could be well approximated by an apparent extinction coefficient and the Beer–Lambert/Bouguer laws. Light was absorbed at greater depths than predicted from fibre optic measurements, with 50% of blue and green light reaching 125 and 240 µm deep, respectively.

    Notes: Changes in specific leaf area (SLA, projected leaf area per unit leaf dry mass) and nitrogen partitioning between proteins within leaves occur during the acclimation of plants to their growth irradiance. In this paper, the relative importance of both of these changes in maximizing carbon gain is quantified. Photosynthesis, SLA and nitrogen partitioning within leaves was determined from 10 dicotyledonous C3 species grown in photon irradiances of 200 and 1000 µmol m−2 s−1. Photosynthetic rate per unit leaf area measured under the growth irradiance was, on average, three times higher for high-light-grown plants than for those grown under low light, and two times higher when measured near light saturation. However, light-saturated photosynthetic rate per unit leaf dry mass was unaltered by growth irradiance because low-light plants had double the SLA. Nitrogen concentrations per unit leaf mass were constant between the two light treatments, but plants grown in low light partitioned a larger fraction of leaf nitrogen into light harvesting. Leaf absorptance was curvilinearly related to chlorophyll content and independent of SLA. Daily photosynthesis per unit leaf dry mass under low-light conditions was much more responsive to changes in SLA than to nitrogen partitioning. Under high light, sensitivity to nitrogen partitioning increased, but changes in SLA were still more important.

    Notes: A modification of the ‘cold plaque’ screening technique (Hodge et al., Plant Journal1992, 2, 257–260) was used to screen a cDNA library constructed from drought-stressed leaf tissue of the desiccation tolerant (‘resurrection’) grass Sporobolus stapfianus. This technique allowed a large number of clones representing genes expressed at low abundance to be isolated. An examination of expression profiles revealed that several of these genes are induced in desiccation-tolerant tissue experiencing severe drought stress. Further characterization indicated that the gene products encoded include an eIF1 protein translation initiation factor and a glycine- and proline-rich protein which have not previously been associated with drought stress. In addition, genes encoding a serine/threonine phosphatase type 2C, a tonoplast-intrinsic protein (TIP) and an early light-inducible protein (ELIP) were isolated. A number of these genes are expressed differentially in desiccation-tolerant and desiccation-sensitive tissues, suggesting that they may be associated with the desiccation tolerance response of S. stapfianus. The results indicate that there may be unique gene regulation processes occurring during induction of desiccation tolerance in resurrection plants which allow different drought-responsive genes to be selectively expressed at successive levels of water loss.
