    Notes: We have identified proteins that control the expression of slpA, the gene encoding the crystalline surface layer of Thermus thermophilus HB8. We cloned three genes from T. thermophilus that specifically repressed the expression of the slpA promoter in Escherichia coli. The proteins encoded by two of them (Rep6 and Rep29) bound in vitro to the slpA promoter, while that from the third (Rep54) bound specifically to the 5′-untranslated region (5′UTR) of the slpA mRNA. Rep6 protein was identified as a C-fragment from a Thermus cytoplasmic basic protein of 28 kDa, whose coding gene, slrA (for S-layer regulator), was characterized. Surprisingly, Rep29 was identified as a C-fragment of SlpM, an S-layer-like protein that is overexpressed in slpA mutants. Insertional inactivation of slrA and slpM demonstrated their in vivo function in the control of slpA transcription: SlrA acts as a repressor, and SlpM as an activator. Even more surprising was the identification of Rep54, the 5′UTR mRNA-binding protein, as a C-terminal fragment of the SlpA protein. This result, in addition to further in vivo evidence presented here, supports the existence of a translational autoregulation in slpA expression. The physiological meaning of overlapping transcriptional and translational controls of S-layer expression, and its relationships with other systems, are discussed.

    Notes: With the aim of improving artificial fertilization (AF) in turbot, Scophthalmus maximus (L.), a series of fertilization experiments was carried out under dry conditions and different wet conditions (eggs/sea water: 2V/V and V/V). Another series of fertilization experiments was carried out with different quantities of sperm pool to determine the optimal ratio of spermatozoa to eggs for each AF method. Sperm pool from two males and eggs from spawns with a viability rate of 〉 70% were used. The sperm pool’s density (0.4–5.18 × 109 sperm mL–1) and motility (1–5) had been assessed previously. Significantly different fertilization rates were found when comparing 2V/V and V/V wet conditions. Significantly higher fertilization rates were found in dry fertilization when the sperm–egg ratio was 〉 9000 spermatozoa per egg and, under wet condition V/V, at 3000–4000 spermatozoa per egg.

    Notes: The development of artificial feeds for marine fish larvae is demanding and, taking into account the difficulties and costs associated with the rearing of the larvae of some fish, preliminary evaluation of feed ingredients using in vitro techniques may be an alternative to in vivo assays. Some proteins used in microfeeds for marine fish were tested in vitro for examination of their effect on the proteases of seabream larvae. Casein and cuttlefish meal did not affect protease activity of crude larval extracts, whereas ovalbumin produced a 60% inhibition. The use of a pH-stat for the assessment of the degree of hydrolysis (DH) of proteins confirmed the low nutritive value of ovalbumin, since a much lower value was obtained for microcapsules prepared using this protein source when compared with those prepared using casein or cuttlefish meal (3.2 vs. 7.3 and 7.6, respectively). Products resulting from such hydrolysis were analysed using sodium dodecyl sulphate–polyacrylamide gel electrophoresis (SDS–PAGE). Image analysis of such gels led to the formulation of an index named the coefficient of protein degradation (CPD) which expressed the extent to which the main protein fractions were hydrolysed by larval proteases in a given time. A significant correlation (r2 = 0.98, P 〈 0.05) was obtained between DH and CPD values measured for either protein sources or microcapsules. The combination of protease inhibition assays with measurements of DH and CPD is proposed as a preliminary evaluation of protein ingredients used in the formulation of artificial feeds for larval fish.

    Notes: The blue-light requirement for the biosynthesis of nitrite reductase and an NO2– transport system was studied in Chlamydomonas reinhardtii mutant S10. The only oxidized nitrogen species that could be taken up by this mutant was NO2–, due to the presence of NO2– transport systems and the absence of high-affinity NO3– transporters. NH4+-grown cells required illumination with blue light to recover the ability to take up NO2– when resuspended in an NO2–-containing NH4+-deprived medium. This blue-light- dependent recovery, which took 1 h, could be suppressed by cycloheximide, indicating that protein biosynthesis was involved. The biosynthesis of nitrite reductase took place in cell suspensions irradiated with red light, even in the absence of NO2–, thus suggesting that the process requiring blue light was the biosynthesis of an NO2– transport system. Nitrite reductase-containing cells (pre-irradiated with red light) took 1 h to start consuming NO2– when they were additionally irradiated with blue light in the presence of this anion, and this process was also cycloheximide-sensitive. The NO2– transport system operated either under red plus blue light or red light only. Thus, in C. reinhardtii mutant S10 cells, blue light was only required for the biosynthesis of an NO2– transport system and not for its activity.

    Notes: A Lower Cretaceous carbonate platform depositional system with a rimmed margin and an adjacent oversteepened slope was analysed in order to determine its depositional architecture and major depositional controls. The platform is made up of coral, rudist, orbitolinid and algal limestones and, in a 12-km dip transect, there is a gradation from lagoon to platform margin, slope and basin environments, each characterized by distinctive sedimentological features and facies associations. The rimmed platform is an aggradational system developed during approximately 4·2 million years of fluctuating relative sea-level rise, and it is bounded by unconformities at its base and top. Internal cyclicity in the construction of the system is evident, mainly in platform interior and slope settings. The seven recognized sequences average 0·6 million years in duration and are related to minor relative sea-level changes. Carbonate deposition occurred in shallow- and deep-water settings during periods of high relative sea level. Reduced rates of sea-level rise led to the development of shallowing upward sequences and, eventually, to the exposure of the shallowest parts of the platform during relative sea-level falls. During low relative sea level, erosion surfaces developed on the slope, and gravitational resedimentation occurred at the toe of slope. Basinwards, resedimented units pinch out over distances of a few hundred metres. Active faults controlled sedimentation at the platform margin, promoting the development of steep slopes (up to 35°) and preventing progradation of the shallow-water platform, despite high sediment production rates. The development of sequences is interpreted to be related to tectonic activity.

    Notes: The western Mediterranean is composed of irregular troughs formed as back-arc basins in the hanging wall of the W-directed Apenninic subduction which retreated eastward during Neogene and Quaternary times. The basins are progressively younger toward the east, ageing from late Oligocene–early Miocene (Valencia, Provençal, Alboran and Algerian basins), to middle Miocene–Pleistocene (Tyrrhenian Sea). The basins isolated boudins of continental lithosphere, the Sardinia–Corsica block representing the largest. The boudinage has a wavelength of 100–400 km and facilitated stretching of the continental lithosphere with formation of new oceanic crust in the Provençal, Algerian, Vavilov and Marsili basins. The boudins developed both in the earlier Alpine–Betic orogen (Alboran basin) and in its foreland (Provençal and Valencia troughs). The extension appears clearly asymmetric due to its eastward polarity, accommodated by E-dipping master low-angle normal faults. Moreover the thinning shows variations in boudinage wavelength and is characterized by several along-strike transfer zones and heterogeneities. The western Mediterranean back-arc setting is comparable with Atlantic and western Pacific back-arc basins associated with W-directed subduction zones that show similar large-scale lithospheric boudinage.

    Notes: The western Mediterranean late Oligocene–Miocene basins (Alboran, Valencia and Provençal basins) are a coherent system of interrelated troughs. In all basins normal faults and thermal subsidence migrated toward the east progressively moving to the Miocene-to-Pleistocene Algerian and Tyrrhenian basins. All those troughs appear elements of the back-arc opening related to the eastward roll-back of the W-directed Apennines–Maghrebides subduction zone, similarly to western Pacific back-arc settings.These late Oligocene–early Miocene basins nucleated both within the Betic cordillera (e.g. Alboran sea) and in its foreland (Valencia and Provençal troughs). The N40–70° direction of grabens is oblique to the coexisting N60–80°-trending orogen and shows its structural independence from the orogenic roots. Thus, as the extension cross-cuts the orogen and developed also well outside the thrust belt front, the westernmost basins of the Mediterranean had to develop independently from the Alps-Betics orogen. Therefore, the Alboran extension, considered a classic example of a basin generated by the collapse of an orogen, cannot be ascribed to the detachment or annihilation of the lithospheric root. In contrast with the eastward migrating extensional basins, the Betic-Balearic thrust front was migrating westward producing interference or inversion structures.
