    Notes: Pyrimidine catabolism in Pseudomonas aeruginosa was investigated. It was found that the pyrimidine bases uracil and thymidine as well as their respective reductive catabolic products could be utilized as sole sources of nitrogen. Reductive degradation of the pyrimidine bases was noted. The reductive catabolic pathway enzymes dihydropyrimidine dehydrogenase, dihydropyrimidinase and N-carbamoyl-β-alanine amidohydrolase were all detected in minimal medium grown cells. Induction of pyrimidine catabolism by uracil was observed in this pseudomonad. Pyrimidine degradation in P. aeruginosa was not subject to catabolite repression.

    Notes: Abstract Proteins present in Saccharomyces cerevisiae ascospores and in germinating ascospores were compared using two-dimensional polyacrylamide gel electrophoresis. Yeast ascospores incorporated labeled methionine efficiently facilitating the electrophoretic analysis. Proteins synthesized in the yeast ascospores differed significantly from those proteins found 15 min after the initiation of germination in the ascospores. An immediate transition from ascospore proteins to proteins required for ascospore germination appears likely.

    Notes: The oxidative pathway of uracil catabolism was found to exist in Enterobacter aerogenes. When this bacterium was grown on a medium containing uracil as the sole nitrogen source, oxidative catabolism of uracil was shown by detection of the products barbituric acid, malonic acid and urea using thin-layer chromatography. The 2 enzyme activities associated with the oxidative catabolic pathway, uracil dehydrogenase and barbiturase, were measurable only when uracil served as the sole source of nitrogen for growth.

    Notes: Abstract Two mutants of the fungus Aureobasidium pullulans ATCC 42023 were isolated that exhibited elevated polysaccharide production. Both mutants were isolated using a combination of chemical mutagenesis and resistance to growth inhibitors. It was found that both mutants elaborated higher polysaccharide levels after 7 days of growth on corn syrup or sucrose, respectively, compared to ATCC 42023. The dry weights of the mutant cells were found not to differ greatly from those of the parent cells whether corn syrup or sucrose served as the carbon source. The pullulan content of the polysaccharide synthesized by the mutants or parent cells on sucrose was consistently lower than polysaccharide synthesized on corn syrup. Using corn syrup as a carbon source, the pullulan content of the polysaccharide elaborated by the parent was higher than either mutant. The inverse was found to occur with respect to pullulan content when the strains were grown on sucrose as a carbon source.

    Notes: Abstract A reduced pigmentation mutant was isolated from Aureobasidium pullulans ATCC 42023 by chemical mutagenesis and was subsequently characterized. The pigment melanin was present not only in A. pullulans cells but also contaminated the elaborated polysaccharide and thus, was measured in both fractions. Cellular and polysaccharide melanin levels of the mutant strain were at least 11-fold and 18-fold reduced, respectivelu, compared toits parent strain after 7 days of growth at 30°C whether sucrose or glucose served as the carbon source in the culture medium. Polysaccharide and cell dry weight levels of the mutant were very similar to those observed for the parent after growth on sucrose or glucose as the source of carbon over a period of 7 days at 30°C. The pullulan content of the polysaccharide produced by the parent or mutant strain was lower for sucrose-grown cells than for glucose-grown cells. It was also noted that the pullulan content of the polysaccharide elaborated by the mutant strain was slightly higher than that of the polysaccharide produced by the parent strain after growth on either sucrose or glucose.

    Notes: Abstract Pyrimidine base and ribonucleoside catabolic enzyme activities of the two type strains of the Pseudomonas diminuta group were investigated for taxonomic classification purposes. The presence of the pyrimidine salvage enzyme nucleoside hydrolase was indicated in both type strains following thin-layer chromatographic analysis. The presence of the hydrolase was also confirmed by enzyme assay. In addition, the activities of the pyrimidine salvage enzymes dihydropyrimidine dehydrogenase and dihydropyrimidinase were measurable in cell-free extracts of both P. diminuta and P. vesicularis. An absence of cytosine deaminase activity was found when assaying extracts of the two type strains. Nucleoside hydrolase and dihydropyrimidine dehydrogenase levels in P. vesicularis were influenced by carbon source while dihydropyrimidinase activity was observed to increase after P. diminuta growth on dihydrothymine as a nitrogen source.

    Notes: The only reported occurrence of quartz-rich sands in Jamaica is on the plains of Black River, St Elizabeth, where they outcrop in isolated pockets. The sands overlie limestones of the White Limestone Supergroup and the Coastal Group, and partly underlie Holocene peat deposits.Monocrystalline quartz, containing fluid and mineral inclusions, is the dominant mineral of the sands, with the heavy minerals. magnetite, haematite, epidote, rutile, anatase, zircon and tourmaline, occurring in minor amounts. Grain surface texture studies of the quartz grains have identified a series of mechanical and chemical features. Microtextures depicting marine, aeolian and pedological environments have been retained on the grains and represent the last three stages in the history of the grains.The petrography of the sands indicates an igneous provenance, with the major source of the detrital minerals being the Cretaceous granitoids that outcrop in the eastern half of the island. These rocks were unroofed during the uplift of Jamaica in the Late Tertiary and were subjected to extensive chemical weathering, which accounted for the breakdown of the majority of their primary silicate minerals. Fluvial transportation and deposition of detritus onto the shelf of the south coast commenced during this period and continued into the Pleistocene, providing terrigenous sand that was modified along the coast to quartz arenite by moderate to high energy marine conditions. Tidal currents, east-west longshore currents and weathering contributed to the maturity of the sand by the time it had been transported along the shelf to the vicinity of Black River. During low sea level stands in the Pleistocene, some of this sediment that became trapped on the shelf was blown inland by south-east winds.
