    Notes: Abstract A new cytotoxicity assay for determining the activity of epsilon toxin produced by Clostridium perfringens type D has been developed. Viability of cultured cells was determined by the ability of only live cells to convert 5-(3-carboxymethoxyphenyl)-2-(4,5-dimethylthiazolyl)-3-(4-sulfophenyl)tetrazolium to the coloured product formazan in the presence of phenazine methosulfate. Of the 12 cell lines tested, only the MDCK cell line was susceptible to epsilon toxin. Specificity was confirmed by the ability of only specific monoclonal antibodies to inhibit cytotoxicity. Good correlation was obtained with the mouse lethality assay (r = 0.991) and over a wide range of viability (15–75%) as determined by ethidium bromide/acridine orange staining (r = 0.995).

    Notes: Xanthomonas albilineans, the causal agent of leaf scald disease of sugarcane, produces a highly potent polyketide-peptide antibiotic and phytotoxin called albicidin. Previous studies established the involvement of a large cluster of genes in the biosynthesis of this toxin. We report here the sub-cloning and sequencing of an additional gene outside of the main cluster and essential for albicidin biosynthesis. This gene encodes a 634-amino-acid protein that shows high identity with the Escherichia coli heat shock protein HtpG. Complementation studies of X. albilineans Tox− mutants confirmed the requirement of htpG for albicidin biosynthesis and revealed functional interchangeability between E. coli and X. albilineans htpG genes. HtpG was co-localised with albicidin in the cellular membrane, i.e., the cellular fraction where the toxin is most probably biosynthesised. Here we show the requirement of an HtpG protein for the biosynthesis of a polyketide-peptide antibiotic.

The seasonal microdistribution of physico-chemical features and plankton were examined at close intervals in a high elevation (2493m) senescent crateri lake (1m deep) in the arid southwestern U.S.A.2. The protective volcanic rim and dense growth of provided highly stable thermal conditions throughout much of the year with the thermocline frequently in the top 30cm of the water column during the summer and dissolved oxygen typically 〈3.0mgl−1 in surface waters and 〈1.0mgl−1 below 50cm.3. The microdistribution of both phytoplankton and zooplankton was examined at close intervals (10cm) throughout the water column with the aid of a peristaltic pump.4. Aglaodiaptomus leptopus, along with Dinobryon sertularia and certain diatom species, were largely restricted to the top 30cm, while the colonial phytoflagellate, Pyrobotrys elongata and certain euglenoids were mostly located in the anaerobic lower strata.5. Cryptomonas obovata demonstrated subsurface maxima at various locations within the water column depending upon season.6. The distribution of plankton was also compared during daylight and darkness on one date.

Cladophora glomerata is the dominant filamentous green alga in the tailwaters of the Colorado River, U.S.A., below Glen Canyon Dam, but becomes co-dominant with filamentous cyanobacteria, Oscillatoria spp., below the confluence of the Paria River (26km below the dam) where suspended sediments are elevated.2. Benthic algal assemblages played an important role in the distribution of the amphipod, Gammarus lacustris, in the dam-controlled Colorado River through Grand Canyon National Park, Arizona. Cladophara and G. lacustris showed a weak positive relationship at ten cobble-riffle habitats in the Colorado River from Lees Ferry (25km below the dam) to Diamond Creek (362km downstream), while no relationship was found between Oscillatoria and G. lacustris.3. The relationship between algal substrata and G. lacustris was tested by a series of in situ habitat choice experiments. G. lacustris showed a significant preference for Cladophora (with epiphytes) over Oscillatoria spp., detritus and gravel in treatment pans at Lees Ferry.4. Epiphytic diatoms (i.e. food) were the overriding determinant of subtratum choice by G. lacustris in laboratory experiments. Gammarus chose the Cladophora/epiphytic diatom community over sonicated Cladophora with few diatoms. The amphipods also chose string soaked in diatom extract over string without diatom extract.5. Importance of mutualistic interactions in aquatic benthic community structure is discussed.

    Notes: SUMMARY. 〈list xml:id="l1" style="custom"〉1 The concomitant diet vertical migrations of the predaceous leech Erpobdella montezuma and its pelagic amphipod prey (Hyalella montezuma) were monitored for 1 year in Montezuma Well, Arizona, U.S.A.2 High densities of H. montezuma occurred in the subsurface strata of the water column during the day, but a substantial portion of the population migrated into the surface 1 m at sunset. E. montezuma remained in the lower water strata during the day, but migrated vertically after sunset to exploit the high densities of H. montezuma near the surface. Densities of E. montezuma progressively increased in the upper strata of the water column after sunset as light energy fell below 1 μEin m−2 s−1.3 It is suggested that the synchronized nocturnal migrations of the predator E. montezuma in relation to its prey H. montezuma, increases the foraging efficiency of the leech in the highly predictable environment of Montezuma Well.

The population dynamics and annual production of the multivoltine. pelagic amphipod Hyalella montezuma were studied over a 3-year period in the thermally constant environment of Montezuma Well. Arizona.2. H. montezuma showed two maxima which coincided with spring and autumn phytoplankton maxima. Juveniles comprised over 85% of the population in the pelagic zone compared to 37% in the littoral vegetation and there were significantly more females in the littoral vegetation. It appears that juvenile and adult H. montezuma show different habitat preferences.3. Total annual mean production for H. montezuma calculated by the size frequency method and adjusted for multiple generations, was 357 kg ha−1 yr−1, which is higher than single-species production estimates reported for most zoobenthic amphipods and multivoltine planktonic crustaceans. Average energy production for H. montezuma was 4640 kJ ha−1 yr−1 in the pelagic zone and 1072 kJ ha−1 yr−1 in the littoral vegetation.4. Average cohort P/B̄ ratios for H. montezuma were higher in the pelagic zone (5.5) than in the litttoral vegetation (3.7). Juveniles had higher cohort PB̄ ratios than adults in the pelagic zone, while the reverse relationship was true in the littoral vegetation. We propose that different size-selective predators may contribute to the differences in PB̄ ratios for juveniles and adults in these two habitats.

    Notes: We present a new survey of several Palaeozoic sections in both the Taurus range and the Border Folds that documents typical glacial features including a glacial pavement and striated dropstones (Halevikdere Formation) and demonstrates the former presence of an ice sheet in southern and south-eastern Turkey. Evidence for the late Ashgill (Hirnantian) age of this episode is provided by macro- and microfossils found within the glacial formation. The extension of ice-related deposits into the northernmost part of the Arabian Platform (Mardin region) implies a much wider distribution of the Ordovician ice sheet than was previously believed, and strongly suggests that southern Turkey lay close to Egypt during the Lower Palaeozoic.
