    Notes: Following NTG mutagenesis, four independent mutants of Anabaena PCC7120 defective in heterocyst differentiation were isolated. These fell into 2 distinct classes; (1) those unable to differentiate heterocysts or show whole-cell acetylene reduction activity; and (2) those unable to differentiate heterocysts but capable of microaerobic acetylene reduction. All mutants grew equally well as the wild type with added nitrogen sources and showed no apparent differences in glutamine synthetase or glutamate synthase activities compared with the wild type. The mutants of class (2) evolved H2 only under microaerobic conditions, suggesting that H2 is evolved via nitrogenase in Anabaena PCC7120.

    Notes: Two detailed vertical profiles through a complex plume of phenolic contaminants in a Triassic sandstone aquifer show that natural attenuation by biodegradation and dispersion is active but very slow. The plume has a microbially active aerobic and NO3 reducing fringe that is less than 2 m thick at both 150 and 350 m downstream of the source. The anaerobic core has evidence of active bacterial populations and degradation at total organic carbon (TOC) concentrations up to at least 1400 mg/L (1800 mg/L total phenolics), although gross half-lives are more than 50 years. There is evidence from the same locations of Mn, Fe, and SO4 reduction, with the latter inhibited by the pollutant matrix and not significant at concentrations more than 1000 mg/L TOC. Degradation of these contaminants in this aquifer is influenced by a range of environmental factors, including the chemical toxicity and pH of the contaminant matrix, and inputs of electron acceptors into the plume by dispersion. The results show that the plume is likely to grow under the present conditions, despite the biodegradable nature of the organic pollutants and availability of suitable electron acceptors. Vertical profiles have proved a cost-effective method of understanding the evolution of the plume.

    Notes: This paper explores the application of a catchment faecal-indicator budget approach to the examination of faecal-indicator sources impacting on coastal recreational water quality.     Notes: : With the recent increased exploration and mining activity in the Northwest Territories, there has been growing interest in streamflows. However, streamflow monitoring in Canada's north is limited, especially in the central Northwest Territories where the exploration activity is concentrated. To complicate matters, the standard approach of measuring discharge with current meters or weirs is often inadequate or prohibitively expensive, as many streams in the region are shallow, braided and rocky. In response, alternative techniques such as salt dilution can be used. A salt tracer's competence in turbulent and rocky channels makes it ideal for discharge measurements in these situations. This paper summarizes the work performed by Indian and Northern Affairs Canada and Canamera Geological Ltd. staff in determining the stream discharge of a lake outlet using a potassium chloride (KC1) tracer. A variety of streamflow measurement methods were performed and compared to determine the viability and rigor of the dilution method. Results suggest the dilution method compares favorably to other measurement techniques both in accuracy and operational ease.

    Notes: The amiE gene encodes an aliphatic amidase capable of converting fluoroacetamide to the toxic compound fluoroacetate and is one of many genes whose expression is subject to catabolite repression control in Pseudomonas aeruginosa. The protein product of the crc gene, Crc, is required for repression of amiE and most other genes subject to catabolite repression control in this bacterium. When grown in a carbon source such as succinate, wild-type P. aeruginosa is insensitive to fluoroacetamide (due to repression of amiE expression). In contrast, mutants harboring the crc-10 null allele cannot grow in the presence of fluoroacetamide (due to lack of repression of amiE). Selection for succinate-dependent, fluoroacetamide-resistant derivatives of the crc-10 mutant yielded three independent pseudorevertants containing suppressors that restored a degree of catabolite repression control. Synthesis of Crc protein was not reestablished in these pseudorevertants. All three suppressors of crc-10 were extragenic, and all three also suppressed a Δcrc::tetA allele. In each of the three pseudorevertants, catabolite repression control of amidase expression was restored. Catabolite repression control of mannitol dehydrogenase production was also restored in two of the three isolates. None of the suppressors restored repression of glucose-6-phosphate dehydrogenase or pyocyanin production.

    Notes: SUMMARY. Populations of Potamogeton richardsonii in Sparrow Lake, Ontario, vary greatly in leaf dimensions and internode length. Leaf length/breadth (L/B) ratio is increased by low irradiance, significantly at 4% daylight and, in a contrary fashion, by ontogenetic drift at high irradiances of shallow water. L/B ratio was not correlated with substrate. These findings suggest that this ratio is of doubtful value taxonomically or as an integrator, as specific leaf area is confirmed to be, of factors like photosynthetically active radiation (PAR). Longest internodes belong to plants from deep(1.8 m) water, and, in a summer-grown high-density population, internode length decreases logarithmically as depth lessens; both observations implicate PAR and ageing. Relative to undisturbed, shallow-water shoots, young transplants in full daylight elongate almost twice as fast because of the production of more, and longer, internodes. Young shoots in 12% daylight lengthen even more rapidly than those in full daylight (and four times faster than undisturbed shoots) in the same period because of quicker elongation of the same number of internodes. At ambient summer temperatures, this rate of elongation is inversely related to PAR. Experimental and seasonal field data indicate that maximal internode extension occurs on young shoots in very low irradiances at temperatures of 9–15°C; in nature, effects of increasing water temperatures are depressed by increasing irradiances and ageing until minimal extension takes place in old shoots at mid- to late-summer temperatures and in high irradiances. In tanks (40 m3) in full daylight, nutrients limit growth of plants on sand before they limit growth on clay (with marl intermediate) whereas, at 12% daylight, light limits growth on sand before soil nutrients do. The leaf area index on silty sand on an exposed shore was 0.4 at 0.5 m depth, in contrast to a cultured population where it was 4.0 in silty loam at the same depth. Thus, while it has not been possible to explain some variations in leaf morphology of P. richardsonii in environmental terms, the differential effects of ontogeny, PAR and temperature on shoot growth have been assessed, along with overall effects of light and nutrients on biomass and of shelter on leaf area index.

    Notes: SUMMARY. Estimates of relative and absolute abundance in field populations of two Gerris species were compared using regression techniques. Separate equations for each gerrid size class allow prediction of population density from timed-catch sampling. Presence or absence of vegetation markedly affected capture rates, but capture rates were not affected by Gerris species or type of emergent vegetation. Availability for capture varied with gerrid leg length; this relationship can be used to estimate availability constants for other water-strider species.
