    Notes: Using 20 bankers as subjects, this exploratory study examined the effects of audit committees on bankers’ perceptions of auditor independence. Results reject the null hypothesis which suggest that bankers place more reliance on financial statements in considering loan applications given information on the presence of audit committees than given information on their absence. Those bankers given information on the presence of audit committees recommended a lower interest rate premium than those given information on the absence of audit committees. These findings suggest that bankers given information on the presence of audit committees perceived a lower loan risk as a result of a higher confidence in the auditor to maintain independence than those given information on the absence of audit committees. It can then be concluded that audit committees had a significant effect on bankers’ perceptions of auditor independence.

    Notes: Abstract Sulphate incorporation into glycopeptides appears to be a key event in the development of a number of organisms. An inhibitor of sulphation, sodium selenate, has been used in this study to examine the possibility that sulphation has a comparable role in the development of Dictyostelium discoideum. At concentrations of 0.1 mM and 1.0 mM, exogenously supplied selenate reversibly arrested the growth of bacterially grown amoebae of D. discoideum. In contrast, the effect of selenate on development was minimal. In the presence of 1.0 mM selenate, aggregation and tip formation were delayed 2–3 h and aggregates were slightly smaller; exogenous 0.1 mM selenate had no visible effect on development. However, the possibility that starved amoebae are impermeable to selenate was not excluded. The vegetative growth and development of an axenic strain in the presence of selenate closely resembled that of the bacterially grown strain. Since an inhibitory effect of 1.0 mM selenate on [35S]sulphate incorporation into acetone precipitable protein was also demonstrated, these results suggest that sulphation is necessary for the growth of D. discoideum.

    Notes: Abstract An ‘actomyosin’ complex was purified from Escherichia coli W3110 using selective precipitation. The complex contains three major components of 19.5, 18.5 and 17 kDa. The 19.5- and 17-kDa proteins were purified by electroelution, peptide mapped and N-terminally sequenced. The structural gene for the 17-kDa protein was found to have been previously identified in an operon containing several other genes including the essential lpxA, lpxB and dnaE. The possible function of the 17-kDa protein and the other ‘actomyosin’ components is discussed.

    Notes: The post-prandial glycemic response of juvenile European sea bass (initial mean weight 10 g) was determined following acclimation to experimental diets that contained a pea seed meal included at two levels. The ingredient provided a source of starch that replaced purified corn starcWyellow dextrin (2:1) in a reference diet based on fishmeal as the main protein component. The total carbohydrate levels amounted to 25% of the dry matter content and contributed (18%) to the overall gross dietary energy value of 20 MJ/kg. After a 12-wk feeding trial, in which the sea bass grew to over 40 g (mean weight), the plasma glucose profile was recorded over a 24-h period (0 h, 1 h, 3 h, 6 h, 9 h, 12 h, and 24 h) following a meal fed to satiation. Results showed that sea bass are able to regulate their glucostatic tolerance to dietary starch, and that the pea seed meal modulated this effect. Peak post-prandial responses varied between 1 to 9 h and were significantly delayed for the reference diet. Efficacy of glucose assimilation improved for the highest level of pea seed meal inclusion (40% by weight). The relevance of carbohydrate complexity with respect to feed formulation and nutritional requirements of sea bass are discussed.

    Notes: Alternative sigma factors are key global regulators that coordinate bacterial responses to environmental changes necessary for adaptation and survival. In turn these sigma factors are controlled by regulators such as anti-sigma and anti-anti-sigma factors. In this report, using a cDNA–total RNA subtractive hybridisation strategy that we have developed previously, we identified increased transcription of a potential sigma factor regulatory gene, Rv1364c, in Mycobacterium bovis BCG upon phagocytosis by macrophages and this was confirmed by Northern blot analysis. Primer extension analysis revealed the use of alternative promotors, P1 and P2, and that the increased expression inside macrophages coincided with promoter switching from P2 to P1. Rv1364c (653 amino acids), originally annotated as RsbU, contains structural domains homologous to the PAS redox sensor, the protein phosphatases anti-anti-sigma factor RsbU/SpoIIE, the protein kinase anti-sigma factor RsbW/SpoIIAB and the anti-anti-sigma factor RsbV/SpoIIAA found in other bacteria. These findings have important implications for understanding coordination of the expression of sigma factors under intra-macrophage conditions. Other potentially differentially expressed genes, including genes for fatty acid metabolism, membrane transportors, heat shock proteins, potential sigma factors and energy metabolic pathways are also listed and their biological significance discussed.

    Notes: Antisera raised to a 109 kDa wall-associated protein (WAP) of Bacillus subtilis 168 cross-reacts with two other WAPs of 220 and 58 kDa. The structural gene for the 109 kDa WAP (designated wapA) was cloned, sequenced, mapped at around 340° on the B. subtilis 168 chromosome and found to encode a precursor of all three wall-bound forms (2334 amino acids and 258 329 Da). The protein has two ligand-binding domains; the N-terminal domain has three direct repeats of 102 residues with 40% identity, which are responsible for wall binding. The C-terminal domain consists of two blocks of residues with a conserved motif repeated a total of 31 times. The motif consensus sequence GXXXX(Y,F)XYDXXG is almost identical to that of the Escherichia coli rearrangement hot spot family and shows similarity to a carbohydrate-binding motif of a number of Gram-positive secreted proteins. A mutant insertionally inactivated in the wapA gene had no distinguishable phenotype apart from lacking the three WAPs. The possible role of WAPA and its two-domain relationship with other ligand-binding proteins is discussed.

    Notes: The peptidoglycan hydrolase profile of Listeria monocytogenes EGD has been characterised under a variety of environmental and physiological conditions, using renaturing sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The profiles show activities ranging from 29 to 186 kDa. The 186-kDa enzyme was only observable under specific medium and aeration conditions. The enzyme activities show differential substrate specificity and sensitivity to incubation conditions. The peptidoglycan hydrolase profile of several different Listeria strains was also compared.

    Notes: Conditions for zymographic detection of a 41-kDa spore cortex hydrolysis-specific autolysin, A6, from Bacillus subtilis 168 were optimised. A6 was present during sporulation from stages II–IV and remained active in the dormant spore. Its expression was controlled by the mother cell-specific early-sporulation sigma factor σE. The characteristic muramic acid δ-lactam of spore cortical peptidoglycan was not necessary for cortex hydrolysis by A6, but it may be important in the inability of the major vegetative autolysin LytC to digest wild-type cortex. Two other minor autolysins were also observed during sporulation. The possible physiological significance of these observations is discussed.
