Notes: Our objective was to determine the effect of a diet rich in tofu (soybean curd) on the main blood components of rats. This was done in comparison to diets with casein, soybean proteins or soybean oil, enabling the observed biological effect to be related to the composition of the tofu. Animals on the tofu diet had very low cholesterol levels and an acceptable HDL cholesterol / total cholesterol ratio. Growth and amino acid profile were also adequate, but the bioavailability of trace elements decreased.

Notes: Aspergillus niger xylanase is a target enzyme of the two wheat proteinaceous inhibitors, XIP-I and TAXI-I. We previously suggested that the xylanase “thumb” region was XIP-I binding site. Here, we expressed the Asp37Ala mutant in Pichia pastoris and showed that the mutation abolished the enzyme capacity to interact with both inhibitors, suggesting a direct contact at the active site. The mutant pH profile was altered, confirming the key role of Asp37 in determining the pH optima of glycoside hydrolase family 11. The results are consistent with a competitive inhibition mode and underline the strategic importance of Asp37 in the inhibition mechanism.

Notes: Abstract Homologous rDNA probes were used to study the number of rRNA genes in a five-generation genealogy of Streptococcus thermophilus CNRZ368. Whereas the CNRZ368 strain contained six rRNA loci, four independant mutants with five rrn loci were obtained. The deletion frequency was 5 × 10−2. Molecular analysis provided identical hybridization patterns for all the deletion mutants. The deletion was shown to occur within the two close rRNA loci, rrnD and rrnE, probably due to a homologous recombination event and to give rise to a hybrid rrnD/E locus.

Notes: Several E-W profiles of Bouguer anomalies across the Precambrian basement were inverted by applying a linearized inversion procedure in the spectral domain and using a simplified two-layer model with a crust-upper mantle interface of constant density contrast. From these inverted profiles, a contour map of Moho topography has been constructed which covers the area of exposed Precambrian basement in the eastern and central parts of the island. Such imaging of the Moho depth in particular exhibits a N20°E-trending zone of substantially thinned crust along the axis of the island. This parallels the east coast margin which resulted from the northward motion of India relative to Madagascar in the Upper Cretaceous. From combined analysis of geological, tectonic and metamorphic observations, this prominent crustal feature in the Precambrian basement of Madagascar is interpreted as the relic of continental lithospheric extension and thinning possibly related to Panafrican collapse tectonics on the eastern edge of the Mozambique belt. This extensional feature would have been reactivated only in part during the rifting and dislocation stages of the Madagascar and India continental blocks as evidenced by an axis of Cretaceous volcanism emphasized by the Ankaratra and Itasy massifs. Final separation nevertheless occurred further east, at the site of the east coast margin.

Notes: A chromosomal repeated sequence from Streptococcus thermophilus was identified as a new insertion sequence (IS), IS1191. This is the first IS element characterized in this species. This 1313 bp element has 28 bp imperfect terminal inverted repeats and is flanked by short direct repeats of 8bp. The single large open reading frame of IS1191 encodes a 391-amino-acid protein which displays homologies with transposases encoded by IS1201 from Lactobacillus helveticus (44.5% amino-acid sequence identity) and by the other ISs of the IS256 family. One of the copies of IS 1191 is inserted into a truncated iso-IS981 element. The nucleotide sequences of two truncated iso-IS981 s from S. thermophilus and the sequence of IS981 element from Lactococcus lactis share more than 99% identity. The distribution of these insertion sequences in L. lactis and S. thermophilus strains suggests that intergeneric transfers occur during co-cultures used in the manufacture of cheese.
