    Notes: Abstract: Proteinase and peptidase activities of the fission yeast Schizosaccharomyces pombe were investigated. Several intracellular proteolytic enzymes were found: two endoproteinases, one carboxypeptidase, one aminopeptidase and one dipeptidyl-aminopeptidase. In addition, proteinase inhibitors were detected. In fresh crude extracts an activation procedure is needed to measure maximal activities of endoproteinases and carboxypeptidase, whose level is markedly dependent on growth medium composition and on growth phase, while aminopeptidase and dipeptidyl-aminopeptidase activities are very little, if at all, regulated by the carbon source.

    Notes: Abstract The steady-state levels of the aminopeptidase Y (APY1) transcript and the levels of assayable aminopeptidase Y activity were measured under a variety of nutritional conditions. Specific APY1 mRNA is less abundant in cells grown in minimal medium than in cells grown in rich medium, while active enzyme levels follow just the opposite pattern. Aminopeptidase Y activity decreases when cells are deprived of glucose without a concomitant fall in mRNA levels. Production of aminopeptidase Y is not markedly affected by nitrogen regulation. APY1 gene expression is not disturbed in heat-shocked cells. Our data support the idea that the main control event in vacuolar aminopeptidase Y expression is a post-transcriptional step.

    Notes: We determined the nucleotide sequence of a 4599-bp DNA genomic fragment including the γ-actin encoding gene from Blakeslea trispora, showing an open reading frame of 1561 bp interrupted by four introns with fungal consensus splice-site junctions. The untranslated regions of the actA gene contain a consensus TATA box, a CCAAT motif, a large pyrimidine stretch, and the polyadenylation sequence AATAAA. The predicted protein (375 amino acids) revealed high identity to γ-actins from fungi (〉90%), and gene phylogenies support the grouping of B. trispora actin close to those from the majority of the filamentous fungi. actA transcript (1.4 kb) level in β-carotene producing conditions was faintly higher than carRA (1.9 kb) and slightly lower than carB (1.8 kb) β-carotene biosynthetic genes. The use of the actA promoter (PactA) for heterologous gene expression was ascertained by the transformation of gene fusions with the bleomycin resistance gene (bleR) from Streptoalloteichus hindustanus and the geneticin resistance marker (aphI) from Tn903, into Escherichia coli and Acremonium chrysogenum.

    Notes: Abstract A yeast mutant lacking non-specific α-mannosidase activity was found as a background marker during our search for dap2 mutants (Suárez-Rendueles, P. and Wolf, D.H. (1987) J. Bacteriol. 169, 4041–4048). The mutant (DPS-15) is characterized in detail. The mutation called amd1 segregates 2:2 in meiotic tetrads, indicating a single chromosomal gene mutation which is recessive. Diploids heterozygous for amd1 show gene dosage. Thus, it appears that AMD1 might be the structural gene for α-mannosidase. Results obtained with this mutant show that α-mannosidase is not a vital component of the vegetative cell cycle. The differentiation process of sporulation is not disturbed in homozygous mutant diploids. Mannose turnover does not seem to be altered in mutant cells.

    Notes: Abstract Mycoplasmas have been incriminated in setting the stage for HIV infection and full-blown AIDS. We tested the possible involvement of mycoplasmas in activation of HIV. Two cell lines, 293 fibroblasts and Jurkat CD4+ T-cells, transfected with plasmids harboring a transcription fusion construct between HIV-long terminal repeat (HIV-LTR) and either luc or cat genes, were infected with several mycoplasmas (M. fermentons; M. penetrons M. pirum and Ureaplasma urealyticum) and the reporter gene expression was monitored. The data presented here suggest that mycoplasmas, and specifically their membranes, play a role in the activation of HIV-LTR mediated transcription.

    Notes: Post-harvest application of acetaldehyde vapors to blueberries, tomatoes, and pears led to the enhancement of the fruit sensory quality including an increase in the sugar content, sugar-acid ratio, and flavor changes judged to be acceptable by test panels. Applied ethanol vapors led to similar but limited enhancement of fruit sensory quality. Comparative applications of ethylene were more effective in stimulating changes in the background color, including an increase in the total carotenoid content in tomato and anthocyanines in blueberry. Ethylene, however, had little or no effect, and occasionally led to deleterious changes in the fruit sensory quality. Acetaldehyde and related volatiles may be important in the development of fruit sensory quality, as occurring normally during ripening, or as a post-harvest application for the improvement of fruit sensory quality.

    Notes: Coeliac disease (CD) is associated with particular HLA genotypes. The susceptibility gene (or genes) has been mapped to the class II region, most probably to the DQ loci. Polymorphism of the upstream promoter region of the DQA1 gene (QAP) has been recently reported. At least ten variants or QAP alleles have been found, some of which are present in the as-acting regulatory sequences. Allelic differences in DQ molecule expression may play a role in susceptibility to CD. We investigated the QAP polymorphism in 102 CD patients and 142 unrelated healthy controls of Czech origin using polymerase chain reaction amplification (PCR) of genomic DNA and oligonucleotide probes. We found a significant frequency increase of the alleles QAP 4.1 (RR = 10.3, p.c. = 10-6) and QAP 2.1 (RR = 2.4, p.c. = 0.017) in patients over controls. An increased susceptibility is provided by the presence of both alleles, as is shown by the higher proportion of QAP 4.1, 2.1 heterozygotes among patients than expected from the Hardy-Weinberg equilibrium and by the comparison of the odds ratios for these alleles. There is a strong linkage disequilibrium between the QAP alleles and the DQA1, DQB1, and DRB1 loci. Two haplotypes carrying the QAP alleles whose frequency is increased are predominant in this group of CD patients: DQB1* 0201, DQA1* 0501, QAP 4.1, DRB1* 0301 and DQB1* 0201, DQA1* 0201, QAP 2.1, DRB1* 0701. Thus, the QAP variants are increased as part of these haplotypes and we cannot discriminate if they are responsible for the primary association.

    Notes: Second generation Penaeus vannamei broodstack, pond-reared and wild populations, were compared for spawning frequency and several variables associated with reproductive performance. For this purpose, 470 pond-reared and 237 wild-caught individually tagged females were stocked with males of the same origin in ten maturation tanks in a commercial hatchery, and several productivity variables were recorded over time. Results of the analysis indicate that wild shrimp had higher mating and spawning frequencies compared to pond-reared broodstock. The number of nauplii per spawn was higher for wild shrimp, but fertilization and hatching rates were higher for pond-reared spawners. A larger proportion of nonmating females were found in pond-reared shrimp; whereas, wild females had a higher potential for multiple matings. The spawn quality (fertilization and hatching rates, percentage of viable spawns, and number of nauplii) in consecutive matings did not decline substantially. The present study indicates that selection of females with multiple spawning capabilities can be an important commercial strategy to improve nauplii production over the short term. Pond-reared broodstock can be an adequate source for applying this strategy because of their year-round availability and an overall acceptable reproductive performance.
