    Notes: Abstract Two Tn917-generated bacitracin deficient mutants of Bacillus licheniformis were isolated. Southern blot analysis of chromosomal DNA extracted from both insertional mutants showed that Tn917 inserted in the vicinity of the gene coding for the enzyme BA2 of the bacitracin synthetase enzyme complex. Measurements of bacitracin synthetase activity in cell-free extracts and positive hybridization signals in the vicinity of the BA2 gene indicate that in both bacitracin deficient mutants Tn917 could be inserted in the BA1 gene or in segments involved in regulation. Thus, it could be possible that the genes for bacitracin synthetase are clustered in the B. licheniformis genome.

    Notes: Deposition from at least three episodes of glaciation left a complex glacial-drift aquifer system in central Illinois. The deepest and largest of these aquifers, the Sankoty-Mahomet Aquifer, occupies the lower part of a buried bedrock valley and supplies water to communities throughout central Illinois. Thin, discontinuous aquifers are present within glacial drift overlying the Sankoty-Mahomet Aquifer. This study was commissioned by local governments to identify possible areas where a regional water supply could be obtained from the aquifer with minimal adverse impacts on existing users.Geologic information from more than 2200 existing water well logs was supplemented with new data from 28 test borings, water level measurements in 430 wells, and 35 km of surface geophysical profiles. A three-dimensional (3-D) hydrostratigraphic model was developed using a contouring software package, a geographic information system (GIS), and the 3-D geologic modeling package, Earth Vision®. The hydrostratigraphy of the glacial-drift sequence was depicted as seven uneven and discontinuous layers, which could be viewed from an infinite number of horizontal and vertical slices and as solid models of any layer. Several iterations were required before the 3-D model presented a reasonable depiction of the aquifer system. Layers from the resultant hydrostratigraphic model were imported into MODFLOW, where they were modified into continuous layers. This approach of developing a 3-D hydrostratigraphic model can be applied to other areas where complex aquifer systems are to be modeled and is also useful in helping lay audiences visualize aquifer systems.

    Notes: Ground water contamination was discovered in 1981 in a monitoring well at the Earthline disposal facility near Wilsonville, Illinois. Organic chemicals had migrated at a rate 100 to 1000 times greater than predicted when the site received its permit to operate in 1978. Postulated failure mechanisms included migration through previously unmapped permeable zones, subsidence of an underground mine, organic-chemical and clay-mineral interactions, acid-mine drainage and clay interactions, trench-cover settlement, and erosion.In this investigation, the Illinois State Geological Survey found the primary reason for the rapid migration: the presence of previously undetermined fractures and joints in glacial till. The inaccurate predictions of hydraulic conductivity were based on laboratory-determined values that did not adequately measure the effects of fractures and joints on the transit time calculations. Field-measured hydraulic conductivity values were generally 10 to 1000 times greater than their laboratory-measured counterparts, thus largely accounting for the discrepancy between predicted and actual migration rates in the transit time calculations. The problem was compounded, however, by the burial of liquid wastes and by trench covers that allowed excess surface runoff to enter the trenches. Organic-chemical and clay-mineral interactions may also have exacerbated the problem in areas where liquid organic wastes were buried.

    Notes: Most guidelines for sampling ground water require the evacuation of multiple bore volumes from the well before a sample is collected. Such recommendations are impractical for wells that are finished in fine-grained deposits with such slow recharge that they cannot recover rapidly enough for the requisite number of well volumes to be removed. In such situations, the sample is usually collected either 24 hours after evacuation or sometime during well recovery. Neither of these strategies has been supported by field evidence.The purpose of this study was to determine the changes in volatile organic chemical concentration during well recovery. Three hundred samples were analyzed for up to 19 different volatile organic compounds from 11 wells that were finished in fine-grained glacial tills. Each well was sampled before purging and at several time intervals up to 48 hours after purging. The experiment was conducted three to five times on each well. Samples were collected with dedicated point-source PTFE bailers equipped with bottom-emptying devices that were designed for collecting samples for volatile organic chemical analysis. All wells were evacuated with a bailer because they were finished in slowly recharging materials having hydraulic conductivities of between 1 × 10−6 and 7 × 10−5 cm/sec.Results of the chemical analyses were examined statistically using a general linear model and the Tukey honestly significant difference test to ascertain whether the changes in water quality with time after purging were statistically significant. At the 95 percent confidence level, there was no significant difference in chemical compositions at any time interval after purging, although samples collected at four hours had slightly higher concentrations than did earlier or later samples. However, concentrations of volatile organics were significantly lower before purging than after purging. These results clearly show that wells finished in fine-grained sediments should be purged before samples are collected for volatile organic chemical analysis.
