    Notes: Abstract In the light leaf spot fungus Pyrenopeziza brassicae, the kinetics of uptake of sucrose, glucose and fructose are all biphasic. At low and high concentrations, glucose and fructose share a high-affinity and a low-affinity hexose carrier respectively, with Kms of 3.5 and 4.6 μM for uptake of glucose and fructose respectively by the high-affinity system, and Kss of 690 and 750 uM for uptake of glucose and fructose by the low-affinity system. The data also suggest the existence of separate high-affinity and low-affinity uptake systems for sucrose. P. brassicae possesses both soluble and paniculate invertase activity, with pH optima of 5.0 and 4.0 respectively. Activity of the particulate invertase is considerably in excess of the highest rates of sucrose uptake.

    Notes: Abstract The higher homologues of cadaverine, aminopropylcadaverine (APC) and N,N-bis(3-aminopropyl)cadaverine (3APC) were formed by a wild-type strain of Saccharomyces cerevisiae, and by two mutant strains, spe 3-1 and spe 4-1, exhibiting point mutations in the genes for spermidine synthase and spermine synthase, respectively. This, together with the incomplete inhibition of APC and 3 APC formation in the presence of inhibitors of 5-adenosylmethionine decarboxylase and spermidine synthase, suggests that the cadaverine derivatives are formed partly by the operation of a different route. However, the yeast strains were unable to utilise [14C]aspartate and lysine to form APC and 3APC. Since the ornithine decarboxylase inhibitor adifluoromethylomithine (DFMO) greatly reduced the formation of APC and 3APC, it is suggested that these compounds are formed preferentially in these yeast strains from cadaverine formed by ODC. APC and 3APC formation in the yeast strains was increased substantially following exposure to 37 °C for 2 h.

    Notes: The predominant filamentous bacteria which are found in bulking activated sludge in the UK have been assessed from a microscopic analysis of samples taken over a three-year period. There were major differences in the floc biology of samples received from plants treating predominantly domestic wastewaters and those of industrial origin. Most samples (84%) of domestic origin were dominated by two filament types: Microthrix parvicella was the dominant filament in 63% of the samples and Type 021N dominated in 21% of the samples. Nostocoida limicola and Sphaerotilus natans dominated in the remaining 16% of samples. The samples of industrial origin showed much greater filament diversity, with eight filament types routinely observed in high numbers. Type 021N was the most prevalent and M. parvicella was not observed in the industrial samples. Many domestic samples from sequencing batch reactors were dominated by M. parvicella. Based upon knowledge of the predominant filament type from microscopic examination. it was possible to identify the likely causes for their proliferation and suggest long-term solutions to achieve their eradication.

    Notes: The fish community of the small (17·5 ha) intermittently open East Kleinemonde estuary was sampled between 1994 and 1997 to estimate population size, standing stock, growth and productivity. The estuarine-spawning species were numerically more abundant (n c. 750 000) but due to their small size contributed only 11·7% to the total biomass. The total annual productivity of all fishes in the estuary (n c. 890 000), with a standing stock of 28·44 g m −2, was calculated at 55·89 g m−2 year−1. The small sparid Rhabdosargus holubi with a production estimate of 41·35 g m−2 year−1 accounted for 〈74% of the total fish production in this estuary.

    Notes: Five transposon mutants of Francisella novicida were isolated that are compromised in their ability to grow in mouse macrophages in vitro. Sequence analysis of the DNA flanking the transposon insertions identified the genes that were interrupted in these mutants. One of the inactivated loci corresponds to the Francisella tularensis gene that encodes a 23-kDa protein that is the most prominently induced protein following macrophage infection. Another insertion was localised to approximately 2 kb upstream of the gene encoding the 23-kDa protein. By analysis of the incomplete Francisella genome sequence it was surmised that these two insertions disrupt different portions of a putative operon that encodes four proteins, none of which have discernible functions. Three other interrupted loci associated with poor intramacrophage growth showed similarity at the deduced amino acid level to alanine racemase, the ClpB heat-shock protease, and the purine biosynthetic enzyme, glutamine phosphoribosylpyrophosphate amidotransferases.

    Notes: In order to identify genes involved in LPS biosynthesis we isolated random mutants generated by transposon insertion in Francisella novicida. The resulting mutant bank yielded mutants with three distinct LPS phenotypes, and three representative mutants were chosen for further study. One mutant that had short O-antigen chains was sensitive to serum; this mutant and one other were more sensitive to killing by deoxycholate than control strains. The third mutant was resistant to deoxycholate killing but slightly sensitive to serum. The three mutants varied in their ability to grow in macrophages. The DNA sequences interrupted by the transposon in two of the three mutants showed similarity to known LPS biosynthetic genes at the deduced amino acid level.

    Notes: Biomass production by Aspergillus fumigatus was greatest at 40–45°C and was associated with an increase in concentration of the diamine putrescine and activity of its biosynthetic enzyme ornithine decarboxylase. Concentrations of the other amines, cadaverine, spermidine and spermine were considerably lower than putrescine concentration and did not change significantly over the temperature range 20–50°C. This is surprising in view of the greatly increased flux of label from ornithine through to spermidine at 45 and 50°C, indicating an increased formation of this triamine. It is suggested that there was increased formation of spermidine derivatives at these temperatures. Interestingly, there was greatly increased formation of the higher homologues of cadaverine, aminopropylcadaverine and N,N′-bis(3-aminopropyl)cadaverine, in A. fumigatus at 45 and 50°C.

    Notes: We have previously shown that when cultured in vitro, peritoneal rat macrophages infected with Francisella novicida spontaneously release nitric oxide in sufficient quantities to inhibit bacterial growth. However, it is not known whether F. novicida can have a similar antimicrobial effect in vivo. Here we show that a co-infection of F. novicida with Francisella tularensis can suppress the number of F. tularensis cells in rat spleens by as much as 100-fold.
