    Notes: 16S rRNA gene libraries were prepared by polymerase chain reaction amplification and cloning from soil samples taken periodically from a field with genetically modified plants. Sequence analyses of the cloned rDNAs indicated that 140 of them clustered apart from known bacterial phyla. Based on 31 full sequences a new phylum could be defined. It includes Holophaga foetida, ‘Geothrix fermentans’ and Acidobacterium capsulatum as the only cultured species so far. Therefore, this line of descent was named the Holophaga/Acidobacterium phylum. About 50 published partial sequences of cloned rDNAs retrieved from soil, freshwater sediments or activated sludge from different continents indicate the occurrence of further representatives of this phylum. Two specific hybridization probes were constructed for members of one of four subclusters. A careful data analysis revealed the importance and problems of identifying and dealing with artefacts such as chimeric structure when defining new phylogenetic groups based mainly upon cloned amplified rDNAs. For the first time, the presence of bacterial cells representing this group could be shown in soil, sediment, activated sludge and lake snow by in situ hybridization.

    Notes: Abstract Photosynthetic and anatomical parameters of leaves from the juvenile and adult part of an ivy plant (Hedera helix L.) have been determined and compared with each other. Light-saturated net photosynthesis (per unit leaf area) was about 1.5 times higher in adult leaves than in juvenile ones. The lower photosynthetic capacity of juvenile leaves was caused by a lower stomatal and especially a lower residual conductance to the CO2-transfer. This corresponds with anatomical features of the leaves, i.e. lower stomatal frequency, fewer chloroplasts per cell, and – most important – thinner leaves, as well as with a less efficient photosynthetic apparatus measured as Hill reaction of isolated broken chloroplasts and activity of ribulose bisphosphate carboxylase. No differences in the respiration in light (relative to net photosynthesis) and in the CO2-compensation concentration could be detected between the two leaf types. These observed anatomical and photosynthetic parameters of the juvenile and adult ivy leaves resemble those reported for shade and sun leaves, respectively, although the leaves investigated originated from the same light environment.

    Notes: This research contributes to the understanding of reactions to different selection screening methods. A sample of students (n=153) experienced one of three types of screening techniques, face-to-face interview screenings, telephone interview screenings, and interactive voice response (IVR) screenings, with identical content in a pre- to post-screening longitudinal study. We further examined the role of two important individual differences, cognitive ability and conscientiousness, in attitudes toward the screenings. IVR is a “non-interpersonal” screening method so it was not surprising that it was rated lower in terms of procedural justice factors such as interpersonal treatment, two–way communication, and openness but what is encouraging is that there were no differences between other labor intensive and costly technologies and IVR on the other procedural justice factors. Therefore, there do not appear to be any major negatives in terms of structural fairness among alternative screening devices implying that organizations can make choices between screening methods based on other factors such as recruitment strategy or cost.

    Notes: Abstract The utilization of quinaldine (2-methylquinoline) by Arthrobacter sp. Rü61a proceeds via 1H-4-oxoquinaldine, 1H-3-hydroxy-4-oxoquinaldine, and N-acetyl-anthranilic acid. By analogy, 1H-4-oxoquinoline is degraded by Pseudomonas putida 33/1 via 1H-3-hydroxy-4-oxoquinoline and N-formylanthranilic acid. Using the purified enzymes from both organisms, the mode of N-heterocyclic ring cleavage was investigated. The conversions of 1H-3-hydroxy-4-oxoquinaldine and 1H-3-hydroxy-4-oxoquinoline to N-acetyl- and N-formylanthranilic acid, respectively, were both accompanied by the release of carbon monoxide. The enzyme-catalysed transformations were performed in an [18O]O2 atmosphere and resulted in the incorporation of two oxygen atoms into the respective products, N-acetyl- and N-formylanthranilic acid, indicating an oxygenolytic attack at C-2 and C-4 of both 1H-3-hydroxy-4-oxoquinaldine and 1H-3-hydroxy-4-oxoquinolone.

    Notes: Rhodopirellula baltica (strain SH 1T) is a free-living marine representative of the phylogenetically independent and environmentally relevant phylum Planctomycetes. Little is known about the regulatory strategies of free-living bacteria with large (7.15 Mb) genomes. Therefore, a consistent, quantitative and qualitative description was produced by comparing R. baltica's transcriptional regulator pool with that of 123 publicly available bacterial genomes. The overall results are congruous with earlier observations that in Bacteria, the proportion of genes encoding transcriptional regulators generally increases with genome size. However, R. baltica distinctly stands out from this trend with only 2.4% (174) of all genes predicted to encode transcriptional regulators. The qualitative investigation of R. baltica's transcriptional regulators revealed a clear shift towards high numbers of two-component systems (66) as well as high numbers of sigma factors (49), with more than 76% (37) belonging to the extra-cytoplasmic function subfamily of sigma-70. Only one predicted sigma factor showed a relatively close phylogenetic relationship to that of another bacterium, the sigma factor SigZ of Bacillus subtilis. In summary, analysis of the R. baltica genome revealed disparate regulatory mechanisms and a clear bias towards direct environmental sensing. This strategy might provide a selective advantage for organisms living in habitats with frequently changing environmental conditions.

    Notes: The paper describes a model and a simple experimental method to determine suspension transport properties, separating the role of particles from the suspending fluid. Shear stresses are described as consisting of 2 components: 1st, a shear stress caused by the flow of the suspending fluid and 2nd, a shear stress caused by direct interaction between suspended particles. We show that the determination of these shear stresses of a complex (food) suspension is largely independent of the definition of its suspending fluid. The rheological behavior of soy milks at low and intermediate moisture contents was studied, and results are compared with those obtained previously. The method evidences that the high viscosity of soy milks is predominantly caused by direct particle interaction.

    Notes: Sparse stations and serious measuring problems hamper analyses of climatic conditions in the Arctic. This paper presents a discussion of measuring problems in the Arctic and gives an overview of observed past and projected future climate variations in Svalbard and Jan Mayen. Novel analyses of temperature conditions during precipitation and trends in fractions of solid/liquid precipitation at the Arctic weather stations are also outlined. Analyses based on combined and homogenized series from the regular weather stations in the region indicate that the measured annual precipitation has increased by more than 2.5% per decade since the measurements started in the beginning of the 20th century. The annual temperature has increased in Svalbard and Jan Mayen during the latest decades, but the present level is still lower than in the 1930s. Downscaled scenarios for Svalbard Airport indicate a further increase in temperature and precipitation. Analyses based on observations of precipitation types at the regular weather stations demonstrate that the annual fraction of solid precipitation has decreased at all stations during the latest decades. The reduced fraction of solid precipitation implies that the undercatch of the precipitation gauges is reduced. Consequently, part of the observed increase in the annual precipitation is fictitious and is due to a larger part of the “true” precipitation being caught by the gauges. With continued warming in the region, this virtual increase will be measured in addition to an eventual real increase.

    Notes: Part of the lysine was destroyed when soybean meal or isolated soybean protein mixed with sucrose was autoclaved 4 hr at 121°C. Little or no loss occurred when isolated proteins were autoclaved by themselves. In vitro digestion with trypsin and erepsin liberated less lysine from the autoclaved than from the unheated soybean proteins if they did not contain the trypsin inhibitor.Partial hydrolysis with hydrochloric acid liberated more aspartie and glutamic acids and lysine from unheated than from heated soybean proteins.Hydrolysis with concentrated hydrochloric acid for 7 days at 40°C and in vitro digestion with trypsin and erepsin liberated similar amounts of lysine (except for the proteins containing trypsin inhibitors), but acid hydrolysis liberated more aspartic and glutamic acids that did enzymatic hydrolysis.The data support the hypothesis that autoclaving soybean protein formed lysine-aspartic acid and lysine-glutamic acid linkages that were resistant to mild hydrolysis.

    Notes: Free amino acid analyses were conducted on 24 muscle tissue samples of chicken, both young and old, fresh and aged, in an attempt to show some properties or constituents of the meat which might he related to quality factors, such as tenderness. In general, ammonia nitrogen remained fairly constant throughout the study. Storage resulted in increases in free amino acids, with proline being a major exception. Light meat showed less free amino acids than dark meat, with major exceptions being lysine and histidine. In most cases, broilers had more free amino acids than hens. Taurine concentration was much higher in the dark meat of both broilers and hens. No relation was found between tenderness and the general pattern of free amino acid concentration or between tenderness and the concentration of any single free amino acid.
