    Production and characterization of a plant α-hydroxynitrile lyase in Escherichia coli (1997)

    Notes: The coding sequence of the cyanogenic α-hydroxynitrile lyase gene of Manihot esculenta Crantz (cassava) was cloned in the plasmid vector pMal-c2 and expressed in Escherichia coli strain JM105. DNA sequencing showed that the recombinant plasmid contained the same sequence as the cDNA clone pHNL10. Peptide sequencing of the recombinant protein showed that the N-terminus was heterogeneous, with either four or six additional amino acid residues compared with the native protein. Circular dichroism spectra indicated similar secondary structure contents for both proteins. Enzyme assays showed that specific activity of native and recombinant proteins were 0.24 and 0.26 mmol CN-/mg/min, respectively; that both proteins had optimal activity at 40°C and pH 5.5; and that both proteins were inhibited by the serine protease inhibitor phenyl-methane sulfonyl flouride (PMSF). Isoelectric focusing of native and recombinant protein revealed multiple isoforms for both proteins; the recombinant protein had a more basic mean isoelectric point (pl) (5.1) than the native protein (4.5). © 1997 John Wiley & Sons, Inc.     Notes: Mathematical programming approaches to the choice of capital expenditure projects under conditions of funds constraint have been widely advocated in the literature of finance. The present paper takes exception to such suggestions, and concludes that programming is not only excessively costly and complex for most practical applications, but is frequently incorrect in the decisions it implies. A technique for allocating funds via an adjusted Excess Present Value Index is developed and proposed as a superior analytical framework.

    Notes: Abstract: A knowledge base management system (KBMS) realises a combination of techniques found in database management systems and knowledge-based systems. At the data model and knowledge representation level, many systems of this kind constitute a marriage of the relational data model and the rule-based reasoning. Experience has shown that either approach is restricted in the way it can express the demanding information and knowledge structures required for applications like decision support systems. Two new technologies offer an exciting new integrated approach to knowledge management. Object-oriented database management systems (OODBMS) provide an object model that supports powerful abstraction mechanisms to facilitate the modelling of highly structured information. Whereas case-based reasoning (CBR) systems are knowledge bases which organise their capabilities around a memory of past cases and the notion of similarity. Both types of system are built upon two fundamental concepts: 1) the retrieval of entities with potentially complex structure, called objects in the former, and cases in the latter type of system; 2) the organisation of those entities in collections with common characteristics. In an OODBMS such collections are termed extents, and in CBR they are usually called categories. In either system, the conceptual meta notion to represent both, objects as well as extents, and cases as well as categories, is the class.Revolving around a Conceptual Case Class and extending a standard object model, this paper proposes a novel and general approach to represent case-knowledge and to build KBMSs. The work presented here is a spin-off of the design of an object query language within the ESPRIT project Lynx.

    Notes: For trade union bargainers one of the infallible laws of political economy is that wages lag behind prices. This means that in an inflationary era the real value of the wage to which their members are entitled will be eroded, and some compensation will have to be made for this at the next round of wage negotiations. Therefore, changes in the cost of living since the last settlement will be regarded by trade unions as one of the most important factors to be taken into account in arriving at a new settlement. This is implicit in the statement made by the TUC in 1971 that “it requires pay increases of at least 10 per cent simply to restore the real disposable pay of a union's previous settlement 12 months earlier”. Economists, on the other hand, argue that once an inflation is under way, then economic agents will develop an inflationary psychology—that is, they will expect it to continue and will adjust their behaviour accordingly. If this is correct, trade union bargainers will attempt to anticipate inflation by trying to fix the money wage at a higher level than they would aim for if the price level were more stable.
