    Notes: Biosynthesis of carotenoids and steroids in tissue cultures of carrot cells I Daucus carota L.) was studied by measuring the change in their contents and incorporation of 14lC-acelate. The rate of synthesis of these plant products varied significantly during the growth cycle in batch culture. Carotenoids were most actively synthesized in the early logarithmic phase and the synthetic rate sharply declined as the culture aged, whereas the rate of accumulation of phytosterols was highest at the end of the growth phase. Newly synthesized carotenes were shown to undergo turnover during the growth. The synthesis of both carotecnoid and steroid was stimulated by 2.- 4-dichlorophenoxyacetic acid.

    Description / Table of Contents: Zusammenfassung Ein Verfahren zur Mikrobestimmung von Fluorid und Oxalat wurde vorgeschlagen, das auf deren Fällungsaustauschreaktion mit Lanthan-Chloranilat beruht. Diese Austauschreaktion wurde mit der Verwendung von Adsorptionszonen in der Ringofenmethode kombiniert. Das in einer Tonerde-Magnesiumhydroxid-Zone adsorbierte Chloranilat wurde ring-kolorimetrisch bestimmt. Die Erfassungsgrenze für Fluorid und Oxalat beträgt 0,75μg, Konzentrationen herunter bis zu 0,4μg/μl sind noch bestimmbar. Die Methode hat den Vorteil, daß eine thermolabile Substanz isoliert und bestimmt werden kann, ohne beim Auswaschen in die Adsorptionszone Schaden zu nehmen.
    Notes: Summary A method for the microdetermination of fluoride and oxalate based on their precipitate-exchange reaction with lanthanum chloranilate was presented. This exchange reaction was combined with adsorption barrier technique in the ring oven method. The chloranilate adsorbed on aluminum-magnesium hydroxide zone was determined by ring colorimetry. The detection limit for fluoride and oxalate was 0.75μg, while fluoride and oxalate concentrations down to 0.4μg/μl have been determined. The method has the advantage that a thermally labile substance can be isolated and determined without thermal damage by washing to the adsorption barrier zone.

    Description / Table of Contents: Zusammenfassung Nach Isolierung aus dem Fruchtfleisch der Dattel wurde ein Gemisch kristallisierter pflanzlicher Sterine der stufenweisen Extraktion und Säulenchromatographie unterworfen. Dessen Zusammensetzung wurde gaschromatographisch untersucht. Außerdem wurde dieses Steringemisch gaschromatographisch-massenspektrographisch nach Überführung in die entsprechenden Trimethylsilylderivate identifiziert. Cholesterin, Campesterin, Stigmasterin,β-Sitosterin und Isofucosterin wurden durch Nachweis der Molekularionen, durch Zuordnung der Fragmentionen und durch Vergleich mit dem spektralen Verhalten der betreffenden authentischen Verbindung festgestellt. Das Steringemisch wurde dann im Wege der Hochleistungs-Flüssigchromatographie untersucht. Hierzu diente eine „Zorbax ods“-Säule und ein Lösungsmittelgemisch aus Acetonitril—Wasser—0,1M Natriumacetat (90 ∶ 10 ∶ 1). Die Peaks wurden durch Lichtstrahlen von 210 nm nachgewiesen. Sechs Peaks wurden gefunden. Nach Identifizierung durch Gaschromatographie und IR-Spektroskopie wurde jede einzelne Verbindung gesammelt. Neben einer unbekannten Zacke entsprachen die Peaks der Reihe nach dem Isofucosterin, dem Cholesterin, Stigmasterin+Campesterin, Campesterin undβ-Sitosterin. Das Gemisch der pflanzlichen Sterine wurde nur mit phasenverkehrten Säulen analysiert, während Corticosteroidhormone sowie männliche und weibliche Steroidhormone sowohl auf normalen wie auf phasenverkehrten Säulenpackungen getrennt wurden.
    Notes: Summary Isolation of a crystalline plant sterol mixture from the sarcocarp of the date was followed by successive extraction and column chromatography. The composition of the mixture was examined by gas chromatography. An additional gas chromatographic-mass spectroscopic identification of the sterol mixture was carried out after conversion of the sample into its trimethylsilyl derivates. Cholesterol, campesterol, stigmasterol,β-sitosterol and isofucosterol were found from the identification of molecular ions, the assignment of fragment ions and the comparison of each spectral feature with that of the authentic compound. The sterol mixture was then examined by high-performance liquid chromatography employing a “Zorbax ODS” column and an acetonitrile-water-0.1M sodium acetate (90∶10∶1) solvent system, and the peaks were detected with radiation at 210 nm. Six peaks were found on the chromatogram. The identification of each peak by means of gas chromatography and infrared spectroscopy was followed by the collection of each component. The peaks were an unknown (ghost) peak, isofucosterol, cholesterol, stigmasterol + campesterol, campesterol andβ-sitosterol, successively. The plant sterol mixture has only been analysed on reverse-phase column packings whereas the hormonal corticosteroids and male or female hormonal steroids have been separated on both normal and reverse-phase column packings.

    Notes: Summary A justification of a finite element scheme for plate bending problems is presented. The finite element treated here is the triangular element proposed by Stricklinet al. and Dhatt using the discrete Kirchhoff assumption. Some error estimates are derived for the finite element solutions of static and eigenvalue problems with the homogeneous Dirichlet conditions. Numerical experiments are also performed to see the validity of the theory.

    Notes: Summary Paternal age distribution for 1279 cases of Down's syndrome born in 1952–1968 was compared with the corresponding distribution for the general population, corrected for the maternal age as well as for the year of birth of the patients. Although there was no difference in the mean paternal age, the two distributions differed significantly, largely due to the excess of fathers aged 55 years and over and to the deficit of those aged 40–44 years in the patients born to mothers aged 30 years and over. The overall pattern of the relative incidence of Down's syndrome with advancing paternal age, with maternal age controlled, seems consistent with the hypothesis proposed by Stene et al. (1977). It increased from 0.8 for fathers aged 20–24 years slowly up to 1.2 for those aged 45–49 years, though with an intermediate drop to 0.8 at the age of 40–44 years, and then sharply to 2.4 for those aged 55 years and over. This rising pattern of the relative incidence with paternal age was essentially the same for the patients born in 1952–1960 and for those born in 1961–1968, although the slope was less steep in the latter than in the former group.

    Notes: Summary A prenatal diagnosis of GM1-gangliosidosis was made in a pregnancy at risk, on the basis of a deficiency of β-galactosidase activity demonstrated in cultured aminiotic fluid cells. Biochemical analyses were performed in the aborted fetus. GM1-ganglioside β-galactosidase activity was reduced to 1% of the control value in both the brain and liver of the affected fetus. Lamellar bodies suggestive of membranous cytoplasmic bodies were found in cells of basal ganglions, while the accumulation of GM1-ganglioside in the brain was not remarkable.
