    Notes: Abstract. Fish surviving infection with the pathogenic ciliated protozoan, Ichthyophthirius multifiliis (Fouquet, 1876), become resistant to subsequent infection by the parasite. The acquired immunity suggests that development of a vaccine against the parasite may be possible. Because of the advantages of immunoprophylaxis for treatment of the disease, an effort has been made to determine whether fish exposed to killed parasite preparations can resist subsequent lethal challenge. Both the route of administration and the effects of stage specific antigens have been examined. Channel catfish vaccinated by intraperitoneal (i.p.) injection or bath immersion with killed I multifiliis tomites show 100% mortality following a standard challenge protocol. Similarly, 100% mortality was observed in test groups injected with tomite cilia. In both cases, a consistent difference in days to death between control and test group animals was observed. Although complete mortality was seen with fish injected with tomite preparations, fish vaccinated with killed trophonts (the feeding stage of the parasite) had a much greater degree of protection with approximately 50% of fish surviving an otherwise lethal challenge. Finally, animals injected intraperitoneally with live tomites showed nearly complete immunity and were identical in their response to fish which survive natural infection. The response of fish vaccinated with live parasites indicates that animals injected intraperitoneally can develop surface immunity and that i.p. injection is a suitable route of administration for potential I. multifiliis vaccines.

    Notes: SYNOPSIS. The morphology of 4 genera of the family Trypanosomatidae has been studied with particular reference to the contractile vacuole, reservoir, flagellum, undulating membrane, kinetoplast, and nucleus. Both the contractile vacuole and reservoir have been found in all species examined. Four morphological types of culture forms are described with special reference to these structures: (1), the Crithidia fasciculata type which is short and membraneless with a rounded posterior end, truncate anterior end and a well developed reservoir; (2) the Herpetomonas muscarum type with no undulating membrane, a truncate posterior end, a rounded anterior end and a reservoir which varies in length from about 1/6 of the length of the body to the full length of the body; (3) the Leishmania type with no undulating membrane, a pointed posterior end, an asymmetrical anterior end and a short reservoir which remains constant in size; and (4), the short-membraned type exemplified by Trypanosoma cruzi, with a pointed posterior end, a short reservoir near the middle of the body and the flagellum attached to the body along a portion of its length. This last form is derived from the blood stream morphology as exemplified by Trypanosoma lewisi in the rat by a shifting of the kinetoplast, flagellum, reservoir and contractile vacuole along the surface of the body from a position posterior to a position anterior to the nucleus.

    Notes: Sera from fish that survive infections with the ciliated protozoon, Ichthyophthirius multifiliis, immobilize the parasite in vitro. In order to identify ceil surface antigens involved in the immobilization response, integral membrane proteins were extracted from tomites with Triton X-l14 and used to immunize rabbits. The rabbit antisera immobilized the parasite in vitro and antigens were localized to cell and ciliary plasma membranes by indirect immunofluorescent microscopy. The membrane protein fractions from both whole cells and tomite cilia were characterized by 1 - and 2-dimensiona! SDS-PAGE. A 43,000-dalton (D) glycoprotein with an isoelectric point of 7.0 is the predominant protein in these fractions, comprising 12% and 60% of the total protein of whole cell and ciliary membranes, respectively. Western blot analysis of ciliary proteins with immune rabbit sera indicated that the 43,000-D glycoprotein is the principal antigen.

    Notes: Recent interest in the constant Kmax (Kcmax) threshold testing procedure has resulted in a more in-depth study of the influence of Kmax level on fatigue response and ΔKth in aluminium alloys. Under Rc= 0.1 conditions, which cause large amounts of closure, ΔKth levels were typically 2 to 4 Mpam. However, under Kcmax test procedures, associated with no measurable closure at threshold, ΔKth was typically 1 Mpam. A slight Kcmax level effect on ΔKth was observed at high Kmax values for some of the alloys, and was deemed to be a pure mean stress effect, separate from closure arguments.

    Notes: An organism diversity index for use with mixed liquor or wastewater samples was developed to assess the species richness and diversity of activated sludge. The index was used in several studies and was found to be fast and simple to perform using basic laboratory equipment. Two bench-scale and two pilot-scale studies found that the resulting ‘organism diversity index’value was a good indicator of process performance and was not influenced by the total concentration of the mixed-liquor suspended solids but by their nature alone. The technique rapidly yielded pertinent information about the health of the sludge and could be used instead of genetic investigations to obtain population information quickly enough for wastewater-treatment plant process control.

    Notes: Sera from channel catfish rendered immune to the protozoan pathogen Ichthyophthirius multifiliis were screened for activity against live parasites. Cells in the infective stage (tomites) were incubated in doubling dilutions of immune and pre-immune sera from fish that had been immunized by exposure to sublethal infections. When examined by light microscopy, tomites were found to agglutinate in the presence of immune sera. While the stength of individual sera varied, agglutination of cells occurred at dilutions as high as 1:128. Cells showed little tendency to agglutinate in pre-immune sera, and virtually no effects were seen with dilutions of pre-immune sera greater than 1:16. Agglutination was usually accompanied by release of mucus from cells, and while tomites appeared to be immobilized, their cilia continued to beat. Low dilutions of immune sera appeared to be toxic. Similar effects on tomites were seen with rabbit antisera prepared against Ichthyophthirius cilia. The involvement of humoral antibodies in agglutination and protective immunity is discussed.

    Description: Phase variation (random ON/OFF switching) of gene expression is a common feature of host-adapted pathogenic bacteria. Phase variably expressed N 6 -adenine DNA methyltransferases (Mod) alter global methylation patterns resulting in changes in gene expression. These systems constitute phase variable regulons called phasevarions. Neisseria meningitidis phasevarions regulate genes including virulence factors and vaccine candidates, and alter phenotypes including antibiotic resistance. The target site recognized by these Type III N 6 -adenine DNA methyltransferases is not known. Single molecule, real-time (SMRT) methylome analysis was used to identify the recognition site for three key N. meningitidis methyltransferases: ModA11 (exemplified by M.NmeMC58I) (5'-CGY m6 A G-3'), ModA12 (exemplified by M.Nme77I, M.Nme18I and M.Nme579II) (5'-AC m6 A CC-3') and ModD1 (exemplified by M.Nme579I) (5'-CC m6 A GC-3'). Restriction inhibition assays and mutagenesis confirmed the SMRT methylome analysis. The ModA11 site is complex and atypical and is dependent on the type of pyrimidine at the central position, in combination with the bases flanking the core recognition sequence 5'-CGY m6 A G-3'. The observed efficiency of methylation in the modA11 strain (MC58) genome ranged from 4.6% at 5'-GCGC m6 A GG-3' sites, to 100% at 5'-ACGT m6 A GG-3' sites. Analysis of the distribution of modified sites in the respective genomes shows many cases of association with intergenic regions of genes with altered expression due to phasevarion switching.

    Description: We developed an innovative hybrid sequencing approach, IDP-fusion, to detect fusion genes, determine fusion sites and identify and quantify fusion isoforms. IDP-fusion is the first method to study gene fusion events by integrating Third Generation Sequencing long reads and Second Generation Sequencing short reads. We applied IDP-fusion to PacBio data and Illumina data from the MCF-7 breast cancer cells. Compared with the existing tools, IDP-fusion detects fusion genes at higher precision and a very low false positive rate. The results show that IDP-fusion will be useful for unraveling the complexity of multiple fusion splices and fusion isoforms within tumorigenesis-relevant fusion genes.
