    Description: Hurricane activity in the Northeastern Gulf of Mexico and its relationship to regional and large-scale climate variability during the Late Holocene is explored. A 4500-year record of hurricane-induced storm surges is developed from sediment cores collected from a coastal sinkhole near Apalachee Bay, Florida. Reconstructed hurricane frequency is shown to exhibit statistically significant variability with the greatest activity occurring between 2700 and 2400 years ago and the least activity between 1900 to 1600 years ago and after 600 years ago. Proxy records of stormrelevant climate variables contain similar timescales of variability and suggest both regional and large-scale mechanisms have influenced hurricane activity on centennial to millennial timescales. In particular, low-frequency migrations of the Loop Current may exercise control over regional hurricane activity by changing the thermal structure of the upper ocean and influencing the role of storm-induced upwelling on hurricane intensification. A new method for estimating the frequency of hurricanegenerated storm surges is presented and applied to Apalachee Bay, Florida. Multisite paleohurricane reconstructions from this region are developed, and the effects of geographic boundary conditions and temporal resolution on estimates of paleohurricane frequency are explored.

    Notes: A new, highly specific colorimetric DNA hybridization method (cDNAH) for detection of Salmonella in foods was developed. It detected 355/362 strains representing 206/213 serovars of Salmonella, and showed no cross-reactivity with 32 different non-Salmonella species, including a variety of Enterobacter and Citrobacter species. The cDNAH method was compared with the BAM/AOAC culture method in two studies: (1) with 20 different inoculated and uninoculated foods overall agreement between the two methods was 99%. (2) with naturally contaminated foods at two independent sites agreement with culture was 99.8% where 9/404 samples were positive for Salmonella.

    Notes: A region of the IncHl plasmid R27 has been found to share very close nucleotide sequence homology with the RepFIA replicon of F. This region has been located on a 1.6 kb segment of R27 plasmid DNA, and corresponds to ori-2 and the E gene of F. The IncC repeat sequence region shows reduced homology, with the F repeats being an imperfect subset of a larger repeated sequence found in R27. The E gene homologue of R27 is able to initiate replication from the F ori-2 sequence and to repress the E gene promoter of F. The results are consistent with the observed incompatibility behaviour of R27, and have a bearing on the specificity of interaction of E protein with its DNA-binding sites.

    Notes: As the control of Oxalis pes-caprae L. is dependent upon treatment after the parent bulb has become exhausted and before the new bulbs have formed, experiments were conducted to test the uniformity of development of plants and to determine if adequate control could be expected from a single herbicide application. Inherent differences were observed in the time of emergence, and further differences could be induced by increasing the depth of planting or withholding water. Competition between plants was shown to retard the development of late-emerging plants.The physical removal of parts of the plant before bulb exhaustion had limited effect on bulb production, but reduced bulb weights and numbers when carried out after bulb exhaustion. Although not all parts of the plant were replaced by regrowth, no major changes in overall development were noted.

    Notes: A survey of the composition and structure of the vegetation of the roadside easement and adjacent farmland was made at eighteen sites on the Mornington Peninsula. Victoria, Australia. The ground cover was grass dominant at all sites, with a predominance of introduced species notably Holcus lanatus, Anthoxanthum odoratum and Briza maxima. Agrostis tenuis and Paspalum dilatatum occurred to a lesser extent. Two alien broad-leafed species, Hypochoeris radicata and Plantago lanceolata, were abundant at most sites.The frequency of species present on the easement was markedly reduced in the adjacent farmland; conversely, the pasture plants Lolium perenne, Trifolium repens and T. subterraneum, did not colonize the easement to any great extent.At some sites, the native trees and shrubs had not been cleared during road construction. They were able to survive on the easement despite the influences of the adjacent road and farmland. Seedlings of all the native species were found.

    Notes: The SopA protein plays an essential, though so far undefined, role in partition of the mini-F plasmid but, when overproduced, it causes loss of mini-F from growing cells. Our investigation of this phenomenon has revealed that excess SopA protein reduces the linking number of mini-F. It appears to do so by disturbing the partition complex, in which SopB normally introduces local positive supercoiling upon binding to the sopC centromere, as it occurs only in plasmids carrying sopC and in the presence of SopB protein. SopA-induced reduction in linking number is not associated with altered sop promoter activity or levels of SopB protein and occurs in the absence of changes in overall supercoil density. SopA protein mutated in the ATPase nucleotide-binding site (K120Q) or lacking the presumed SopB interaction domain does not induce the reduction in linking number, suggesting that excess SopA disrupts the partition complex by interacting with SopB to remove positive supercoils in an ATP-dependent manner. Destabilization of mini-F also depends on sopC and SopB, but the K120Q mutant retains some capacity for destabilizing mini-F. SopA-induced destabilization thus appears to be complex and may involve more than one SopA activity. The results are interpreted in terms of a regulatory role for SopA in the oligomerization of SopB dimers bound to the centromere.
