    Notes: SUMMARY-The development of food mixtures containing vegetable protein concentrates, minerals, and vitamins represents an important approach to the problem of protein-calorie malnutrition of preschool children in many parts of the world. In Brazil, soy flour is the most readily available good-quality protein for this purpose. Protein efficiency-ratio studies in rats demonstrated that soy flour with a PER value of 1.99 was available in large quantities. Accordingly, a formula containing 19.50% soy flour, 4.50% nonfat milk powder, 32.28% corn starch, 39.00% sucrose, and 4.72% vitamins and minerals was thoroughly tested as an infant food. The PER value of the protein in rats was 2.31. Amino acid assay demonstrated that the protein compared favorably with the essential amino acid pattern of the FAO/WHO reference protein (FAO/WHO, 196.5) except for methionine-cystine, The formula was well accepted and tolerated by children and adults. Average nitrogen absorption and retention of the formula as the sole food of malnourished children were 72.68 and 23.82%; values for milk protein were 80.17 and 30.13%. Average nitrogen absorption and retention were 63.4 and 22.0% when the formula provided one-half (15 g) of the daily protein for normal children consuming a typical diet of the region, comparing favorably with values of 69.3 and 21.3% for milk protein.

    Notes: Stress factors studied included 0.95 aw adjusted with sucrose and glycerol, pH (4.0, 3.5, and 3.0), potassium sorbate (0, 500, 1000, and 1500 ppm) and sodium bisulfite (0, 100, and 150 ppm). These were tested on a strain of Zygosaccharomyces rouxii isolated from an intermediate moisture (IM) papaya product. Z rouxii was very sensitive to SO2. A combination of 100 ppm SO2, 500 ppm sorbate and pH 4.0 is recommended to prevent spoilage of IM fruits by Z. rouxii. The minimum aw for growth in sucrose-glycerol broth was between 0.62 and 0.64 in the absence of antimicrobials and in the range of optimal pH for this yeast.

    Notes: The galE gene of Pasteurella multocida has been isolated by complementing galE-defective mutants of Salmonella typhimurium with a plasmid library of this organism. The complete nucleotide sequence of the P. multocida galE gene consists of 1017 nucleotides, encoding a predicted polypeptide of 339 amino acids. The deduced amino acid sequence displayed the highest identity (85%) to the GalE protein of Haemophilus influenzae. However, the gene organization surrounding the galE locus was different from that of H. influenzae. A galE-defective mutant of P. multocida was obtained by replacement of the active galE gene by a copy inactivated in vitro. The resulting galE mutant was highly attenuated as seen in a biological test carried out in a mouse model.

    Notes: . Growth by serial transfers of the trypanosomatid Crithidia deanei in culture medium containing 1 mg/ml of the β-lactam antibiotics ampicillin or cephalexin resulted in shape distortion of its endosymbiont. The endosymbiont first appeared as filamentous structures with restricted areas of membrane damage. An increase of electron lucid areas was also observed in the endosymbiont matrix. The continuous treatment with β-lactam antibiotics, resulted in endosymbiont membranes fragmentation; and later on the space previously occupied by the symbiont was identified as an electron lucid area in the host cytoplasm. The putative targets of β-lactam antibiotic were two membrane-bound penicillin-binding proteins (PBPs) detected in the Sarkosyl-soluble fraction of purified symbionts labeled with [3H]-benzylpenicillin. The apparent molecular weight of the proteins were 90 kDa (PBP1) and 45 kDa (PBP2). PBP2 represented 85% of the total PBP content in the membrane fraction of the endosymbionts. Competition experiments using the tested antibiotics and [3H]-benzylpenicillin showed that ampicillin and cephalexin have half saturating concentrations considerably higher than [3H]-benzylpenicillin and indicated that PBP1 is the probable lethal target of the antibiotics tested. These results suggest that a physiologically active PBP is present in the cell envelope of C. deanei endosymbionts and may play important roles in the control of processes such as cell division and shape determination.

    Notes: Abstract Hydroxylamine is an intermediate in the oxidation of ammonia to nitrite, but until now it has not been possible to grow Nitrosomonas europaea on hydroxylamine. This study demonstrates that cells of N. europaea are capable of growing mixotrophically on ammonia and hydroxylamine. The molar growth yield on hydroxylamine (4.74 g mol−1 at a growth rate of 0.03 h−1) was higher than expected. Aerobically growing cells of N. europaea oxidized ammonia to nitrite with little loss of inorganic nitrogen, while significant inorganic nitrogen losses occurred when cells were growing mixotrophically on ammonia and hydroxylamine. In the absence of oxygen, hydroxylamine was oxidized with nitrite as electron acceptor, while nitrous oxide was produced. Anaerobic growth of N. europaea on ammonium, hydroxylamine and nitrite could not be observed at growth rates of 0.03 h−1 and 0.01 h−1.

    Notes: Abstract β-d-Xylosidase production was maximal for Humicola grisea var. thermoidea grown on xylan as the sole carbon source. The main β-d-xylosidase activity was localised in the periplasm. β-Xylosidase was purified from crude extracts by heat treatment, ammonium sulfate precipitation and chromatography on DEAE-cellulose and Sephadex G-100. The purified enzyme was a monomer of molecular mass estimated to be 43 kDa by SDS-PAGE and gel filtration. Optima of pH and temperature were 6.0 and 50 °C, respectively. The enzyme activity was stimulated by Ca2+, Fe2+, and Mg2+. The purified β-xylosidase did not exhibit xylanase, carboxymethylcelullase, galactosidase, glucosidase, fucosidase or arabinosidase activities. The purified β-xylosidase hydrolysed xylobiose and xylo-oligosaccharides of up to five monosaccharide units. The enzyme had a Km of 0.49 mM for p-nitrophenyl-β-d-xylopyranoside and was not inhibited by its product, xylose.

    Notes: Abstract Leishmania (Leishmania) infantum is the causative agent of both the cutaneous and visceral forms of leishmaniasis in southwest Europe; the dog is the main reservoir. In order to identify the L. (L.) infantum zymodemes present in Spain, a total number of 85 Leishmania stocks isolated from dogs (31), HIV-positive patients (46) with visceral or cutaneous leishmaniasis, a patient with visceral leishmaniasis complicating renal transplantation (1) and immunocompetent patients (7) with visceral or cutaneous leishmaniasis, have been characterized by isoenzyme typing. All canine stocks were MON-1, which is the most widespread zymodeme in the Mediterranean area. In immunocompetent patients three zymodemes were found: MON-1 (2), MON-24 (2) and MON-34 (3). Nine different zymodemes were obtained in stocks from HTV co-infected patients, indicating a higher variability of L. (L.) infantum amongst them: MON-1 (in 21 stocks), MON-24 (7), MON-28 (1), MON-29 (3), MON-33 (7), MON-34 (1) and MON-183 (4). Two new zymodemes, MON-198 (1) and MON-199 (1), were described among HIV patients from Spain. The stock from the renal transplanted patient was MON-1.     Notes: Abstract The major outer membrane proteins (OprF) from Pseudomonas fluorescens MFO and OE 28.3 were purified by a new method involving native electrophoresis in octyl-polyoxyethylene media. Both proteins, characterized by the same size, heat-modifiability and N-terminal sequence were re-incorporated in virtually solvent-free planar lipid bilayers. They displayed very similar channel-forming properties: the major conductance level was between 250 pS and 270 pS in l M NaCl. From experiments of zero-current potential, both porins were determined weakly cation selective. Amplification by PCR and sequencing of the oprF gene of strain MFO allowed to point out 94% identity between the amino acid sequences of these two OprFs isolated from ecological niches as different as milk (strain MFO) and soil (strain OE 28.3).

    Notes: —Commercial samples of bleached and normal paprika were extracted with acetone, separated by countercurrent distribution fractionation, purified by column chromatography and identified by spectral and chemical methods. The bleaching treatment of the paprika samples resulted in a loss of nearly 96% in total extractable pigment expressed as beta-carotene. The fractionation of the cartenoid extracts revealed a great variation in the distribution patterns of the pigments. Most grades of paprika showed a predominace of diolpolyol carotenoids, but one grade had a far greater amount of hydrocarbons. There were 54 and 37 pigments isolated from bleached and unbleached Domestic paprikas, respectively. Of the 54 and 37 pigments, only 33 and 21, respectively, were completely or tentatively identified. Sixteen of the known pigments in unbleached paprika were also observed in the bleached sample. It is evident that the bleached paprika gave more isomers and oxicative products than did the unbleached sample. Most of the compounds appearing with the capsanthin and capsourbin fractions seemed to be isomers of more fully oxygenated derivatives of these two major pigments. Most of the more oxygenated products appeared to possess ketonic group(s).

Pre-fractionation of the complex essencew ontaining components in widely differing concentrations—was performed on a packed column previous to further analysis on the capillary column. A large number of esters, aldehydes, acetals, and alcohols were identified.
