    Description: Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder primarily affecting motor neurons. Mutations in optineurin cause a small proportion of familial ALS cases, and wild-type (WT) optineurin is misfolded and forms inclusions in sporadic ALS patient motor neurons. However, it is unknown how optineurin mutation or misfolding leads to ALS. Optineurin acts an adaptor protein connecting the molecular motor myosin VI to secretory vesicles and autophagosomes. Here, we demonstrate that ALS-linked mutations p.Q398X and p.E478G disrupt the association of optineurin with myosin VI, leading to an abnormal diffuse cytoplasmic distribution, inhibition of secretory protein trafficking, endoplasmic reticulum (ER) stress and Golgi fragmentation in motor neuron-like NSC-34 cells. We also provide further insight into the role of optineurin as an autophagy receptor. WT optineurin associated with lysosomes and promoted autophagosome fusion to lysosomes in neuronal cells, implying that it mediates trafficking of lysosomes during autophagy in association with myosin VI. However, either expression of ALS mutant optineurin or small interfering RNA-mediated knockdown of endogenous optineurin blocked lysosome fusion to autophagosomes, resulting in autophagosome accumulation. Together these results indicate that ALS-linked mutations in optineurin disrupt myosin VI-mediated intracellular trafficking processes. In addition, in control human patient tissues, optineurin displayed its normal vesicular localization, but in sporadic ALS patient tissues, vesicles were present in a significantly decreased proportion of motor neurons. Optineurin binding to myosin VI was also decreased in tissue lysates from sporadic ALS spinal cords. This study therefore links several previously described pathological mechanisms in ALS, including defects in autophagy, fragmentation of the Golgi and induction of ER stress, to disruption of optineurin function. These findings also indicate that optineurin–myosin VI dysfunction is a common feature of both sporadic and familial ALS.

    Description: The goal of the Papillomavirus Episteme (PaVE) is to provide an integrated resource for the analysis of papillomavirus (PV) genome sequences and related information. The PaVE is a freely accessible, web-based tool ( http://pave.niaid.nih.gov ) created around a relational database, which enables storage, analysis and exchange of sequence information. From a design perspective, the PaVE adopts an Open Source software approach and stresses the integration and reuse of existing tools. Reference PV genome sequences have been extracted from publicly available databases and reannotated using a custom-created tool. To date, the PaVE contains 241 annotated PV genomes, 2245 genes and regions, 2004 protein sequences and 47 protein structures, which users can explore, analyze or download. The PaVE provides scientists with the data and tools needed to accelerate scientific progress for the study and treatment of diseases caused by PVs.

    Description: Next generation genomic technologies have made a significant contribution to the understanding of the genetic architecture of human neurodevelopmental disorders. Copy number variants (CNVs) play an important role in the genetics of intellectual disability (ID). For many CNVs, and copy number gains in particular, the responsible dosage-sensitive gene(s) have been hard to identify. We have collected 18 different interstitial microduplications and 1 microtriplication of Xq25. There were 15 affected individuals from 6 different families and 13 singleton cases, 28 affected males in total. The critical overlapping region involved the STAG2 gene, which codes for a subunit of the cohesin complex that regulates cohesion of sister chromatids and gene transcription. We demonstrate that STAG2 is the dosage-sensitive gene within these CNVs, as gains of STAG2 mRNA and protein dysregulate disease-relevant neuronal gene networks in cells derived from affected individuals. We also show that STAG2 gains result in increased expression of OPHN1 , a known X-chromosome ID gene. Overall, we define a novel cohesinopathy due to copy number gain of Xq25 and STAG2 in particular.

    Description: We investigate the enhancement of heat transfer in the nucleate boiling regime by inducing high frequency acoustic waves ( f ∼ 10 6 Hz) on the heated surface. In the experiments, liquid droplets (deionized water) are dispensed directly onto a heated, vibrating substrate. At lower vibration amplitudes ( ξ s ∼ 10 −9 m), the improved heat transfer is mainly due to the detachment of vapor bubbles from the heated surface and the induced thermal mixing. Upon increasing the vibration amplitude ( ξ s ∼ 10 −8 m), the heat transfer becomes more substantial due to the rapid bursting of vapor bubbles happening at the liquid-air interface as a consequence of capillary waves travelling in the thin liquid film between the vapor bubble and the air. Further increases then lead to rapid atomization that continues to enhance the heat transfer. An acoustic wave displacement amplitude on the order of 10 −8 m with 10 6 Hz order frequencies is observed to produce an improvement of up to 50% reduction in the surface temperature over the case without acoustic excitation.

    Description: First order reversal curves (FORC) method has been reported to be an efficient tool to study interaction between grains and layers of magnetic materials. Although a few studies have been carried out on perpendicular recording media in the past, a study on the effect of systematic variation of exchange interaction in granular perpendicular magnetic recording media on FORC contours has not been carried out in detail. Such a study will help to understand the use of FORC better. In this paper, we have made a systematic set of samples in order to study the variation in exchange coupling and its effect on FORC contours. The pressure during the deposition of the second ruthenium layer and the magnetic layer was varied to alter the separation between the grains and hence the exchange interaction between the grains in the CoCrPt-oxide recording layer. In addition, the thickness of Co-alloy cap layer was used as an additional tool to control the exchange interaction between the magnetic grains. The results indicated that the interaction field obtained from the FORC does not vary in a significant manner when the changes in exchange interaction are small. In comparison, the peak intensity of the FORC shows a clear trend as the exchange coupling is varied, making it a more suitable parameter to study the exchange and magnetostatic interactions in systems such as magnetic recording media.

    Description: Class-D audio amplifiers convert an audio signal into a high-frequency square wave whose switching times are modulated according to that audio signal. Thus, they operate using so-called pulse-width modulation. When the high-frequency components of the output square wave are removed by filtering, the remaining lower frequency components can reproduce the audio signal with very little distortion. This low distortion, coupled to the high efficiency of class-D amplifiers, is responsible for the increasing use of these devices in a wide variety of applications. However, to date there has been very little predictive mathematical modelling of those class-D amplifiers that employ negative feedback, as is usually the case in practice. Here, we derive and investigate a mathematical model for a novel design of the output stage for a class-D audio amplifier, which employs negative feedback to remove the effects of power supply noise on the amplifier's performance.

    Description: :  Biomedical Entity-Relationship eXplorer (BEReX) is a new biomedical knowledge integration, search and exploration tool. BEReX integrates eight popular databases (STRING, DrugBank, KEGG, PhamGKB, BioGRID, GO, HPRD and MSigDB) and delineates an integrated network by combining the information available from these databases. Users search the integrated network by entering key words, and BEReX returns a sub-network matching the key words. The resulting graph can be explored interactively. BEReX allows users to find the shortest paths between two remote nodes, find the most relevant drugs, diseases, pathways and so on related to the current network, expand the network by particular types of entities and relations and modify the network by removing or adding selected nodes. BEReX is implemented as a standalone Java application.     Notes: We have performed the first experiments to examine the neutral fragmentation paths following direct core-level excitation in a molecule. Using monochromatized synchrotron radiation in the range 100–140 eV, we have monitored the dispersed UV/optical fluorescence resulting from excitation of a Si 2p electron in SiF4. The main features in the fluorescence spectrum have been identified as emission from the SiF+4 D state and from excited SiF, Si, F, and Si+. Features in the fluorescence excitation spectra are assigned to excitation of a Si 2p electron to unoccupied valence orbitals, Rydberg orbitals, and shape resonances. There is a large enhancement in the yield of excited-state fragments following core-to-Rydberg excitation, which is due to the greater probability of the core-excited Rydberg state decaying, via a resonant Auger process, to highly excited, unbound states of SiF+4.

    Notes: Using monochromatized synchrotron radiation, the gas phase photoelectron spectra of the Hg 5d and valence levels of Hg(CH3)2 have been obtained between 21 and 100 eV photon energies. Experimental valence band branching ratios have been compared with theoretical branching ratios from MS-Xα calculations. Our results strongly suggest that the ordering of valence levels is 2a‘2〈3a1〈2e'〈2e‘, rather than 2a‘2〈3a1〈2e‘〈2e' from previous theoretical calculations. For the 3a'1 orbital, two shape resonances are predicted at 30 and 50 eV kinetic energy. Only the one at 30 eV is observed. Interchannel coupling with Hg 5d photoemission is important for the outermost 2a‘2 and 3a'1 orbitals. Shape resonances and interchannel coupling strongly suggest that He ii:He i cross section ratios cannot generally be used to infer orbital character. The Hg 5d intensity ratio in Hg(CH3)2 differs from that of atomic Hg in the low photon energy region. This effect is probably due to the differences in the outgoing f wave potential between Hg and Hg(CH3)2.

    Notes: Using monochromatized synchrotron radiation, gas phase photoelectron spectra of CF3I have been obtained between 21 and 100 eV photon energies. Experimental valence band branching ratios have been compared with theoretical branching ratios from MS-Xα calculations. The generally good agreement between experiment and theory confirms the orbital assignment: 4e〈4a1〈1a2∼3e〈2e〈3a1〈1e∼2a1 〈1a1, in order of increasing binding energy. In contrast to CF4, MS-Xα calculations predict two shape resonances for CF3I at 15.2 eV (e channel) and 17.2 eV (a1 channel). Experimentally, weak shape resonances are observed on e orbitals at ∼14 eV kinetic energy. The cross sections are very useful for correlating orbitals in CF3I with those of CF4. For example, the cross sections suggest that the 3e orbital in CF3I correlates with the 1t1 orbital rather than the 4t2 orbital in CF4. After the onset of the I 4d level at ∼57 eV, intershell coupling becomes important in enhancing valence band levels having high I 5p character such as the 4e orbital. The I 4d branching ratio from 66 to 110 eV is similar to the Xe 4d branching ratio in atomic Xe, indicating atomic-like behavior for the I 4d subshell.
