    Notizen: A new type of instrument has been designed and built to make Fourier transform nuclear-magnetic-resonance (NMR) spectroscopic measurements at temperatures to at least 1200 °C, with a resolution of about 1 ppm. A sample is rapidly and repeatedly shuttled between the furnace and the radio frequency (rf) coil of the NMR probe (both located within a high field superconducting magnet), spending most of the time in the furnace. During each cycle, a series of rf pulses may be given and a single free-induction decay sampled. The rf coil is located outside of the furnace and is maintained at room temperature despite its close proximity to the sample. Electronic stability and coil "filling factor'' are thus both optimized. The sample temperature drops below that of the furnace when cycling begins, but rapidly approaches a nearly constant value with only small fluctuations. 99% BN has been successfully used as a sample container in experiments on 27Al, 29Si, and 23Na in molten silicates in the system Na2O–Al2O3–SiO2.

    Notizen: Abstract Cells of diploid yeast,Saccharomyces cerevisiae, were exposed to a variety of energetic heavy ions (provided by the UNILAC facility at the Gesellschaft für Schwerionenforschung, GSI),241Amα-particles and 80-keV x-rays after which they were assessed for DNA double-strand breaks (DSB) using either the neutral sedimentation or the pulsed-field gel electrophoresis (PFGE) technique. Both yielded comparable results. The DSB production cross-sections are compared with inactivation studies performed for the same cells under identical conditions. The measurements show that with lighter ions DSB induction cross-sections increase with linear energy transfer (LET), but the situation is less clear with the heavier ions. A close parallelism was found between DSB induction and cell inactivation in these yeast cells.

    Notizen: Abstract The effect of a crack on time-of-flight of shear waves (4.5 MHz) polarized in perpendicular (t ⊥) and parallel (t ‖) directions to the crack surface, propagating parallel to the direction of crack growth is investigated. The first and second back-wall echoes are used instead of the weak crack-tip echo for the measurement of time-of-flight. The measurement is made for fatigue cracks grown by different loading histories in ferritic steel (pressure vessel steel A533B-1) under the condition of no loading. The normalized time-of-flight (t ⊥−t ‖)/t ⊥ at the crack position is found to change proportionally as the ratio of crack depth to specimen width increases. The change is mainly due to the effect of plastic deformation occurring around the crack ont ‖. It is shown that the depth of tightly closed fatigue crack in austenitic stainless steel (AISI 304) also can be evaluated under the condition of no loading by using this relationship.

    Notizen: Abstract The induction of resistance to 6-thioguanine by heavy ion exposure was investigated with various accelerated ions (oxygen-uranium) up to linear energy transfer (LET) values of about 15000 keV/µm.31 y Survival curves are exponential with fluence; mutation induction shows a linear dependence. Cross-sections (δi: inactivation, δm: mutation) were derived from the respective slopes. Generally, δi rises over the whole LET range, but separateas into different declining curves for single ions with LET values above 200 keV/µm. Similar behaviour is seen for δm. The new SIS facility at GSI, Darmstadt, makes it possible to study the effects of ions with the same LET but very different energies and track structures. Experiments using nickel and oxygen ions (up to 400 MeV/u) showed that inactivation cross-sections do not depend very much on track structure, i.e. similar values are found with different ions at the same LET. This is not the case for mutation induction, where very energetic ions display considerably smaller induction cross-sections compared with low-energy ions of identical LET. Preliminary analyses using the polymerase chain reaction (PCR) demonstrate that even heavy ions cause “small alterations” (small deletions or base changes). The proportion of the total deletions seems to increase with LET.

    Notizen: Abstract The problem of detection and sizing of fatigue cracks that are closed due to the reverse plastic deformation is addressed in this paper. In particular, it is suggested that the zone of plastic deformation can be identified by measuring the changes in the wave speeds in the plastically deformed material. This is supported through experimental results, first by determining the changes in wave speed with monotonic plastic deformation and then by investigating the plastic zones near fatigue cracks. The characteristic changes in the wave speeds are interpreted in terms of the theory of wave propagation in nonlinearly deformed media and elastic plastic fracture mechanics.

    Notizen: Abstract Using Concanavalin A (Con A) labeled with fluorescein isothiocyanate, we studied the intracellular localization of receptor molecules in the calyptra of 24-h dark-grown cress roots. Fixation in glutaraldehyde gave positive binding of the distal complex of the endoplasmic reticulum and the nucelus in the statocytes. In contrast, fixation in formaldehyde did not preserve the membrane-associated receptors, but revealed Con A affinity of the starch grain surface within the amyloplasts. Treatment of glutaraldehydefixed sections with non-ionic detergents led to partial solubilization of membrane components: the starch grain surface turned positive, though the positive binding of Con A to the endoplasmic reticulum and the nucleus remained unaffected. We therefore conclude that the Con A receptor in the membrane is a glycoprotein tightly inserted in other components of the compartment.

    Notizen: Abstract Phosphorylase was purified from young and senescent potato tubers. Antibodies raised against the enzyme from young tubers crossreacted with phosphorylase from old tissue, although the latter exhibited different physico-chemical properties. In polyacrylamide gel electrophoresis it migrated with higher mobility, its subunit molecular weight was determined in the range of 40,000 in contrast to 100,000 of the phosphorylase in young tubers. The enzyme of senescent tubers displayed an isoelectric point of 5.4 different from the one of young tubers with 5.0, and the diffusion coefficients of the two enzymes varied. The appearance of the phosphorylase form typical for senescent tissue is connected with changes in the intracellular localization as revealed by immunofluorescence. Before massive starch accumulation is initiated, non-vacuolated subepidermal cells contain antigenically active material in their cytoplasm. During starch accumulation in fully differentiated storage parenchyma, only amyloplasts fluoresce, indicating the presence of adsorbed phosphorylase protein. Cytoplasmic phosphorylase can be detected in the continuance of senescence and, finally, after 16 months of tuber storage, the particle-bound enzyme had mostly disappeared. Simultaneously, we observed membrane destruction and decomposition on the ultrastructural level. The phosphorylase from senescent potatoes is a converted molecule and seems to be formed by proteolytic cleavage. The location of phosphorylase in the amyloplasts during starch synthesis indicates that it also plays a role in starch synthesis and not only in its degradation.
