    Description: High-throughput sectioning and optical imaging of tissue samples using traditional immunohistochemical techniques can be costly and inaccessible in resource-limited areas. We demonstrate three-dimensional (3D) imaging and phenotyping in optically transparent tissue using lens-free holographic on-chip microscopy as a low-cost, simple, and high-throughput alternative to conventional approaches. The tissue sample is passively cleared using a simplified CLARITY method and stained using 3,3'-diaminobenzidine to target cells of interest, enabling bright-field optical imaging and 3D sectioning of thick samples. The lens-free computational microscope uses pixel super-resolution and multi-height phase recovery algorithms to digitally refocus throughout the cleared tissue and obtain a 3D stack of complex-valued images of the sample, containing both phase and amplitude information. We optimized the tissue-clearing and imaging system by finding the optimal illumination wavelength, tissue thickness, sample preparation parameters, and the number of heights of the lens-free image acquisition and implemented a sparsity-based denoising algorithm to maximize the imaging volume and minimize the amount of the acquired data while also preserving the contrast-to-noise ratio of the reconstructed images. As a proof of concept, we achieved 3D imaging of neurons in a 200-μm-thick cleared mouse brain tissue over a wide field of view of 20.5 mm 2 . The lens-free microscope also achieved more than an order-of-magnitude reduction in raw data compared to a conventional scanning optical microscope imaging the same sample volume. Being low cost, simple, high-throughput, and data-efficient, we believe that this CLARITY-enabled computational tissue imaging technique could find numerous applications in biomedical diagnosis and research in low-resource settings.

    Description: Close proximity to roadways has been associated with higher exposure to traffic-related air pollutants. However, analyses of the effects of season and meteorological parameters on horizontal gradient patterns of traffic-generated air pollutants still need to be elucidated. Our objectives were to: (1) determine effects of season on horizontal gradient patterns of polycyclic aromatic hydrocarbons (PAHs), total suspended particles (TSP), and PM2.5 near a heavily trafficked highway; and (2) examine the effect of day-of-the-week variations (weekday versus weekend) associated with traffic counts on measured airborne-contaminant levels. PAHs (Σ8PAHs [MW 228–278]; gas + particulate), TSP and PM2.5 were monitored at nominal distances (50, 100, and 150 m) from the New Jersey Turnpike every 6 days for periods of 24 h, between September 2007 and September 2008. Seasonal variations in the horizontal gradient patterns of Σ8PAHs were observed. In the summer, Σ8PAHs declined significantly between 50–100 m from the highway (23% decrease), but not between the furthermost distances (100–150 m). An inverse pattern was observed in the winter: Σ8PAHs declined between 100–150 m (26% decrease), but not between the closest distances. Σ8PAHs and TSP, but not PM2.5, concentrations measured on weekends were 12–37% lower than those on weekdays, respectively, corresponding to lower diesel traffic volume. This study suggests that people living in the close proximity to highways may be exposed to varying levels of Σ8PAHs, TSP, and PM2.5 depending on distance to highway, season, and day-of-the-week variations.

    Description: 〈p〉Stretchable conductors are essential components in next-generation deformable and wearable electronic devices. The ability of stretchable conductors to achieve sufficient electrical conductivity, however, remains limited under high strain, which is particularly detrimental for charge storage devices. In this study, we present stretchable conductors made from multiple layers of gradient assembled polyurethane (GAP) comprising gold nanoparticles capable of self-assembly under strain. Stratified layering affords control over the composite internal architecture at multiple scales, leading to metallic conductivity in both the lateral and transversal directions under strains of as high as 300%. The unique combination of the electrical and mechanical properties of GAP electrodes enables the development of a stretchable lithium-ion battery with a charge-discharge rate capability of 100 mAh g〈sup〉–1〈/sup〉 at a current density of 0.5 A g〈sup〉–1〈/sup〉 and remarkable cycle retention of 96% after 1000 cycles. The hierarchical GAP nanocomposites afford rapid fabrication of advanced charge storage devices.〈/p〉

    Description: We demonstrated an experimental technique based on stimulated light scattering to continuously sample the relative phase of two spatially separated Bose-Einstein condensates of atoms. The phase measurement process created a relative phase between two condensates with no initial phase relation, read out the phase, and monitored the phase evolution.     Description: Spin-polarized gaseous Bose-Einstein condensates were confined by a combination of gravitational and magnetic forces. The partially condensed atomic vapors were adiabatically decompressed by weakening the gravito-magnetic trap to a mean frequency of 1hertz, then evaporatively reduced in size to 2500 atoms. This lowered the peak condensate density to 5 x 10(10) atoms per cubic centimeter and cooled the entire cloud in all three dimensions to a kinetic temperature of 450 +/- 80 picokelvin.     Description: We used radio-frequency spectroscopy to study pairing in the normal and superfluid phases of a strongly interacting Fermi gas with imbalanced spin populations. At high spin imbalances, the system does not become superfluid even at zero temperature. In this normal phase, full pairing of the minority atoms was observed.     Description: 〈p〉In comparison with the advancement of switchable, nonlinear, and active components in electronics, solid-state thermal components for actively controlling heat flow have been extremely rare. We demonstrate a high-contrast and reversible polymer thermal regulator based on the structural phase transition in crystalline polyethylene nanofibers. This structural phase transition represents a dramatic change in morphology from a highly ordered all-trans conformation to a combined trans and gauche conformation with rotational disorder, leading to an abrupt change in phonon transport along the molecular chains. For five nanofiber samples measured here, we observe an average thermal switching ratio of ~8x and maximum switching ratio of ~10x, which occurs in a narrow temperature range of 10 K across the structural phase transition. To the best of our knowledge, the ~10x switching ratio exceeds any reported experimental values for solid-solid and solid-liquid phase transitions of materials. There is no thermal hysteresis observed upon heating/cooling cycles.〈/p〉

    Description: 〈p〉Genes implicated in neuropsychiatric disorders are active in human fetal brain, yet difficult to study in a longitudinal fashion. We demonstrate that organoids from human pluripotent cells model cerebral cortical development on the molecular level before 16 weeks postconception. A multiomics analysis revealed differentially active genes and enhancers, with the greatest changes occurring at the transition from stem cells to progenitors. Networks of converging gene and enhancer modules were assembled into six and four global patterns of expression and activity across time. A pattern with progressive down-regulation was enriched with human-gained enhancers, suggesting their importance in early human brain development. A few convergent gene and enhancer modules were enriched in autism-associated genes and genomic variants in autistic children. The organoid model helps identify functional elements that may drive disease onset.〈/p〉
