    Description: The Genome Database for Rosaceae (GDR, http:/www.rosaceae.org ), the long-standing central repository and data mining resource for Rosaceae research, has been enhanced with new genomic, genetic and breeding data, and improved functionality. Whole genome sequences of apple, peach and strawberry are available to browse or download with a range of annotations, including gene model predictions, aligned transcripts, repetitive elements, polymorphisms, mapped genetic markers, mapped NCBI Rosaceae genes, gene homologs and association of InterPro protein domains, GO terms and Kyoto Encyclopedia of Genes and Genomes pathway terms. Annotated sequences can be queried using search interfaces and visualized using GBrowse. New expressed sequence tag unigene sets are available for major genera, and Pathway data are available through FragariaCyc, AppleCyc and PeachCyc databases. Synteny among the three sequenced genomes can be viewed using GBrowse_Syn. New markers, genetic maps and extensively curated qualitative/Mendelian and quantitative trait loci are available. Phenotype and genotype data from breeding projects and genetic diversity projects are also included. Improved search pages are available for marker, trait locus, genetic diversity and publication data. New search tools for breeders enable selection comparison and assistance with breeding decision making.

    Notes: Zusammenfassung 1. Der Einfluß zweier Kohlenstoffquellen auf die Ruhe- und Vermehrungsatmung INH-sensibler und-resistenter Zellen von M. phlei wird untersucht. Zur Erweiterung der Aussagemöglichkeiten erfolgt parallel zur Messung der Sauerstoffaufnahme die Bestimmung des in der Nährlösung vorhandenen Glycerins. 2. Der Einfluß der zugesetzten Substrate ist mit einigen Ausnahmen auf ruhende und sich vermehrende Bakterien praktisch gleichsinnig. Manche Substrate, wie Essigsäure, werden anscheinend gleichzeitig mit Glycerin verbraucht. Die Wirkung anderer, z. B. Malon- und Glyoxylsäure, setzt erst nach Verbrauch des Glycerins ein. 3. In der Ruheatmung zeigen sich zwischen INH-sensiblen und-resistenten Bakterien deutliche Unterschiede hinsichtlich der Zuordnung der einzelnen Substrate zu den einzelnen Gruppen. Die Sauerstoffaufnahme der sensiblen Keime wird durch Ameisen-, Essig- und Bernsteinsäure besonders charakteristisch beeinflußt. 4. Die Sauerstoffaufnahme sich vermehrender Keime wird durch Zusatz einer 2. Kohlenstoffquelle in den ersten Stunden erniedrigt. 5. Der Zusatz von Citronensäure ist nur bei resistenten Keimen von Einfluß. Er äußert sich in der Ruheatmung wie ein entsprechender Essigsäurezusatz und unterbindet die Vermehrung in der Vermehrungsphase.

    Notes: Summary The acute effect of a single oral dose of isradipine 5 mg on blood pressure, renal haemodynamics, electrolyte excretion and plasma renin activity was studied in 10 healthy males. Isradipine did not produce a significant change in systolic or diastolic blood pressure, and glomerular filtration rate, renal plasma flow, renal vascular resistance, and urinary albumin excretion remained constant. There was a marked natriuretic and diuretic effect about 1–3 h after isradipine. Plasma renin activity showed a slight, insignificant increase 1 h after dosing. Uric acid clearance and β2-microglobulin excretion showed no significant changes, despite an increase in sodium clearance, suggesting an additional mechanism of action other than the proximal tubular natriuretic effect of isradipine in normotensive volunteers.

    Notes: Abstract The pharmacokinetics of torasemide, a new loop diuretic, as well as its active metabolites M1 and M3, and its inactive main metabolite, M5, were studied in 12 patients with end-stage renal failure during single i.v. (n=6) or single oral (n=6) dosing of 200 mg torasemide, and during chronic oral treatment for 9 days (n=12). The elimination half-life (t1/2) of torasemide was unchanged in renal failure, whereas t1/2 of the torasemide metabolites M1, M3, and M5 were markedly prolonged. However t1/2 as well as the area under the plasma level time curve of torasemide and its metabolites were unchanged during chronic compared to acute administration. The results of this study suggest that despite the increased half-life of torasemide metabolites M1, M3 and M5 in end-stage renal failure patients, no accumulation of the parent drug torasemide and its metabolites during chronic dosing is demonstrable.

    Notes: Abstract The availability of ATR devices equipped with a diamond internal reflection element makes it possible to obtain IR absorption spectra of organic substances deposited on inorganic substrates, for example, metals, glass or quartz. The quantitative comparison of these spectra performing mass calibration by Langmuir-Blodgett (LB) layers are reported. The measurements were made in the common configuration with the specimen placed between the internal reflection element and the non-transparent support and in a second configuration with a thin transparent substrate (silicon) placed between the specimen and the internal reflection element. The band areas of the CH2 bands near 2900 cm–1 measured for the two ATR configurations were compared with those obtained from transmission measurements. The results can be partially interpreted by the equation for the effective thickness of thin layers on internal reflection elements. The relative absorbances of several bands of the TSE spectra obtained under different measuring conditions were also investigated.

    Notes: Summary By in situ hybridization, Y-specific DNA sequences were localized on Xp22.3-Xpter of one of the two X chromosomes in all of eleven XX males studied. In nine of the cases the presence of the Y-specific DNA did not affect random X inactivation in fibroblasts. Fibroblasts of the other two cases showed a preferential inactivation of the Y DNA-carrying X chromosome. In only one of these two exceptions blood lymphocytes could also be studied, and here, random inactivation of the Y DNA-carrying X chromosome occurred. Furthermore, the gene dosage of steroid sulfatase (STS) was examined by Southern blot analysis. In ten of the cases including the one showing random X-inactivation in lymphocytes but not in fibroblasts, a double dosage of the STS gene is present. The remaining case with non-random inactivation shows a single STS gene dosage. This case was reported previously to have STS enzyme activity in the male range. It is assumed that, as a consequence of an unequal X-Y interchange, a deletion of X-specific DNA sequences may result in the preferential inactivation of the Y DNA-carrying X chromosome.
