    Notizen: Abstract Transcription of the Trichoderma longibrachiatum egl1 gene is induced in the presence of lactose and β-methylglucoside and repressed by glucose. A DNA fragment containing 722 bp upstream of the ATG codon has been sequenced. The gene has two major transcription start points (20 and 24 nucleotides upstream from the ATG codon) and several transcription termination points (located in a region around 130 nt downstream of the stop codon). Two 6-mer sequences (5′-CTGGAG-3′) separated by 16 bp are present in the egl1 gene promoter. These sequences match the Aspergillus nidulans consensus CreA binding site and might be implicated in carbon catabolite repression of egl1 transcription.

    Notizen: The long-time effect of phosphinothricin (PPT) on gas exchange and nitrate metabolism in intact plants of lucerne (Medicago sativa L. cv. Aragón) was investigated. Photosynthetic CO2 uptake, stomatal conductance, and transpiration were measured with an Infra-Red Gas Analyzer (IRGA). Under photorespiratory conditions, CO2 uptake continuously decreased after PPT treatment. The decrease of photosynthesis led to an increase in the internal CO2 concentration, which in turn caused stomatal closure and a reduction of transpiration rate. Nitrate reduction from plants sprayed with PPT was assayed both in vitro and in vivo. In vivo nitrate reductase was measured with and without nitrate in the infiltration medium. Both types of nitrate reductase assays indicated that the enzyme was inhibited in plants treated with PPT; however, the enzyme appeared more affected when the in vivo assay was used than when the one in vitro was applied. The nitrate reduction was pronouncedly affected after 24 h of PPT treatment, when glutamine synthetase (GS, EC 6.3.1.2.) activity and gas exchange were inhibited by more than 60%. The data suggest that the inhibition of GS leads to inhibition of photosynthesis, which, in turn, means lack of NADPH and nitrate, the substrates for nitrate reductase. The inhibition of GS also leads to a high ammonia level, which will produce a secondary inhibition of nitrate reductase activity.

    Notizen: Aspergillus species included in section Nigri are common in plant products and processed food, such as grapes, cereals, coffee and derivatives, particularly in warm and tropical climates. Two of these species, A. carbonarius and A. niger, are known to produce ochratoxin A (OTA), a potent nephrotoxin and carcinogenic to human (group 2B). Recognition of the several species of this section is difficult and requires considerable expertise using conventional methods based on morphological features. In this work we describe rapid, sensitive and robust assays based on the PCR technique to discriminate the main species included in section Nigri: A. japonicus, A. heteromorphus, A. ellipticus and the two morphologically indistinguishable species of the A. niger aggregate: A. niger and A. tubingensis. The species-specific primers have been designed on the basis of ITS (internal transcribed spacers of rDNA units) sequence comparisons obtained from several Aspergillus strains and have been tested in a number of strains from different origins and hosts. These PCR assays, based on multi-copy sequences, are highly sensitive and specific and represent a good tool for an early detection of OTA-producing Aspergillus species in order to prevent OTA from entering the food chain.

The application of an immunocytochemical method to identify precystic stages and to analyse the encystment kinetics, by using a polyclonal antiserum against isolated cyst walls from the ciliate Colpoda inflata, is reported for the first time. Three different precystic phases were chosen on the basis of morphological changes and degree of cyst wall formation. By using this procedure a better identification of mature resting cysts with regard to precystic cells or young cysts is provided. An average consensus encystment kinetics of C. inflata, by using an accumulated class frequency analysis, is reported.

Free-living amoebae belonging to the species Naegleria fowleri are known to be the etiological agents for a form of fulminant meningoencephalitis that is generally fatal (primary amoebic meningoencephalitis). In a broad bacterial screening from soil and water we have isolated three strains (M-4, D-13 and A-12) belonging to the species Bacillus licheniformis that have remarkable amoebicidal activity against Naegleria sp. and also against different Gram-positive and Gram-negative bacteria. Physical-chemical characteristics, partial purification and biological activities of a substance produced by the M-4 strain have been investigated. This substance (m-4) is stable at high temperature (up to 100°C) and extremes of pH (2.5-9.5) and also at -20°C for months. Its production is greatly influenced by oxygenation of the cultures and is probably related to the sporulation process of the bacterium. Scanning electron microscope observations reveal that amoebae are lysed after a few minutes contact with m-4.

    Notizen: Nuclear and cortical phenomena during dividing and resting cyst formation of Colpoda inflata are described. Cell division forms a cyst and produces two or four tomites. In each tomite, the right oral field results from the proliferation of the anterior extreme of a single kinety, and the left oral field results from the proliferation of four, five, or six somatic kineties. After macronuclear division, each macronuclear mass undergoes a chromatinic extrusion process. During resting cyst formation, the oral infraciliature of the vegetative cell is resorbed. The somatic kineties dispose in a radial way and some pairs of kinetosomes disappear. As in cell division, there is an extrusion process. From these results we conclude that the resting cysts of Colpoda inflata cannot be included in any group of the previous classifications for hypotrich resting cysts. Thus, we propose a new additional group to Walker and Maugel's classification called PKR (partial-kinetosome-resorbing) cysts.

    Notizen: SYNOPSIS. The “Laredo strain” of Entamoeba histolytica has been reported to have different temperature requirements for growth than the usually known strains of the species, although morphological and antigenic tests have thus far indicated no significant recognizable differences. Further genetic studies show the Laredo strain to exhibit at least four characteristics different from the usual. Also, a freshly isolated strain (BA) was shown to have the same characteristics as those previously reported for some well-known strains.

Total or kinetoplast DNA (kDNA) from 72 isolates and clones of Trypanosoma cruzi as well as from nine related trypanosomatids were analyzed by dot hybridization using nonradioactive kDNA or cloned minicircle fragments as probes. Biotinylated-kDNA probes generated by nick-translation proved reliable for distinguishing Zymodeme 1 and Zymodeme 2bol of T. cruzi parasites. In contrast, digoxigenin-labeled kDNA obtained by random-priming did not distinguish among T. cruzi isolates but did distinguish among New World leishmanias. Cloned minicircle fragments labeled with digoxigenin gave the same results as digoxigenin-labeled kDNA, except for a 10-fold decrease in sensitivity. Digoxigenin-labeled DNA probes proved useful in unambiguously detecting T. cruzi from different geographic regions of America. However, T. rangeli and T. cruzi marinkellei were not distinguished by these probes.

A field study was conducted to assess the effect of the nitrification inhibitor 3,4-dimethylpyrazole phosphate (DMPP), applied at a rate of 1 kg ha−1, on nitrous oxide (N2O) emissions, forage production and N extraction from a grassland soil after cattle slurry applications in autumn and spring. Nitrous oxide emissions were measured daily or weekly using the closed chamber technique. DMPP efficiency after slurry application was lower in spring (16.7 °C mean soil temperature) than in autumn (11.4 °C mean soil temperature). Thus, DMPP was able to maintain soil mineral N in the ammonium form for 22 days and reduce cumulative N2O emissions by 69% in autumn, while in spring its effect on soil mineral N lasted for 7–14 days, reducing cumulative N2O losses by 48%. Furthermore, application of DMPP after slurry did not decrease biomass yield or N uptake.

    Notizen: DNA oligotyping was used to determine HLA-A28 subtypes in 25 unrelated Caucasian individuals living in or around Seville, Spain. Results showed that HLA-A*6802 was the most frequent allele, found in 14 individuals (53.8%), followed by HLA-68.3, which was present in eight subjects (30.8%), and both combined represented 84.6% of A28+ individuals in the area. The HLA-A*6801 allele was found in three individuals (11.5%), whereas HLA-A*6901 was present in one subject only (3.8%). Results indicate that the distribution of HLA-A28 alleles can vary among different Caucasoid populations. In this way, the high frequency obtained for A*6802 supports previous studies suggesting that the HLA-A*6802 allele was prevalent in people of the Mediterranean basin, in contrast to A*6801, prevalent in northern European populations.
