Notes: Summary Polluted water flowing over stabilized and active deposits is purified in 3 ways. 1. Physical adsorption and biochemical degradation. 2. Biochemical processes in deposits over which water flows can be quantitatively complete since, unlike conventional systems, they do not depend on the nutrients contained in the pollution load itself, but take place in the sediment where supply of available food can become less easily unbalanced or exhausted than the food contained in the pollution. Thus, if a polluted water is allowed to flow through and over an active stabilized sediment, degradation may become one hundred percent. 3. In contrast to existing opinions, bottom deposits of maturation ponds and natural waters are anaerobic or semi-aerobic systems which supplement the aerobic processes regarding the removal of carbon by gassification as CH4, nitrogen as N2 and possibly sulphur as H2S. Our conventional concept of pond systems being purely aerobic has to be revised since even in aerobic systems, processes taking place in the bottom deposits, are anaerobic. These aspects should be considered in future research leading to attempts to design more efficient sewage works.

Notes: Summary A method has been described which permits the quantitative and qualitative determination of the toxicity of effluents on biochemical purification and self-purification processes. The test is designed so that it can indicate whether or not adaptation to the toxic substances occurs and is based on comparing measurements of the reduction of the dehydrogenase activity of a system when treated with known toxic standards and effluents of unknown toxicity. The effect of known toxic standards, such as Ag+, Hg++, Cr VI, formaldehyde, phenol and of some industrial effluents on the dehydrogenase activity, was determined under standardized conditions and the results are presented graphically. The method may be modified to suit particular local requirements. The advantage of this method is that it does not require dilution as is the case with the BOD bottle technique nor does it require expensive or elaborate equipment as the Warburg Apparatus and the method is suitable for series analysis.

Notes: Summary Some physiological considerations led to the following conclusions: Although the calculation of the ultimate oxygen demand, as proposed by Gameson et al., might be a practical and rapid way to estimate roughly the theoretical polluting potential of effluents, no existing theory based on physiological principles can support the soundness of the U.O.D. formula to predict the oxygen distribution in effluents or receiving waters which are unbalanced in their organic and mineral available constituents. It is, therefore, felt that caution should be exercised when U.O.D. results are interpreted in terms of the oxygen distribution and polluting potential of effluents not balanced with respect to nutritional factors or containing inhibitors.

Notes: Summary Samples of bottom deposits were taken from a total of twenty-six stations in five areas along the Vaal River and analysed for dehydrogenase activity, organic carbon, Kjeldahl nitrogen, inorganic carbon, humic acids, degree of humification, cationic adsorption capacity and exchangeable bases. BOD determinations on the flowing water were also done at these stations. The relation between the characteristics of the sediments and the presence or absence of pollution was examined. It was concluded that analysis of the sediments can provide a valuable index of the state of pollution of natural waters.

Notes: Summary A method has been described for the determination of protease activity in sediments of sewage stabilization ponds. As a result of the experiments standard conditions for the method regarding pH, buffer, incubation time, temperature and sample quantity were established and the results shown graphically. The method, which is suitable for serial analysis, characterizes the actual protein degrading efficiency of stabilization pond sediments.

Notes: Summary A rapid quantitative method for the determination of nitrotase activity based on principles of enzyme kinetic was devised. Factors determining the rate of denitrification likepH, substrate concentration, reaction time and temperature were studied. This method enables the operator of industrial effluent purification plants to study and control the performance of denitrifying plants at all times.
