    Notes: —Free amino acid (FAA) analyses were conducted on 87 I. dorsi and 58 b. femoris samples from cattle varying in sex, breeding and age to study the relationships between these muscle constituents and Warner-Bratzler Shear values. The b. femoris contained greater amounts of FAA than the I. dorsi and was significantly tougher. Muscles from a line of cattle that tended to be more tender possessed greater amounts of FAA. With few exceptions the I. dorsi of steers contained greater amounts of FAA than the same muscle from bulls. Differences in bull muscles due to age were not significant for any FAA. Steer muscle with low shear values tended to possess greater amounts of FAA than steers with high shear values. Although not significantly correlated individual and total FAA increased slightly with increasing tenderness within beef muscles.

    Notes: —The effect of starches on utilization of protein and lipid composition in the liver of rats was investigated. The carbohydrates tested were rice starch, corn starch, wheat starch, and arrow root starch. Criteria for evaluation of protein utilization were growth response, digestibility of protein, and availability of lysine. Arrow root starch was inferior to corn starch, rice starch, and wheat starch in effect upon protein utilization. Corn starch and wheat starch were similar in effects on protein utilization. Studies of growth and protein efficiency ratios indicated that rice starch is inferior to corn starch and wheat starch. The distribution of fatty acids in the liver lipid was somewhat different in rats fed arrow root starch from that in rats fed the other three starches. Lysine supplementation did not alter the fatty acid pattern in the liver lipid of rats from that in the unsupplemented group.

    Notes: Pectinesterase (PE) was inhibited by sucrose at the same concentrations that delayed gelation of papaya purée. The inhibitory effect was linear with sucrose concentration throughout the range investigated (up to 50% sucrose). Conditions for optimum PE activity, pH 7.5 and 0.21M NaCl, were not affected by inhibitory concentrations of sucrose. Evidence was obtained against competitive inhibition, transferase activity, and an effect on the binding of PE as possible mechanisms for inhibition. Other sugars tested, including glucose, maltose, and corn syrups of varying degrees of hydrolysis, as well as glycerol were also inhibitory.

    Notes: Volatile compounds that may contribute flavor to a wide variety of foods can be isolated in an apparatus which is described. It utilizes the principle of flash evaporation and vaporization from a continuous thin heated film. Liquid foods, such as fruit juices or beer, need no preparation, but foods such as meat and potato chips are first made into a fine water slurry. Vaporized food constituents and water are recovered by condensation in a series of specially designed cold traps. Features and performance of the apparatus are discussed.

    Notes: SUMMARY: Steaks from 24 bulls 20 months old, 89 bulls 14 months old and 109 steers 20 months old were used to relate thickness of epimysial tissue to the amount of intra-muscular connective tissue, and to correlate these factors with Warner-Bratzler shear values in muscles cooked to 140 or 160°F. The thickest portion of the LD epimysium was greater in lighter weight bulls than it was in heavier steers. Epimysial extension and thickness varied according to muscle location but at the same location it was not significantly different in steaks from tough versus tender steers. Differences in hydroxyproline content and connective tissue of steers and young bulls were small but older bulls had slightly greater amounts of both. The LD epimysium was tougher in older bulls than in younger bulls.Shear values of epimysial tissue varied considerably within animals of the same age, especially when roasted to 140°F. Most connective tissue differences due to line of cattle were small. Correlations within groups of cattle between thickness of epimysial tissues of different muscles or between epimysial thickness and intramuscular connective tissues were low. With few exceptions neither hydroxyproline, epimysial thickness nor connective tissue scores were significantly related to shear force of muscle or shear force of epimysial tissue itself. Correlations between shear force of the LD and BF muscles were not significant (P 〈 0.05) in animals of the same age. There was a tenderness interaction between age of animal and type of muscle which suggested that collagen gelatinization occurred at a faster rate in the LD than in the BF as age increased.

    Notes: SYNOPSIS. The distribution of cytoplasmic DNA as determined by H3-thymidine incorporation in the Fernald strain of Hartmannella rhysodes was studied by electron microscope autoradiography. Of a total of 1,313 silver grains counted over the cytoplasm in thin sections, 488, 271, 202 and 230 were attributed to DNA in mitochondria, cytoplasmic matrix, plasmalemma and ectoplasm, respectively. On the basis of the ratio of grains associated with the relative area occupied by the various compartments, the plasmalemma accounted for 3 times more grains than the mitochondria and about 20 times that attributed to DNA in cytoplasmic matrix and ectoplasm. These findings are interpreted to indicate the presence of endosymbionts in this strain of soil amoeba. Since no definitive microorganism could be seen in this cell, the most likely endosymbiont is defective DNA virus(es) or episome-like genetic element(s).

    Notes: Corn seeds were treated with 0.01 M sodium fluoride for various time periods. The treated seeds were germinated and grown until the seedling roots reached a standard size of 12±3 mm.Analyses were made for RNA and DNA contents of 3-mm seedling root tips. Determinations also were made for growth rate, rate of cell elongation, cell multiplication, and tissue maturity of 12-mm roots.RNA contents of 3-mm root tips were found to be directly proportional to the growth rates of the entire seedling root of corn seeds treated with sodium fluoride for various periods of time. The RNA content was reduced on a cell basis and was independent of the root tip cell number. The amount of DNA was not related to the growth rate of the intact seedling roots. Since fluoride reduced the number of mitotic figures, it was likely that fluoride inhibited DNA synthesis during the interphase of the mitotic cycle.Growth by cell multiplication was inhibited more than that by cell elongation in the sample treated with fluoride for a shorter period. The two types of growth, however, showed a similar level of growth reduction in the sample treated with fluoride for a longer period. Fluoride seemed to reduce the rates of cellular elongation and multiplication not more than about 40 per cent of the control value in these tissues under present experimental conditions. Fluoride also induced maturity in the seedling roots in proportion to the periods of fluoride treatment.

    Notes: Environmental factors affecting basidiospore germination were studied with Volvariella volvacea, the edible straw mushroom which is common in South East Asia. A relatively mild heal shock is necessary for spore germination. The spores give best germination at 40°C although early hyphal growth is better at 35°C the germination of spores is affected by temperature. pH. a presoaking treatment and spore density. Higher pH supports more germination but seems unfavourable for early mycelial growth. Presoaking treatment in phosphate buffer solution or distilled water also stimulates germination markedly.

    Notes: (1) The value of and the applications for the augmented histamine test are discussed. Proposals are put forth in reference to the nomenclature of gastric acid and the interpretation of the augmented histamine test for routine clinical purposes.(2) The results of the augmented histamine test in 14 patients with proven gastric ulcer and in 68 subjects with proven duodenal ulcer are presented. The mean acid outputs of men with gastric ulcer (all sites) for basal and histamine-stimulated secretion were about one-half that of men with duodenal ulcer. These differences were significant, statistically.(3) Duodenal ulcer patients with complications had a significantly higher maximal acid output to histamine than those without complication. The finding of a maximal acid output greater than 40 mEq/hr is highly suggestive of the presence or the imminence of a complication.(4) We prefer the use of the augmented histamine test as a routine gastric analysis procedure, since it is a quantitative test and, as such, gives reproducible acid output in the same individual, provides an estimate of the parietal cell mass, and gives a positive lead to the diagnosis of Zollinger-Ellison syndrome.
