Zusammenfassung 1. An jeweils 6–7 Organen des Flußkrebses Orconectes limosus wurden die Aktivitäten der Alanintransaminase (GPT), Aspartattransaminase (GOT), Glutamatdehydrogenase (GDH), D-Aminosäureoxydase und L-Serindehydratase, die Transaminierung von verschiedenen L-Aminosäuren auf α-Ketoglutarat und die Steigerung der Homogenatatmung durch L- und D-Aminosäuren untersucht. 2. Mit dem gekoppelten optischen Test wurden GPT und GOT in allen untersuchten Organen nachgewiesen; mit Ausnahme des Bauchmarks ist die Aktivität der GPT stets mindestens ebenso hoch wie die der GOT. Die Gleichgewichtskonstanten der GPT und GOT stimmen mit denen der Säugerenzyme etwa überein. 3. In der Antennendrüse wurde mit Hilfe von Glutamatdecarboxylase Transaminierung von 15 weiteren L-Aminosäuren auf α-Ketoglutarat nachgewiesen; einige dieser Reaktionen wurden auch in Enddarm, Kiemen und Mitteldarmdrüse gefunden. 4. GDH wurde in allen untersuchten Organen nachgewiesen; in der Mitteldarmdrüse vorhandene Hemmfaktoren verhinderten dort die quantitative Bestimmung. Die optimalen Bedingungen der GDH-Reaktion wurden an der Antennendrüse untersucht. Sie entsprechen etwa denen der Säugerenzyme; das Krebsenzym zeigt jedoch kein scharfes Optimum der NADH-Konzentration. Die Reaktion verläuft in Richtung Glutamatsynthese etwa 15–20fach rascher, als in Richtung Glutamatabbau. 5. D-Aminosäureoxydase war nur in der Antennendrüse nachzuweisen; das Enzym ist spezifisch für D-Glutamat und -Aspartat. Mitteldarmdrüsen-Extrakte hemmen die D-Aminosäureoxydase der Hammelniere. Unter Bedingungen, unter denen zugesetztes Säugerenzym von dem Hemmfaktor abgetrennt und rückgewonnen werden kann, ist jedoch keine D-Aminosäureoxydase aus der Mitteldarmdrüse zu isolieren. Diese enthält also entgegen der Vermutung von Sarlet u. Mitarb. (1950) keine dem Säugerenzym ähnliche D-Aminosäureoxydase. 6. L-Prolin bewirkt eine Erhöhung der O2-Aufnahme an Homogenaten der Antennendrüse, des Enddarms und der Kiemen, L-Tyrosin nur an Homogenaten der Kiemen. 7. L-Serindehydratase konnte in allen untersuchten Organen nachgewiesen werden; mit D-Serin, L- und D-Threonin erfolgte keine signifikante Bildung von Ketosäuren. 8. Die spezifischen Aktivitäten der GPT, GOT, GDH und L-Serindehydratase liegen beim Flußkrebs in der gleichen Größenordnung wie bei den Säugetieren. 9. Die höchsten Aktivitäten der untersuchten Enzyme des Aminosäurestoffwechsels wurden in Antennendrüse und Herz gefunden, die niedrigsten in Mitteldarmdrüse und Muskel.

Summary 1. In each of 6 or 7 tissues of the crayfish Orconectes limosus the activities of alanine transaminase (GPT), aspartate transaminase (GOT), glutamate dehydrogenase (GDH), D-amino-acid oxidase and L-serine dehydratase were determined. Transamination from different L-amino acids on α-oxoglutarate and stimulation of homogenate respiration by L- and D-amino acids were assayed. 2. GPT and GOT were demonstrated by spectrophotometric methods in all the tissues studied; with the exception of nervous tissue the activity of GPT was always at least as high as the activity of GOT. The equilibrium constants of GPT and GOT corresponded to those of mammalian enzymes. 3. Amino-group transfer from 15 other L-amino acids to α-oxoglutarate was detected in the antennal gland with the aid of glutamate decarboxylase; some of these reactions could be shown to be present also in hindgut, ctenidia and hepatopancreas. 4. GDH was found in all organs examined. Inhibitory factors present in hepatopancreas made quantitative determination of GDH impossible in this organ. Optimal conditions of GDH reaction as studied in the antennal gland corresponded closely to those of mammalian enzymes; contrasting to mammalian GDH, the crayfish enzyme displayed no sharp optimum of NADH concentration. The reaction proceeded in the direction of glutamate synthesis about 15–20 times faster than in the opposite direction. 5. D-amino-acid oxidase was demonstrable in antennal gland only. The enzyme was specific for D-glutamate and D-aspartate. Extracts of crayfish hepato-pancreas inhibited the D-amino-acid oxidase of sheep kidney. Purification methods, which allowed to separate and recover added mammalian enzyme from the inhibitory factor, did not yield any D-amino-acid oxidase, if applied to hepato-pancreas extracts. Thus it can be stated that contrarily to suppositions of Sarlet et al. (1950) the hepato-pancreas of crayfish contains no enzyme comparable to mammalian D-amino-acid oxidase. 6. L-proline stimulated respiration in homogenates of antennal gland, hindgut and ctenidia, L-tyrosine in ctenidia only. 7. L-serine dehydratase could be demonstrated in all the tissues studied. With D-serine, L- and D-threonine there was no significant production of keto-acids. 8. The specific activities of GPT, GOT, GDH and L-serine dehydratase in the crayfish equaled those in mammals in order of magnitude. Although the intensity of energy metabolism in the crayfish is lower than that of a mammal of the same body weight by a factor of 50–100, there is no corresponding difference in the activities of many enzymes. 9. The highest activities of the enzymes examined were found in antennal gland and heart, the lowest in hepato-pancreas and muscle.
