Notes: Summary Macromolecular orientation (evaluated from linear dichroism findings) and isoelectrical point were investigated on periodontal fibers of adult rats and compared with data on tendon and cementum collagen bundles. Linear dichroism data pointed out similarities on protein orientation, but some differences concerned with patterns of acid mucopolysaccharides orientation could be shown, as for instance, the absorption maxima on periodontal fibers wavelength, what is in accordance with findings on the rat cementum, but differs from patterns for adult tendon collagen fibers under same experimental conditions. On the other hand, they are quite similar to those found in the early stages of experimental regenerations of the tendon collagen. Also differences concerned with the wavelength of the absorption peak in the parallel direction were shown in sections impregnated with silver and gold. These modifications of dichroism patterns which are indicative of differences in molecular orientation are assumed as being related to a less evolved degree of maturity of the collagen fibers in the rat periodontium, which is supported by morphology and birefringence observations. Less orientation of the macromolecules is shown in the plexus region of the periodontium as compared with the juxtacementar zone. The apparent isoelectrical point was found as being placed within the range which is usually reported for collagen bundles.

Notes: Summary The larval peritrophic membrane ofMelipona quadrifasciata (Hym., Apoidea) was studied with respect to morphological, histochemical, and histophysical properties in the four larval instars prior to intestinal discharge. The peritrophic membrane is composed of acid and neutral polysaccharides, proteins, and lipids, whose concentrations apparently vary with the larval instar. There are also granules of calcium and acid mucopolysaccharides. The aggregation of macromolecules (mainly acid polysaccharides and proteins) in the peritrophic membrane is revealed by birefringence and dichroism phenomena. Since the same components which make up the membrane are also found in the midgut cells and in most cases they show a similar ordering molecular organization, it is assumed that the larval peritrophic membrane is secreted by the entire length of the midgut epithelium. Migration of enzymic products from the midgut cells to the lumen and of digested food in the reverse direction may depend on concentration and aggregation of the substances (mainly mucopolysaccharides) which make up the membrane. The main function assumed for the peritrophic membrane is that of a diffusion controller.

Notes: Summary Acid and neutral polysaccharides exist in the secretion of the silk glands of Melipona quadrifasciata during the entire functional stage, except in the late 5th instar. However, only from the 3rd instar on is a molecularly oriented structural protein present. The following evidence indicates that this protein is not collagen: (1) the optical sign of its birefringence does not become reversed after von Ebner's reaction, and (2) its secretion can be detected when the cell nucleus attains a ploidy degree optimal for secretory activity. This is at first observed in the distal part of the glands and then throughout the organ.
