    Description / Table of Contents: Zusammenfassung Hybridisierungskinetiken der DNA mit markierter (18s+28s)rRNA aus HeLa-Zellen wurden ermittelt bei Patienten mit Trisomie 21, gesunden Probanden mit normalem Karyotyp und Trägern einer zentrischen Fusion t(DqGq). Die Ergebnisse bestätigen die bereits früher ermittelten Sättigungswerte von Hybridisierungen bei diesen Probanden: 3 Patienten mit Trisomie 21 zeigten erhöhte Sättigungsniveaus. Bei 2 dieser Patienten übersteigt der Überschuß an rDNA den Erwartungswert erheblich. 3 der untersuchten Translokationsträger t(14q21q) hatten signifikant erniedrigte Sättigungswerte, während die vierte Probandin mit dem dizentrischen Chromosom t(15q21q) im Normbereich lag. Die unterschiedlichen Befunde werden mit den cytogenetischen Eigenschaften der akrozentrischen Chromosomen in Zusammenhang gebracht. Es wird erwartet, daß diese Unterschiede sich durch Familienuntersuchungen aufklären lassen.
    Notes: Summary The hybridization kinetics of DNA with labeled (18s+28s)rRNA from HeLa cells was determined in patients with trisomy 21, healthy probands with normal karyotype and in carriers of a t(DqGq) centric fusion. The results are in, accordance with the saturation values obtained earlier for these probands: Three patients with trisomy 21 showed an increased saturation level. In 2 of these patients the excess of rDNA exceeded the expected value considerably. Three of the investigated translocation carriers t(14q21q) showed significantly reduced saturation values, while results of a fourth proband with a dicentric chromosome t(15q21q) were found to be within the range of normal probands. The heterogeneous results were correlated with the cytogenetic characteristics of the acrocentric chromosomes. It is expected that these differences could be explained on the basis of family analysis.

    Description / Table of Contents: Zusammenfassung Es wird über Befunde zur Entstehung von Bandenmustern berichtet, die durch verschiedene Färbungen unterschiedlich vorbehandelter Metaphasechromosomen gewonnen wurden. Aus der Diskussion dieser Befunde im Zusammenhang mit den Befunden anderer Autoren ergibt sich: 1. Eine Alteration des DNA-Protein-Komplexes ist bestimmend für alle Banden, die sich mit Giemsa anfärben lassen. 2. Über eine Entsprechung von Basenverhältnis der DNA und Bandenmustern hinaus existiert eine dem Basenverhältnis spezifisch zugeordnete Besetzung der DNA mit sauren Kernproteinen.
    Notes: Summary Results are reported on the production of banding patterns stained with Giemasa. They are obtained from the application of different dyes and dye mixtures to metaphase chromosomes which were pretreated in various ways. The discussion of our findings in addition to those of other authors allows the following conclusions: 1. Alterations of the DNA-protein complex determine the production of all banding patterns stained with Giemsa. 2. In addition to the accordance between base ratio of the DNA and banding pattern, there exists a base specific attachement of qualitative different non-histone proteins to DNA.

    Description / Table of Contents: Zusammenfassung Die Assoziationsfrequenzen individueller, durch Fluorescenz-Marker identifizierbarer, akrozentrischer Chromosomen wurden bei 11 Probanden bestimmt und in Beziehung gesetzt zur jeweiligen Länge der NOR-Region. Hierbei ergab sich: 1. Nicht nur Assoziationsfrequenzen von Chromosomenpaaren, sondern auch die homologer Chromosomen können sich signifikant unterscheiden. Die Assoziationsfrequenz its jeweils ein konstantes Charakteristikum eines individuellen akrozentrischen Chromosoms. Die oft sehr unterschiedlichen Assoziationsfrequenzen der jeweiligen Homologen bestimmen somit das Assoziationsverhalten der akrozentrischen Chromosomenpaare. 2. Bei homologen Chromosomen zeigt dasjenige mit der größeren NOR-Region auch stets eine höhere Assoziationsfrequenz. Verschiedene Interpretationsmöglichkeiten dieser Befunde werden diskutiert.
    Notes: Summary We analyzed individual acrocentric chromosomes identified by fluorescence markers as to their association behavior and measured the lengths of their secondary constrictions. Significant differences were found to occur not only between the frequencies of association of pairs of acrocentric chromosomes, but also between the individual homologues. The frequency of association is a constant and characteristic property of an individual acrocentric chromosome. Differences between the frequencies of association of pairs of acrocentric chromosomes result from widely differing contributions of their respective homologues. Differences between the association frequencies of homologous acrocentric chromosomes correlate with differences between the lengths of their nucleolar constrictions: the homologue with the longer constriction shows the higher frequency of association. Several alternative interpretations of these results are discussed.

    Description / Table of Contents: Zusammenfassung Chromosom 21 weist stets eine schwächere C-Bande am Zentromer auf als Chromosom 22, wodurch diese Chromosomen eindeutig unterscheidbar sind. Belegt wird dieser Befund durch Karyotypen mit zentrischer Fusion t(Dq21q), Trisomie 21, und durch einen Vergleich des C-und Q-Banden-Polymorphismus der kurzen Arme.
    Notes: Summary Chromosome No. 21 consistently shows a lighter C band at the centromere than chromosome No. 22, allowing reliable differentiation. This distinction was demonstrated in karyotypes with centric fusions t(Dq 21q), trisomy 21 and by comparing the C-and Q-band polymorphism of the short arms of the G chromosomes.

    Notes: N-Allyl-2-methylaniline (12) forms on heating at 140° in xylene in the presence of zinc chloride 2-allyl-6-methylanline (19) as major compound and 4-allyl-2-methylaniline (20) as well as 2,7-dimethyl-indoline (21) as minor products. Compound 21 is also formed when 19 is heated in the presence of zinc chloride (scheme 2). That 19 arises from a charge-induced [3s, 3s] sigmatropic rearrangement of 12 - and 20 from two consecutive [3s, 3s]-sigmatropic transformations - follows from the reaction of N-crotyl-2-methylaniline (13) in the presence of zinc chloride at 140°. 2-(1′-Methylallyl)-6-methylaniline (22) and 4-crotyl-2-methylaniline (23) are formed exclusively. Small amounts of 2,3,7-trimethyl-indoline (24) and 2-(cis- and trans-1′-methyl-propenyl)-6-methylaniline (cis- and trans-25) are observed as by-products. Compound 24 arises from 22 in the presence of zinc chloride (scheme 3). Similar results are obtained when N-allyl and N-(2′-methylallyl)-N-methyl-aniline (14 and 15, respectively) are heated in the presence of zinc chloride. Whereas 14 gives nearly exclusively 2-allyl-N-methyl-aniline (28) and only small amounts of the corresponding 1, 2-dimethyl-indoline (29) and of 2-(cis- and trans-propenyl)-N-methyl-aniline (cis- and trans-27), 15 forms comparable amounts of 2-(2′-methylallyl)-N-methyl-aniline (30), 1,2,2-trimethyl-indoline (31), and 2-isobutenyl-N-methyl-aniline (32) (scheme 4). Compound 30, and also 32, are transformed into 31 on heating in the presence of zinc chloride.Charge-induced aromatic amino-Claisen rearrangements are also observed when N-allylated anilinium tetraphenylborates are heated at 100-105° in hexamethyl phosphoric acid triamide. Thus, N-allyl- and N-crotyl-N, N-dimethyl-anilinium tetraphenylborate (16 and 17, respectively) yield 2-allyl- and 2-(1′-methylallyl)-N,N-dimethyl-aniline (33 and 34, respectively) besides small amounts of N, N-dimethyl-aniline. N-Cinnamyl-N, N-dimethyl-anilinium tetraphenylborate (18) gives, besides appreciable amounts of N,N-dimethyl-aniline, a mixture of 2-(1′-phenylallyl)-,2-cinnamyl-, and 4-cinnamyl-N, N-dimethyl-aniline (35, 36, and 37, respectively) in which the first two compounds predominate.
