Notizen: A process for encapsulating cotton fibers in the form of woven clothes at ambient temperatures with polyoxymethylene is described. It involves pretreatment of the cotton with a carbon tetrachloride solution of tributylamine and diphenylamine to deposit the basic catalyst on the surface of the fibers. Subsequent exposure to dry, purified vapors of monomeric formaldehyde at 25°C results in the encapsulation of individual fibers with high molecular weight polyoxymethylene. The process is relatively fast (30% fiber weight gain in approx. 20 min) with the polymer located only at the fiber surfaces. Fiber cementation does not become pronounced for weight increases below approximately 60%-65% based upon the weight of the fibers. Thickness of the polymer sheath increases with the time of the polymerization reaction. Because of the uniformity of the polymer layer deposition, the encapsulated cotton cellulose fibers can be expected to exhibit substantially different physical and chemical surface properties even at low polymer add-on. The tensile and tear strengths of the polyoxymethylene-encapsulated cotton fibers are generally improved, with some gains in abrasion resistance also noted. Differential staining techniques and polarized light microscopy have shown the location and uniformity of the polymer sheaths.

Notizen: The origin of chloride ion in chloroform and methylene chloride solutions of tertiary amines is studied. The applicability of such solutions in the analytical control of solid phase peptide synthesis must be seriously questioned. An alternative procedure, amenable to automation, is suggested.

Notizen: The systematic analysis of solid phase peptide synthesis by Edman degradation becomes possible by the development of a technique (high pressure liquid chromatography) for the determination of phenylthiohydantoins in trace amounts. By its qualitative beside the quantitative results the degradation method offers the possibility to follow formation and propagation of failure sequences over more than one synthetic cycle. Scope and limitations of the method are discussed.

Notizen: 3-Hydroxyhydantoins, representing a new class of compounds, are easily accessible from esters of α-isocyanato fatty acids and hydroxylamine via esters of α-hydroxyureido fatty acids. Structural assignment is based on Exner correlations of infrared spectra and other physical data. Chemical properties are as expected from those of hydantoins.

Notizen: -1. FDP aldolase from pupae of Drosophila melanogaster is shown by the following findings to be a tetrameric molecule with two non-identical subunits: (a) Disc electrophoresis on polyacrylamide gel in the presence of sodium dodecylsulfate yields a subunit molecular weight of 40000 (equal to that of rabbit muscle aldolase). (b) Hybridisations of drosophila aldolase with type C aldolase from rabbit and calf brain lead to a five-membered set of isoenzymes.2. The non-identity of the subunits is suggested by the behaviour on isoelectro focussing. While rabbit muscle aldolase and human liver aldolase show five isoenzymes (hybrids from the non-identical subunits), drosophila aldolase can be separated into three peaks only. This result may be interpreted in two ways: (a) The isoenzymes α4 and α3α′ are not separated from each other in the pH range 5--8, neither are αα′3 and α′4. Or (b) α2 and α′2 are not split under the condition of isoelectro focussing and therefore 3 isoenzymes α4, α2α′2, and α′4 are obtained only.(3) Isoenzyme I (supposedly mainly α4) and isoenzyme III (supposedly mainly α′4) are identical in the following respects: Michaelis constant, pH profile, FDP/F-1-P activity ratio, inactivation by carboxypeptidase A, disc electrophoresis on polaycrylamide and molecular weight. The only difference found between isoenzyme I and III is the specific activity: Isoenzyme I is nearly twice as active as isoenzyme III.4. The fact that hybridisation between drosophila aldolase and type C aldolase from rabbit and calf brain yields a set of isoenzymes proves that interspecies hybridisation is possible between FDP aldolases from vertebrates and invertebrates. FDP aldolases from vertebrates and invertebrates seem to be homologous enzymes. Our results suggest that the conformation of aldolases remained relatively unchanged during evolution.

Notizen: Hydrazoacetic acid (7) and a variety of derivatives by alteration of the hydrazo or the carboxyl groups were synthesized. 7 forms complexes with metalions. Under certain conditions N-acety-N′-glycyl-hydrazoacetic acid (17) undergoes rearrangement into N-(N-acetyl-glycyl)-hydrazoacetic acid (18).

Notizen: Byproducts of peptide synthesis, formed in the course of accidental attack by Pd-activated oxygen on ethanol and triethylamine in reaction media, have been identified as products of condensation between peptides and acetaldehyde, and demonstrated to represent N,N′-alkylidenepeptides, i.e. derivatives of 4-imidazolidinones. The same products are obtained by direct action of acetaldehyde on the respective peptides, and analogs resulted in some cases when acetaldehyde was replaced by formaldehyde or acetone. The condensation is reversible, imidazolidinone stability depending on structural features of both the involved peptide and carbonyl compound. There are evidently implications of these findings with respect to the chemistry and biochemistry of peptides and proteins.

Notizen: Sedimentation of collagen or cellulose fibers freshly precipitated from aqueous solutions containing certain soluble, flexible polyelectrolytes compacts them to pellets which may incorporate considerable amounts of the polyelectrolyte. The amount of polyelectrolyte incorporated increases with its chain weight and with its concentration in the solution from which the fibers were precipitated. The amount of water held by the pellets after compaction under a standard set of conditions generally increases with the amount of polyelectrolyte which has been incorporated into the pellet. Pellets formed by the compaction of cellulose fibers in the presence of polyelectrolyte are coherent although those formed in the absence of polyelectrolyte are not; all collagen pellets are coherent. All coherent pellets can be dried and, subsequently, reconstituted by being soaked in.15 M KCl. Reconstituted collagen pellets hold over two times as much water as the initial wet pellet formed by compaction; reconstituted cellulose pellets hold on the average about ⅓ the amount of water as the initially compacted pellets. The reconstituted pellets can again be dried and reconstituted with generally little further loss of polyelectrolyte or change in capacity to take up water.

Notizen: Composite hydrophilic materials (CHM) containing collagen or cellulose as the insoluble fibrous component and potassium salts of proteinpolysaccharide or carboxymethylcellulose as the soluble polyelectrolyte were prepared as simple models of the connective tissue, cartilage. The tensile properties of uncrosslinked and formaldehyde-crosslinked collagenous CHM were measured and compared to those of bovine nasal cartilage (BNC). The compressive properties of uncrosslinked and formaldehyde-crosslinked cellulosic CHM were measured and compared to bovine nasal cartilage. Although elongating to approximately the same extent before breaking (20%), the uncrosslinked collagenous CHM had lower tensile strengths than BNC (330 psi). Formaldehyde crosslinking increased the tensile strength of the collagenous CHM to as much as 3 times that of BNC but reduced the elongation at failure to only 7%. The uncrosslinked cellulosic CHM attained a maximum of only ¼ the compressive strength of BNC (940 psi). Formaldehyde crosslinking extended the compressive strength to as high as ⅓ that of BNC. The results indicate that the simple uncrosslinked and crosslinked models can only partially account for the mechanical properties of BNC.
