    Notizen: Summary In the human heart, it is well known that the polyploidization of working heart-muscle cells increases in proportion to increases in heart weight, but there has been no investigation of the process of polyploidization in the specialized heart-muscle cells of the cardiac conduction system which have a nerve-like function. In order to investigate the process of polyploidization in these cells, the nuclear DNA content of atrioventricular nodal cells was measured using cytofluorometry. Tissue samples taken from autopsied hearts without arrhythmias were embedded in paraffin blocks after Carnoy fixation. Blocks containing the atrioventricular conduction system were cut according to the serial sectioning method of Lev et al. The compact atrioventricular nodes were removed from thick paraffin sections (150 μm) under a stereomicroscope. The cells were then isolated by enzyme digestion and ultrasonic treatment. Smears of the isolated cells were double stained with azocarmin-G and acriflavine-Feulgen. Cytofluorometric DNA determinations of the DNA content of atrioventricular nodal cells were performed. Atrioventricular nodes were found to be composed of a large number of diploid cells and a small number of tetraploid cells. No octaploid cells were found. These findings reveal that the process of polyploidization in atrioventricular nodal cells is different from that found in working heart-muscle cells.

    Notizen: Summary In an attempt to analyse the kinetics of angiogenesis in the brain, we developed a new lectin-histochemical staining technique for identifying the vasculature. Three horseradish-peroxidase-conjugated lectins, i.e., Griffonia simplicifolia agglutinin 1 (GS1), Ricinus communis agglutinin 1 (RCA1) and soybean agglutinin (SBA), selectively stained vascular walls in brain-tissue sections. When these lectins were injected into the circulation of ether-anesthetized animals via the pulsating left ventricle, they bound specifically to the inner surface of endothelial cells and revealed the three-dimensional architecture of the vascular network within thick tissue preparations. When this technique, referred to a lectin angiography, was combined with 5-bromo-2-deoxyuridine (BudR) immunohistochemistry, proliferating capillary cells could be easily identified in three-dimensional structures of the developing vasculature. Because of its simplicity and wide applicability, lectin angiography should be useful for analysing the kinetics of angiogenesis in developmental, regenerative, and pathological conditions in various tissues and organs.

    Notizen: Summary We describe a method for measuring the DNA content of the component cells of the organ of Corti using serial sections of human cochleae obtained at autopsy. Cochleae were fixed in Carnoy's solution and embedded in Acrytron E, a water-miscible methacrylate resin. A procedure was developed to reduce the background fluorescence in methacrylate-embedded sections; the resin was pretreated with ion-exchange resin (Amberlite IRA-410). Experiments showed that pretreatment reduce the background fluorescence practically to zero. Seventy 3 μm-thick serial sections were prepared on fluorescence free glass slides and stained with azocarmin G and acriflavine-Feulgen. After postirradiation using blue excitation light, the amount of Feulgen-DNA present in the target nucleus in each section was determined using a microfluorometer. The amount of DNA in the entire nucleus was determined by adding together the DNA contnet of the segments of the nucleus. The characteristic appearance of the organ of Corti made it easy to detect these cells; under green excitation light the cells of this organ exhibited red cytoplasmic azocarmin-G fluorescence. Due to the relatively wide internuclear spaces, cytofluorometry of individual nuclei could be performed without interference from the neighboring cells. Our technique using serial sections allowed us to measure the DNA contnet of individual cells and obtain histological information about particular cells and their neighboring cells. Several polyploid cells were found among the Hensen's cells in the cochlea, while all other component cells of the organ of Corti were diploid.

    Notizen: Abstract Multiple optimum solutions of a multistage allocation problem, well-known to chemical engineers, are analyzed. The number of local optima becomes greater with a decrease in the initial-condition value of the first stage or with an increase in the total stage number. The fact that this behavior is closely related to the flat portion of the profile of a curvef(x), which determines the objective function, is revealed. A construction method by Aris is used to give an excellent insight into this behavior. Moreover, the construction curves ensure that all stationary points are found. Finally, a theorem to discriminate local optima from stationary points, without evaluating second-order derivatives, is presented.

    Notizen: The densities and expansion coefficients of several Nafions in the acid and salt form were measured. For some samples, effects of variations in the equivalent weight, moisture content, and degree of uniaxial orientation were explored. The densities, which show considerable experimental scatter, seem to be independent of the equivalent weight but to be a strong function of the moisture content. Thin films of the as-received, unbacked polymer are strongly uniaxially oriented. After annealing, reproducible values of the expansion coefficient can be obtained. Two distinct breaks in the linear expansion curve are seen; in the salts, the upper one correlates with the glass transition of the material, while the lower one is found in the vicinity of the mechanical β-dispersion of the polymer. The large scatter of the experimental densities is consistent with the hypothesis that the sample is subject either to a microphase separation process which is strongly influenced by the sample history and which affects the density of the material very strongly, or to partial crystallization of the polymer, or both.

    Notizen: Sorption phenomena of water and aqueous salt solutions by a perfluorinated polymer containing sulfonic acid groups (Nafion) were investigated. The temperature and concentration dependencies of the sorption by the membranes in the acid and salt forms were studied. The apparent activation energies for the diffusion of water in the H-form membrane and in the K-salt form were obtained as 4.9 and ca. 13.0 kcal/mole, respectively. The sorption kinetics during the neutralization of the membranes were observed in several aqueous solutions. A maximum in the sorption curve during the neutralization process was found and explained as resulting from the differences in the diffusion coefficients of water and of the cations and from the different number of water molecules absorbed by a SO3-H+ (acid) site and a neutralized site. The diffusion coefficients D of several cations (K+, Cs+, Ba2+, and Ca2+) were determined and found to be considerably smaller than that of water. For the various cations, log D was related linearly to q/a, where q is the cation charge and a is the separation between centers of charge of the cation and anion. The dependence of water sorption upon the degree of neutralization of the membrane was also studied at room temperature. It was observed that for membranes of a low degree of neutralization a secondary sorption process existed, while no such secondary sorption could be found for the pure acid or the highly neutralized membranes. This secondary sorption was attributed to a structural rearrangement in the polymer. The apparent diffusion coefficient of water and the number of water molecules absorbed at equilibrium by an ionic site, ns, were obtained as a function of the degree of neutralization. The diffusion coefficient of water was dependent strongly on both the degree of neutralization and type of the salt, but no quantitative relation could be established. For all the salts studied in this paper, ns was linearly related to the degree of neutralization, x, supporting the assumption that the value of ns could be divided into those water molecules absorbed by an SO3-H+ (acid) site, nh, and those absorbed by a neutralized site, nm. It was found that the value of q × nm had a strong correlation with a characteristic constant of the cations since a plot of q × ns versus log (q/r) yielded a straight line (r being the radius of the cation).
